血管新生は、小さな局所的な新生物からより大きな、潜在的に転移性腫瘍1、2への癌の進行に不可欠なプロセスである。腫瘍の増殖と血管新生との相関関係は、腫瘍学研究の分野で重点の一つとなる。しかし、形態学的変化を測定する従来の方法は、視覚化されたアプローチを使用して、生きた動物の腫瘍の進行と血管新生を同時に監視することができません。
腫瘍細胞の生物発光イメージング(BLI)は、その非侵襲性、感受性、および特異性3、4、5、6のために腫瘍の増殖を監視するための特に適切な実験方法である.BLI技術は、ルシフェラーゼが生物発光を放出しながら特定の基質の酸化を触媒することができるという原理に基づいています。移植された腫瘍細胞で発現されるルシフェラーゼは、生体イメージングシステムによって検出することができる注入された基板と反応し、シグナルは生体内6、7における細胞数または細胞局在化の変化を間接的に反映する。
腫瘍増殖を除いて、腫瘍血管新生(癌進行の重要なステップ)はまた、Vegfr2-Fluc-KIトランスジェニックマウス8、9、10を用いてBLI技術を用いて可視化することができる。血管内皮増殖因子(Vegf)受容体2(Vegfr2)は、Vegf受容体の1つのタイプであり、主に成体マウス11の血管内皮細胞において発現される。Vegfr2-Fluc-KIトランスジェニックマウスでは、ホタルルシフェラーゼ(Fluc)のDNA配列が内因性Vegfr2配列の最初のエキソンにノックされる。その結果、Flucは、マウスの血管新生のレベルと同じ方法で(BLIシグナルとして現れる)発現する。数ミリメートルを超えて成長するために、腫瘍は、腫瘍細胞1からの成長因子によって引き起こされるVegfr2を高度に発現する既存の血管から新しい血管を新たに産生する。これにより、BlIによる腫瘍血管新生を非侵襲的に監視するためにVegfr2-Fluc-KIトランスジェニックマウスを使用する可能性が開かれる。
このプロトコルでは、ホタルルシフェラーゼ(Fluc)およびレニラ・ルシフェラーゼ(Rluc)イメージングを介して単一のマウスにおける腫瘍進行および血管新生をそれぞれモニタリングする腫瘍性マウスモデルが確立される(図1)。4T1細胞株(4T1-RR)を作成し、Rlucイメージングにより細胞増殖を追跡するためにRlucおよび赤色蛍光タンパク質(RFP)を安定的に発現させる。腫瘍の進行および回帰における血管新生の動的変化をさらに調べるため、自殺遺伝子ヘルペス単純ヘルペスを発現する別の4T1細胞株(4T1-RRT)が作成され、チミジンキナーゼ(HSV-ttk)、Rluc、およびRFPが切り捨てられた。ガンシクロビル(GCV)の投与により、HSV-ttk発現細胞は選択的にアブレーションされる。これらの細胞株に基づいて、Vegfr2-Fluc-KIマウスにおける腫瘍担持モデルは、生体内で腫瘍進行および腫瘍血管新生を橋渡しする実験モデルとして機能する。

血管新生は腫瘍転移および進行において重要な役割を果たす。このプロトコルでは、乳癌のマウスモデルにおける動的腫瘍進行および血管新生をモニタリングするために、二重生物発光イメージング法を用います。このモデルでは、ホタルルシファーゼとレニラルシメラーゼイメージングを介して、単一マウスの腫瘍増殖と血管新生を同時に追跡することができます。このモデルは、抗腫瘍薬スクリーニングおよび腫瘍学研究に広く適用され得る。この二重生物発光モデルは、腫瘍の進行と退行を追跡し、治療戦略に応答して腫瘍関連の分子プロセスを検出するために使用することができます。血管形成は腫瘍進行の重要なプロセスである。したがって、効果的な腫瘍治療を開発するためには、視覚および敏感な方法で腫瘍の進行および血管新生をモニタリングすることが必要である。この手順は、私たちの研究室の学生である張海、陳王、そして上陳が示します。レンチウイルス包装および生産種子用 10〜 293T 細胞の 2 ミリリットルの DMEM で 10% 牛血清を補った摂氏 37 度と 5%の炭酸ガスで加湿インキュベーターで一晩培養のための 6 ウェル プレートに.翌朝、7.5マイクロリットルのリポソームと250マイクロリットルのミニマルエッセンシャルミディアムを2つの別々の1.5ミリリットルチューブに混ぜ、室温で5分間インキュベーションします。DNA溶液を調製するには、表に概説されているように、個々の1.5ミリリットルチューブの最小必須培地のプラスミドレンチウイルスRRベクターおよびヘルパープラスミドを250マイクロリットルに添加する。インキュベーションの最後に、DNA RR溶液を滴下方式でリポソーム懸濁液に加え、DNAを室温で20分間脂質膜に結合させます。混合物がインキュベートされている間、293T細胞培養物の上清を1ミリリットルの新鮮な培養培地に置き換え、適切なリポソームDNA混合物を各ウェルに0.5ミリリットル加えて穏やかに加える。各ウェルの上清を抗生物質を添加した新鮮な培養培地の1ミリリットルで置き換える前に、細胞を12〜16時間培養インキュベーターに戻す。さらに36時間のインキュベーションの後、上清をレンチウイルス株当たり1つの円錐チューブにプールし、遠心分離によって293T細胞を沈降させる。