Deze methode heeft verschillende kritieke stappen en vereist mogelijke wijzigingen om te zorgen voor een geslaagde implementatie. Ten eerste moet de dosering en timing van de intraveneuze injectie van antilichaam/antilichaam conjugaat worden afgestemd op de specifieke toepassing. In het algemeen moeten doseringen worden gebruikt die consistent zijn met hoe het antilichaam conjugaat meestal zal worden gebruikt, d.w.z. overeenkomende doseringen van het therapeutische antilichaam of contrastmiddel op basis van antilichamen....

Monoklonale antilichamen (mAb) zijn grote glycoproteïnen (ongeveer 150 kDa) van de immunoglobuline superfamilie die worden uitgescheiden door B-cellen en die een primaire functie in het immuunsysteem hebben om de biologische functie van, of markeren voor vernietiging, bacteriële of virale pathogenen, en kan abnormale eiwit expressie op kankercellen herkennen1. Antilichamen kunnen een extreem hoge affiniteit hebben met hun specifieke epitopen tot en met femtomollaire concentraties, waardoor ze veelbelovend zijn in biogeneeskunde2. Met de ontwikkeling van hybridoma-technologie door Milstein en Köhler (bekroond met de Nobel prijs in 1984), werd de productie van mAbs mogelijk3. Later werden menselijke MABS gegenereerd met behulp van de faag display technologie of transgene muis stammen en revolutioneerden hun gebruik als nieuwe research tools en Therapeutics4,5.
Kanker is een wereldwijd gezondheidsprobleem en een belangrijke doodsoorzaak die de behoefte aan nieuwe benaderingen voor preventie, opsporing en therapie6creëert. Tot nu toe, MABS hebben toegestaan verdrievoudiging van de rol van genen en hun eiwitten in tumorvorming en wanneer gericht tegen kanker biomarkers, kan tumor detectie en karakterisering voor patiënt stratificatie. Voor kankertherapie, bispecifieke mAbs, antilichaam-drug conjugaten, en kleinere antilichaam fragmenten worden ontwikkeld als Therapeutics, en voor de beoogde drug delivery ter verbetering van de therapeutische werkzaamheid7. Daarnaast dienen antilichamen voor de biomarker-targeting van contrastmiddelen voor moleculaire beeldvormings modaliteiten zoals fluorescentie geleide chirurgie, photoacoustic (PA) beeldvorming, echografie (VS) moleculaire beeldvorming en klinisch gebruikte positron-emissie tomografie (PET) of enkelvoudige foton emissie computertomografie (SPECT)8. Tot slot kunnen antilichamen ook worden gebruikt als theranostische middelen die stratificatie van patiënten mogelijk maken en respons monitoring voor gerichte therapieën9. Daarom beginnen nieuwe mAbs een cruciale rol te spelen bij de opsporing, diagnose en behandeling van kanker.
Ondanks kritische vooruitgang in de ontwikkeling en productie van nieuwe en zeer specifieke mAbs, diagnostische en therapeutische toepassingen kunnen worden weergegeven ineffectief als gevolg van de complexiteit van de tumor omgeving. Interacties tussen antilichamen zijn afhankelijk van het type epitope,d.w.z. ofhet lineair of conformationeel10is. Naast de herkenning van antigenen moeten antilichamen natuurlijke barrières overwinnen, zoals de wanden van het vat, basale membranen en de tumor stroma om doelcellen te bereiken die het antigeen uitdrukken. Antilichamen communiceren niet alleen met het weefsel via het variabele fragment antigen binding (FAB) domein, maar ook via het constant kristallijne fragment (FC) dat verder leidt tot off-site interacties11. Targeting wordt ook bemoeilijkt door de heterogene expressie van tumormarkers in de tumor bulk en heterogeniteit in tumor vascularisatie en het lymfestelsel12,13. Bovendien, de tumor micro omgeving is samengesteld uit kanker-geassocieerde fibroblasten die tumorcellen ondersteunen, tumor immune cellen die anti-tumor immuunreacties onderdrukken, en de tumor endotheel die het transport van zuurstof en voedingsstoffen ondersteunt, alle van die interfereren met de penetratie, distributie en beschikbaarheid van op antilichamen gebaseerde therapieën of diagnostiek. Algemene, deze overwegingen kunnen beperken therapeutische of diagnostische werkzaamheid, vermindering van de behandeling reactie, en kan resulteren in tumor resistentie.
