Met het dreigende visuele stimuli systeem, een meerderheid (97%) van gezonde Eye-muizen toonde vlucht reactie. Één van 29 muizen toonde geen duidelijke vlucht reactie. Het visuele systeem begint bij het netvlies, waar visuele signalen worden gevangen door fotoreceptoren, gekanaliseerd naar bipolaire cellen (2ND-orde neuronen), en uiteindelijk doorgegeven aan ganglioncellen (3RD-orde neuronen). Retinale 2ND-en 3RD-orde neuronen worden verondersteld om veelvoudige neurale wegen te vormen die bepaalde aspecten van het visuele signaleren zoals kleur, motie, of vorm overbrengen. Deze diverse visuele kenmerken worden doorgegeven aan de laterale geniculate kern en de visuele cortex. Cellulaire en moleculaire technologieën hebben bijgedragen aan ons begrip van visuele verwerking en mogelijke ziektemechanismen die kunnen voortvloeien uit afwijkende visuele verwerking. Een dergelijk begrip kan bijdragen tot de ontwikkeling van kunstmatige ogen. Hoewel cellulaire onderzoeken en analyses bieden diepgaande kennis op een cellulair niveau, een combinatie van gedrags-experimenten en cellulaire experimenten zou aanzienlijk vergroten onze huidige begrip van minieme visuele processen. Bijvoorbeeld, de Starburst amacrine cellen zijn de belangrijkste neuronen voor bewegingsdetectie in de muis retina. Meerdere methoden zijn beschikbaar voor het meten van de muis visie. Deze methoden hebben voordelen en beperkingen. Leerling licht reflex maatregelen leerling vernauwing in reactie op lichtprikkels, die niet nodig anesthesie en vertoont objectieve en robuuste reacties 19. Hoewel de pupil reflex de retinale licht respons in vivo simuleert, wordt de reflex voornamelijk bemiddeld door de intrinsiek fotogevoelige KNOOPCELLEN van het netvlies (ipRGCs) 27. Omdat ipRGCs vertegenwoordigen een kleine minderheid van RGCs en niet dienen als conventionele beeld-vormende ganglioncellen, deze meting biedt geen informatie met betrekking tot de meerderheid van de ganglioncellen.
Het dreigende licht experiment is niet eerder beschouwd als een belangrijke test voor het meten van de muis visie. Nochtans, is het ook een robuuste en betrouwbare visie test over diverse soorten, zoals muis28,29, zebravis30, sprinkhanen31,32, en menselijk33,34, 35. belangrijk is dat het dreigende experiment is een van slechts een paar methoden om het imago te testen-vormende pathway-het is niet een reflex pad-gezien de visuele en de limbische systemen in het centrale zenuwstelsel zijn betrokken bij dit circuit36, 37,38. We hebben een dreigende visuele stimulus systeem en hebben aangetoond dat het vermogen om bewegingsdetectie te lokken in de muis, die we gebruiken als een proxy om de intactheid van de muis visueel systeem te beoordelen.

Het visuele systeem in het centrale zenuwstelsel verwerkt diverse visuele signalen. Hoewel de totale structuur is gekenmerkt van het netvlies door de laterale geniculate kern aan de visuele cortex, het systeem is complex. Cellulaire en moleculaire studies zijn uitgevoerd om de mechanismen ter ondersteuning van visuele verwerking en, bij uitbreiding, ziektemechanismen te verhelderen. Deze studies kunnen bijdragen tot de ontwikkeling van kunstmatige visuele systemen. Om de resultaten van deze studies te valideren, is gedrags visie testen noodzakelijk. Hier laten we zien dat de dreigende stimulatie experiment is een betrouwbare muis visie test die een relatief eenvoudige setup vereist. Het dreigende experiment werd uitgevoerd in een grote behuizing met een schuilplaats in een hoek en een computer monitor gelegen aan het plafond. Een CCD-camera gepositioneerd naast de computer monitor diende om het gedrag van de muis te observeren. Een muis werd geplaatst in de behuizing voor 10 minuten en mag acclimatiseren naar en verken de omgeving. Dan, de monitor geprojecteerd een programma-afgeleide dreigende stimulus 10 keer. De muis reageerde op de prikkels, hetzij door bevriezing of door de vlucht naar de schuilplaats. Het gedrag van de muis voor en na de dreigende stimuli werd opgenomen, en de video werd geanalyseerd met behulp van Motion Tracking software. De snelheid van de muisbeweging aanzienlijk veranderd na de dreigende stimuli. In tegenstelling, geen reactie werd waargenomen bij blinde muizen. Onze resultaten tonen aan dat de eenvoudige dreigende experiment is een betrouwbare test van de muis visie.

Een muis met gezonde ogen werd geplaatst in de behuizing en toegestaan om acclimatiseren voor 10 min. Using Looming Visual Stimuli to Evaluate Mouse Vision

Terwijl veel muis visie tests beschikbaar zijn, de dreigende experiment is een van de weinige methoden om het beeld vormen route te testen, dat is niet een visuele reflex traject.Terwijl veel andere visuele gedragtest uitgebreide training in dierchirurgie vereisen, vereist de dreigende techniek weinig experimentele training. Begin met de bouw van een rechthoekige open deksel behuizing met behulp van aluminium framing en PVC-panelen om de muis huis tijdens dreigende visuele stimuli presentatie. Gebruik vervolgens analysesoftware om de beweging van het dier rond de arena te volgen voorafgaand aan, tijdens en na de stimuluspresentatie. Bereken de snelheid en de afstand van de muis van het toevluchtsoord.Als het beeld van de arena is vervormd als gevolg van de videohoek, corrigeert u de x- en y-coördinatie voorafgaand aan de berekening. Vergelijk ten slotte de parameters voor en na het opdoemende stimulus begin om te bepalen hoe de muis reageerde op de stimuli. Of het nu door bevriezing, vluchten, of het aantonen van geen verandering in gedrag.Vastgelegde videoclips werden geanalyseerd met behulp van een commerciële analysesoftware met een handmatige trackingfunctie. Met behulp van de trackingfunctie werd de positie van de muis geïdentificeerd in elk frame van de video voor, tijdens en na de dreigende stimuli. Toen de vlucht plaatsvond, nam de snelheid abrupt toe en de afstand tot de schuilplaats verminderde dienovereenkomstig.Daarentegen was de snelheid bijna nul millimeter per seconde toen muizen bevroor. De snelheid werd verminderd in vergelijking met de controle voor het opdoemen en de afstand tot de koepel veranderde niet tijdens deze periode. Het is belangrijk om te onthouden om een basislijn van beweging te krijgen voorafgaand aan het begin van de stimulus of een kwantitatieve reactie op de stimulus kan niet worden bepaald.Het is belangrijk om meerdere tests uit te voeren voor een nauwkeurige visie-evaluatie. Pupil lichtreflex is een veelgebruikte methode, die ervoor zorgt dat de muis gezonde ogen heeft. Het mechanisme van dreigende reactie is niet volledig begrepen.Deze techniek kan mogelijkheden bieden om mechanismen van bewegende objectherkenning en angstreacties voor neurowetenschappers te verkennen.