その後、レンチウイルスRR含有上清を個々の1.5ミリリットルの滅菌ポリプロピレン貯蔵チューブに移し、マイナス80°Cで貯蔵します。4T1乳腺腫瘍細胞における遺伝子発現用レンチウイルス伝達については、6ウェル4T1細胞培養板の各ウェルの上清を、牛胎児血清と1ミリリットルのレンチウイルスストックを添加した新鮮なRPMIベースの細胞培養培地の1ミリリットルに置き換える。その後、各井戸にポリブレンのミリリットルあたり8マイクログラムを追加し、穏やかにミペットを数回混合します。プレートを遠心分離して、トランスダクション効率を高め、細胞を細胞培養インキュベーターに4〜12時間戻す。インキュベーションの終わりに、レンチウイルス粒子およびポリブレンを除去するために血清および抗生物質を添加した新鮮な培養培地で各ウェルの上清を置き換える。レンチウイルス導入細胞に対して選択するには、トランスデュースされた4T1細胞培養物を選択培地との1対3~1対4の比率で通過し、2~3日ごとに培地を変化させる。7日間の選択培地培養後、蛍光反転相造影顕微鏡下でトランスデューテッド細胞培養を見て、赤色蛍光タンパク質陽性またはRFP-4T1細胞の数と4T1細胞の全ての数を3つの視野領域で数え、RFP陽性比を推定する。腫瘍を持つマウスモデルを設定するには、PBSで80%コンフルエントスエントリンジ細胞培養液を洗浄してから、60ミリメートル培養プレートあたり2ミリリットルのトリプシン-EDTAで細胞を取り外します。細胞がプレート底から持ち上がったとき、1プレートあたり5〜10ミリリットルの血清補充培地で反応を停止します。そして、カウントするための個々の15ミリリットル遠心分離管に培養を移す。血清濃度を含まない培地100マイクロリットル当たり6細胞を1倍10~6細胞に希釈する。そして、4T1-RR細胞株の100マイクロリットルを麻酔腫瘍軸受モデルマウスの左肩に皮下に注入し、4T1-RRT細胞株の100マイクロリットルを同じマウスの右肩に注入する。注射後、完全に回復するまで、各マウスを熱支持を持つ適切な回復領域に置きます。マウスが腫瘍を持つものであることを確認するために、毎日7日間腫瘍腫瘤を触診する。7日目の移植後、腹腔内は実験終了まで1日2回、ガンシクロビルの1キログラム当たり50マイクログラムを各腫瘍を持つマウスに注入する。腫瘍の成長のために、生きた画像システムを開く。リビングイメージングソフトウェアを初期化し、システムを初期化します。システムが初期化されている間に、インスリン注射器を使用して、適切な量のコエンテラジンをレトロブルバーに画像化する各マウスを注入する。注射したマウスをカメラチャンバーに入れ、すぐにマウスの写真を数枚得て、生物発光シグナルが消えるまで4T1細胞からレニラルシファーゼ信号を取得します。レニラルシメラーゼイメージングの10分後、各動物に適切な量のD-ルシフェリンを腹腔内注入するためにインスリン注射器を使用する。D-ルシフェリン注射の10分後、マウスをカメラチャンバーに入れ、マウスの後回しの画像を複数得て、血管新生からホタルルシフラーゼ信号を得る。レンチウイルス導入後、99%以上の細胞がRFP陽性であり、2つの導入細胞株間で観察された細胞培養形態の違いはない。続いて、トランスデューセイングされた細胞の生物発光イメージングは、両方の細胞株が同様の強度の強い生物発光シグナルを放出することを明らかにする。トランスジェニック腫瘍を持つマウスモデルにトランスデューセ細胞株を皮下注射した後、血管新生誘発腫瘍増殖は、同じ動物中のD-ルシフェリンの存在下でホタルルシフェラーゼシグナルによって評価することができる。7日間の移植後、ガンシクロビル投与は4T1-RRT細胞で死を誘発し、これらの細胞によって確立された腫瘍におけるレニラルシファーゼシグナルの劇的な減少をもたらす。ホタルルシメラーゼシグナルは、レニラルシメラーゼ発現の増加と共に増加し、ルシファーゼの減少後に減少します。これらの結果をまとめて、腫瘍血管新生と腫瘍増殖との間に直接的な相関関係があることを示唆している。実際、ガンシクロビル誘発腫瘍細胞死は、腫瘍血管新生の阻害につながる可能性がある。また、抗血管内皮増殖因子受容体IIに対する免疫染色は、血管新生のマーカーとして、4T1-RRT腫瘍からの切片よりも4T1-RR腫瘍組織切片においてより顕著に確立された微小血管構造を明らかにし、ガンシクロビル投与後に観察されたホタルルシファーゼシグナルの低下と一致する。このプロトコルの最も重要なステップは、細胞と血管新生の同時追跡にFLucとRLucのような2種類のルシファーゼを使用することです。この技術は、異なる場所のがん細胞を治療するために使用することができ、それはウイルス癌モデルで広く使用することができます。このマウスモデルは、生きている動物の中で動的に可視化されるHSV-ttk/GCV追跡システムの腫瘍増殖および抗腫瘍効果を可能にする。培地にはヒトに有害なレンチウイルス粒子が含まれているため、レンチウイルストランスフェクションを防ぐためにバイオセーフティレベルII施設が必要です。
