Los ratones humanizados (hu-mice) han transformado el estudio de muchos aspectos de la salud humana y la enfermedad, incluyendo hematopoyesis, inmunidad, cáncer, enfermedad autoinmune y enfermedad infecciosa1,2,3,4 ,5,6,7,8,9. Estos hu-mice tienen la clara ventaja sobre otros modelos de ratón en que tienen un sistema inmunológico humano funcional y pueden ser infectados con patógenos específicos humanos. Sin embargo, la importancia del microbioma intestinal ha quedado demostrada por su papel en muchas enfermedades humanas como la obesidad, el síndrome metabólico, las enfermedades inflamatorias y el cáncer10,11,12, 13. El sistema inmunitario mucoso y el microbioma intestinal están regulados recíprocamente para mantener la homeostasis intestinal y sistémica. El sistema inmunitario está formado por antígenos presentados por el microbioma intestinal y recíprocamente el sistema inmunitario desempeña un importante papel regulador en la promoción de bacterias intestinales comensales y la eliminación de patógenos14,15, 16. Sin embargo, el microbioma intestinal de hu-mice no se ha caracterizado bien y el microbioma intestinal murino difiere sustancialmente en composición y función de los seres humanos17. Esto se debe a diferencias evolutivas, fisiológicas y anatómicas entre el intestino murino y humano, así como otros factores importantes como la dieta, que pueden influir en los resultados experimentales de los modelos18de la enfermedad de hu-mice. Por lo tanto, más allá de la clasificación del microbioma intestinal murino de hu-mice, se necesita un modelo animal con un sistema inmune humano y microbioma intestinal humano para estudiar las complejas interacciones de la enfermedad humana in vivo.
El estudio de las enfermedades humanas directamente en seres humanos es a menudo poco práctico o poco ético. Muchos modelos animales no se pueden utilizar para estudiar patógenos humanos como el virus de la inmunodeficiencia humana tipo 1 (VIH-1). Los modelos de primates no humanos son genéticamente endogámicos, muy caros y no son susceptibles a muchos patógenos humanos. Cg-PrkdcscidIl2rgtm1Wjl/SzJ (NSG), no están disponibles comercialmente como GF. Los animales GF también pueden sufrir de deficiencias inmunitarias duraderas que no se invierten completamente por el injerto de microbios21. Por lo tanto, creamos un doble hu-mice con un sistema inmune humano funcional y microbioma intestinal estable similar al humano bajo condiciones específicas libres de patógenos (SPF). Para generar doble hu-mice, se realizó una cirugía en ratones NSG para crear ratones de médula ósea, hígado, timo humanizados (hu-BLT). Los ratones hu-BLT fueron tratados con antibióticos de amplio espectro y luego se les administraron trasplantes fecales con una muestra de donante humano saludable. Caracterizamos el microbioma intestinal bacteriano de 173 muestras fecales de 45 ratones dobles hu-BLT y 4 muestras de donantes fecales humanos. Los ratones de doble hu-BLT tienen perfiles únicos del gen rRNA 16S basados en la muestra de donante humano individual que se trasplanta. Es importante destacar que el microbioma trasplantado similar al humano se mantuvo estable en ratones durante la duración del estudio hasta 14,5 semanas después del trasplante. Además, los metagenomas pronosticados mostraron que los ratones de doble hu-BLT tienen diferente capacidad funcional pronosticada que los ratones de hu que es más similar a las muestras de donantes humanos.

Los ratones humanizados (hu-mice) que cuentan con un sistema inmunitario humano funcional han cambiado fundamentalmente el estudio de patógenos humanos y enfermedades. Se pueden utilizar para modelar enfermedades que de otro modo son difíciles o imposibles de estudiar en seres humanos u otros modelos animales. El microbioma intestinal puede tener un profundo impacto en la salud y la enfermedad humanas. Sin embargo, el microbioma intestinal murino es muy diferente al que se encuentra en los seres humanos.  Hay una necesidad de modelos preclínicos de hu-mice mejorados que tengan un microbioma intestinal humano injertado. Por lo tanto, creamos doble hu-mice que cuentan con un sistema inmunológico humano y microbioma intestinal humano estable. Cabeceo. Nuestro protocolo es significativo, porque se puede utilizar para crear ratones de forma reproducible con un microbioma intestinal humano y un sistema inmunitario humano. Llamamos a estos ratones, ratones humanizados dobles. El modelo de ratón doble humanizado se puede utilizar para la investigación biomédica básica y traslacional, incluyendo el estudio de las relaciones in vivo entre el microbioma intestinal humano y el sistema inmunológico humano.Demostrando el procedimiento estarán Jianshui Zhang, un postdoctorado, y Yilun Chang, un estudiante de doctorado de nuestro laboratorio. Los cambios fenotípicos en el bazo y el cecum inducidos por el tratamiento antibiótico son similares a los observados en animales libres de gérmenes. Esta gráfica de análisis de componentes principales de los datos representativos de secuenciación de ARN ribosomal 16S revela que los ratones humanizados dobles tienen microbiomas intestinales similares a los humanos que son exclusivos de la muestra de donante humano.El aspecto más crítico de este protocolo es limitar la introducción de bacterias no deseadas para asegurar que los ratones permanezcan sanos y mantengan el microbioma intestinal humano.