Daarom is het voor de ontwikkeling van efficiënte op antilichamen gebaseerde therapieën en diagnostiek cruciaal om de biodistributie en interactie van het op antilichamen gebaseerde geconjugeerde in de tumor micro omgeving te beoordelen. Momenteel, in preklinische studies, marker expressie in tumor Research modellen wordt geanalyseerd ex vivo door immunofluorescentie (indien) kleuring van tumor secties14. Standaard indien kleuring wordt uitgevoerd met primaire marker-specifieke antilichamen die vervolgens worden gemarkeerd door secundaire fluorescently gelabelde antilichamen op ex vivo tumorweefsel segmenten die zijn geïsoleerd van het dier. Deze techniek benadrukt de statische locatie van de marker op het moment van weefsel fixatie en geeft geen inzicht in hoe de op antilichamen gebaseerde therapieën of diagnostiek in fysiologische omstandigheden kunnen verspreiden of ermee omgaan. Moleculaire beeldvorming door Pet, SPECT, US en PA kan informatie verschaffen over de verdeling van antilichamen-geconjugeerde contrast agenten in levende preklinische modellen8,15. Aangezien deze beeldvormings modaliteiten niet-invasief zijn, kunnen longitudinale onderzoeken worden uitgevoerd en kunnen tijdgevoelige gegevens worden verzameld met een minimaal aantal dieren per groep. Deze niet-invasieve moleculaire beeldvormings benaderingen zijn echter niet gevoelig genoeg en hebben onvoldoende resolutie voor de lokalisatie van antilichaam distributie op cellulair niveau. Bovendien kunnen de fysische en biologische kenmerken van het primaire antilichaam drastisch worden gewijzigd door de conjugatie van een contrastmiddel16.
Om de in vivo fysiologische en pathologische omstandigheden rekening te houden met de interactie van op antilichamen gebaseerde therapieën en diagnostiek binnen de tumor omgeving en om cellulaire en zelfs sub-cellulaire distributie met een hoge resolutie te verkrijgen profielen van niet-geconjugeerde antilichamen, stellen wij een IF-benadering voor, die wordt geacht in vivo immunofluorescentie lokalisatie (IVIL), waarbij het antigeen-specifieke antilichaam in vivo intraveneus wordt geïnjecteerd. De op antilichamen gebaseerde therapeutische of diagnostische, optredend als primair antilichaam, circuleert in functionele bloedvaten en bindt zich aan zijn doel proteïne in de uiterst nauwkeurige, levende tumor omgeving. Na isolatie van in vivo-gelabelde tumoren met het primaire antilichaam, wordt een secundair antilichaam gebruikt om geaccumuleerde en bewaarde antilichaam conjugaten te lokaliseren. Deze benadering is vergelijkbaar met een eerder beschreven als histologie benadering die fluorescently gelabelde antilichamen17injecteert. Hoewel hier, het gebruik van niet-geconjugeerde antilichamen vermijdt een mogelijke verandering in bioverdelings kenmerken veroorzaakt door modificatie van antilichamen. Onze etiketterings aanpak weerspiegelt in vivo biodistributie van op antilichamen gebaseerde geneesmiddelen en doelstoffen en kan belangrijke inzichten bieden voor de ontwikkeling van nieuwe diagnostische en therapeutische middelen.
Hier beschrijven we twee toepassingen van de IVIL-methode zoals toegepast in eerdere studies die de biodistributie onderzoeken en de toegankelijkheid van antilichamen-gebaseerde contrastmiddelen voor moleculaire beeldvormings benaderingen voor borstkanker detectie. Ten eerste is de biodistributie van een antilichaam-near infrarood kleur conjugaat (anti-B7-H3-antilichaam gebonden aan de Near Infrared fluorescentie Dye, indocyanine Green, B7-H3-ICG) en het Isotype-controlemiddel (ISO-ICG) voor fluorescentie en fotoakoestisch moleculair beeldvorming is verkend18. De methode van deze toepassing wordt beschreven in het protocol. Vervolgens wordt de bioverdelings resultaten van een conformatief gevoelig antilichaam tegen netrin-1, meestal niet detecteerbaar met traditionele als beeldvorming, gebruikt met echografie moleculaire beeldvorming, gekwantificeerd en gepresenteerd in de representatieve resultaten19. Aan het einde van dit protocol papier moeten lezers zich comfortabel voelen bij het aannemen van de IVIL-methode voor hun eigen op antilichamen gebaseerde onderzoekstoepassingen.

