该方法对选择的抗原恢复原生人类抗体的效用有几个显著的局限性。首先，只有可标记的可溶性蛋白质才能用作流动细胞仪的"诱饵"。对于代表性结果中呈现的种类，我们使用了一系列来自tau蛋白的合成重叠肽，因为使用整个蛋白质证明是困难的。线性肽的使用可能不是最佳的，因为它可能会限制对非线性或结构表位抗体的识别。然而，我们能够识别几种针对tau的独特抗体，这些抗体目前正在进一步评估8，9。另一个限制是IgGs的分子生物学恢复和克隆的低通量。最初的分选策略允许筛选数百万个细胞，但随后的分子生物学处理由多个阶段组成。正在进行的优化工作，以纳入新技术，如吉布森组装10已经简化了几个步骤，但瓶颈仍然是克隆单个重链和轻链对。
"BSelex"方法利用在记忆B细胞表面表达的BCR来识别显示抗原反应的细胞，然后通过流式细胞测定11、12恢复这些单个细胞。由于这种对BCR的依赖，该方法仅限于记忆B细胞室，并且不捕获抗体分泌细胞（ASC），如血浆细胞。然而，与ASCs相比，这种方法可能有利于恢复更广泛的抗原特异性免疫球蛋白。流感疫苗接种研究表明，虽然反应性ASC可以由少量扩热B细胞克隆主导，但抗原特异性记忆B细胞群很少克隆12。T细胞表面的T细胞受体（TCR）在基因排列和重组中与B细胞受体相似，以最大化多样性。单细胞方法类似于这里介绍的方法，已经开发用于评估T细胞的表谱，包括α和β链的恢复和单细胞克隆13。然而，TCR识别需要由MHC分子呈现肽，为标记诱饵方法增加显著的复杂性，以识别肽特异性T细胞。与B细胞不同，T细胞不经过整个可变区域的亲和成熟，因此识别短CDR3区域和一些侧翼序列是识别和重组所需的全部。最后，该方法描述了使用流式细胞测定法识别IgG分子的方法，但也可以使用替代型型标记来识别不同等型的B细胞。

single-cell screening method for the selection and recovery of antibodies with desired specificities from enriched human memory b cell populations

该方法涵盖从人类捐赠者中分离抗原特异性抗体的多步骤过程。在此显示的代表性数据中，细胞用荧光标记肽池孵育，这些肽代表tau蛋白的几个不同领域，包括磷酸化肽，以模拟假定磷酸化位点。这些肽被用作"诱饵"，以识别与感兴趣的tau表皮反应的细胞。为了准备分拣，使用荧光标记的荧光标记组来识别富集B细胞群中的不同细胞群。设计了一系列细胞测量门来分离目标记忆B细胞（图1）。淋巴细胞被分离基于其细胞大小和粒度使用正向散射（FSC）和侧散射（SSC）图在流细胞学6，7。排除多个细胞（"双细胞"）和死细胞后，型型标记允许通过IgG、CD19（B细胞）和CD27（内存）分离IgG+记忆B细胞。在这种方法中，CD27标记不分布到两个离散群体，因此CD27的前45%和表达单元包含在最终门。最后，APC和PE荧光团的细胞双阳性分布到门控图的右上象限中，指示对两个标记版本的肽的反应。落在绘制门内的细胞被分离，并分类到96孔板的单个井。使用两个不同的标签使用抗原可提高信噪比，并减少随后分子生物学过程中的误报数。排序的细胞表示最终门中大约 0.1% 的内存 B 细胞，以及起始细胞样本的 0.001%。
单细胞克隆的第一次读出是确认各自重变链和轻变量链的扩增（图2）。由于需要两种放大器的成对恢复，PCR反应在agarose凝胶上并排评估，并从凝胶和提取的DNA片段中切除匹配的对。典型的放大效率是30-50%重链和50-70%轻（卡帕）链。配对的放大器的回收效率通常在 25-40% 之间。这些效率因供体池而异，代表性数据（图3）是24个单细胞（42%成对恢复）高效扩增的一个例子。在IgG克隆之后，IgG表达载体被转染成人类胚胎肾（HEK-293）悬浮细胞，用于最大化表达。使用无血清介质可减少重组抗体制备中的污染蛋白，并有助于将后续结合检测中的噪音降至最低。回收的抗体对原陶肽板进行筛选，并由ELISA进行反应性评分（图4）。OD = 高于背景0.5的初始阈值用于指示阳性抗原反应性，β-actin蛋白用作非特异性结合的控制。如果流式细胞学和分选步骤使用混合肽池，则筛选 ELISA 是去解回收 IgGs 特定反应的第一步。10个IgG测定中，有3种对磷酸化CBTAU22.1肽表现出反应性，1种对非磷酸化CBTAU27.1有反应（图4A）。使用相同重组抗体样本的浓度曲线对初始屏幕中识别的肽进行进一步确认，并添加一个肽作为阴性对照（图4B）。对于每个阳性命中，从转换后的池中分离出单个质粒克隆，并通过相同的 ELISA 方法重新确认。只显示了克隆34的数据，并且确认对非磷酸化CBTAU27.1的反应，同时用磷酸化27.1肽观察到较低的亲和结合（图4C）。