in vivo immunofluorescence localization for assessment of therapeutic and diagnostic antibody biodistribution in cancer research

Monoklonale antilichamen (mAbs) zijn belangrijke instrumenten voor de opsporing, diagnose en behandeling van kanker. Ze worden gebruikt om de rol van eiwitten in tumorigenesis ontrafelen, kan worden gericht aan kanker biomarkers waardoor tumor detectie en karakterisering, en kan worden gebruikt voor kankertherapie als mAbs of antilichaam-drug conjugaten te activeren immune Effector cellen, remmen signalering van trajecten, of direct dodencellen die het specifieke antigeen dragen. Ondanks klinische ontwikkelingen in de ontwikkeling en productie van nieuwe en zeer specifieke mAbs, diagnostische en therapeutische toepassingen kunnen worden aangetast door de complexiteit en heterogeniteit van de tumor micro omgeving. Voor de ontwikkeling van efficiënte therapieën en diagnostiek op basis van antilichamen is het dus cruciaal om de biodistributie en interactie van het op antilichamen gebaseerde geconjugeerde met de levende tumor microomgeving te beoordelen. Hier beschrijven we in vivo immunofluorescentie lokalisatie (IVIL) als een nieuwe benadering om interacties van op antilichamen gebaseerde therapieën en diagnostiek in de in vivo fysiologische en pathologische omstandigheden te bestuderen. In deze techniek wordt een therapeutisch of diagnostisch antigeen-specifiek antilichaam intraveneus geïnjecteerd in vivo en gelokaliseerde ex vivo met een secundair antilichaam in geïsoleerde tumoren. IVIL weerspiegelt daarom de in vivo biodistributie van op antilichamen gebaseerde geneesmiddelen en doelstoffen. Twee IVIL-toepassingen worden beschreven bij het beoordelen van de biodistributie en de toegankelijkheid van contrastmiddelen op basis van antilichamen voor moleculaire beeldvorming van borstkanker. Dit protocol stelt toekomstige gebruikers in staat om de IVIL-methode aan te passen voor hun eigen op antilichamen gebaseerde onderzoekstoepassingen.

De IVIL-methode werd hier gebruikt om de in vivo biodistributie en weefsel interactie van B7-H3-ICG en ISO-ICG te onderzoeken, door de agentia, na intraveneuze injectie in een levend dier, toe te staan om met het doelweefsel te communiceren voor 96 h, en vervolgens zodra de weefsels als primaire antilichamen tijdens ex vivo-immunokleuring. Dit protocol voor in vivo immunofluorescentielokalisatie, of IVIL, beoordeelt de in vivo biodistributie van diagnostische en therapeutische antilichamen of op antilichamen gebaseerde middelen voor kankeronderzoek, detectie en behandeling. In tegenstelling tot conventionele immunofluorescente kleuring waar cellulaire expressie van antigenen ex vivo wordt onthuld, verduidelijkt de IVIL-methode hoe antilichamen en antilichaamgebaseerde agentia zich verspreiden in het levende dier. Deze video helpt bij het begrijpen van de algehele workflow van de IVIL-methode.Het toont belangrijke details voor een succesvolle reproductie van het protocol voor andere toepassingen en antilichamen. Observeer muizen van het gewenste kankermodel voor de juiste tumorgroei via palpatie of remklauwmeting voordat u verdergaat. Zuiveren konijn anti-muis B7-H3 en konijn IgG isotype controle antilichamen op een desalting kolom om conserveringsmiddelen en opslag buffers volgens de instructies van de fabrikant te verwijderen.Aliquot doseringen van 33 microgram van elk antilichaam conjugaat in individuele microcentrifuge buizen. Gebruik tangen om alleen de buitenste laag van de huid te grijpen tussen de set van borstklieren het dichtst bij de staart, en maak een kleine incisie met een paar chirurgische schaar.Introduceer de gesloten schaar in de snede en open de punt langzaam om de huid zorgvuldig te scheiden van het onderliggende buikwandmembraan, waardoor deze intact blijft. Maak een verticale incisie van de buik, blijven de huid te scheiden van het binnenste membraan. Tussen de derde en vierde borstklieren, maak een horizontale snede over de buik om intrekking van de huid en visualisatie van de borstklieren mogelijk te maken.Grijpen elke tumor of normale klier met tangen, zorgvuldig trimmen weg de bijgevoegde huid met behulp van chirurgische schaar. Plaats gesneden weefsels in weefsel wegwerp basis mallen die zijn voorgelabeld en gevuld met optimale snijtemperatuur inbedding medium. Bevries snel de mallen door plaatsing op droogijs.Om off-target levering te bestuderen, accijnzen andere weefsels of organen van belang. Met behulp van een cryostat, sectie bevroren weefsel blokken op 10-micron dikte, en plaats aangrenzende secties op vooraf gelabelde adhesie glazen dia's. Spoel bevroren weefselglijbanen met kamertemperatuur PBS gedurende vijf minuten om het inbeddingsmedium te verwijderen.Afbakenen weefselsecties met een hydrofobe barrièrepen om het volume van de oplossingen die nodig zijn tijdens het bevlekken te verminderen. Bevestig de weefselsecties met 4% paraformaldehydeoplossing gedurende vijf minuten. Na het spoelen van de dia's in PBS gedurende vijf minuten, permeabiliseren weefselsecties met 0,5%Triton X-100 in PBS gedurende 15 minuten.Spoel de glijbanen opnieuw in PBS gedurende vijf minuten. Blokkeer de weefsels met PBS met een gewicht van 3%, per volume runderserum albumine en 5% volume per volume geitenserum gedurende een uur bij kamertemperatuur. Na het spoelen van de dia's in PBS gedurende vijf minuten, inbroed de secties met het bijhouden van primaire antilichamen zoals een gemeenschappelijke nucleaire, vasculaire of cytoplasmische marker.Hier wordt rat anti-muis CD31 gebruikt bij een één-op-100 verdunning in de blokkeringsoplossing. De dia's met primair antilichaam worden 's nachts bij vier graden Celsius achtergelaten, beschermd tegen uitdroging op een schuiflade. Na incubatie, spoel de dia's in PBS gedurende vijf minuten drie keer.Wijzig de PBS telkens weer. Incubeer de dia's met secundaire antilichamen om de primaire antilichamen te etiketteren. Visualiseer voor deze toepassing anti-B7-H3-antilichaam met Alexa Fluor 546 geconjugeerd anti-konijnantilichaam van geiten en visualiseer CD31 met Alexa Fluor 488 geitenanti-rat secundair antilichaam in blokkeringsoplossing.Bescherm de glijbanen tegen licht en uitdroging op een schuiflade gedurende een uur bij kamertemperatuur. Na het spoelen van de dia's in PBS als voorheen, breng een druppel van het montagemedium in het midden van het weefsel slice. Plaats voorzichtig een coverslip, het vermijden van beknelling van luchtbellen.Sluit de randen van de coverslip af met heldere nagellak en laat drogen. Ga verder met confocale microscopie beeldvorming en kwantitatieve beeldanalyse zoals beschreven in het tekstprotocol. Representatieve confocale micrografen tonen de vergelijking aan van specifieke B7-H3-antilichaam-ICG-vervoeging en niet-specifieke isotypecontroleantilichaam-ICG vervoeging lokalisatie in murine borstklieren die normale of carcinoomweefsels bevatten.Normale murine borstklieren van een dier dat intraveneus wordt geïnjecteerd met Iso-ICG of B7-H3-ICG en tegengehouden met CD31 vertoont geen vlekken van de antilichaamconjugaten. Normale weefsels bleken geen uiting te hebben van B7-H3 door standaard ex vivo immunofluorescentie kleuring. Bij invasieve borsttumoren bindt B7-H3-ICG zich sterk aan de vasculatuur, het eerste contactpunt in vivo, en kan vervolgens extravaseren van de vasculatuur om het tumoreptheel heterotheel heterogeen te bevlekken.Iso-ICG toont niet-specifieke accumulatie in de tumorweefsels. Standaard ex vivo immunofluorescente kleuring toont een uniforme expressie van de B7-H3 marker op epitheel en en endotheelcellen. Representatieve confocale micrografen tonen de in vivo immunofluorescentielokalisatiemethode om netrin-1-expressie of isotypecontrole-expressie in murinecarcinoom en normale borstklieren te detecteren.In vivo immunofluorescentie lokalisatie bevestigt epitheel signaal voor netrin-1 in MMTV-PyMT tumoren, maar niet in normale borstklieren. Bovendien is er een sterk netrin-1 signaal op endotheelcellen in borsttumoren, zoals aangegeven door het gekoloniseerde gele signaal. Het signaal is aanzienlijk zwakker in normale borstklieren.De IVIL-methode moet worden geoptimaliseerd in termen van antilichaamdosering, weefselverzamelingstiming en secundaire verdunning van antilichamen voor een succesvolle implementatie. IVIL maakt het mogelijk om conformatie-epitopen te targeten die niet kunnen worden gedetecteerd met behulp van standaard immunofluorescentiekleuring, waarvoor weefselverwerking vereist is. Ook kunnen gezonde organen van tumordragende muizen worden verzameld om off-target levering van therapeutische antilichamen te beoordelen.Met een toenemend gebruik van op antilichamen gebaseerde therapieën en contrastmiddelen in kankeronderzoek en -beheer is de IVIL-methode zeer toepasbaar op farmaceutisch onderzoek en -ontwikkeling. Onderzoekers in kankertherapieën, moleculaire beeldvorming, ontsteking, en andere gebieden kunnen vinden dat de IVIL-methode nuttige informatie biedt.
