1. 大塔的小鼠牺牲和微解剖
<ol><li>通过CO2窒息和记录重量牺牲鼠标。</li>

	<li>在右侧灌注液排水器中切开。以颅方向插入一根27G的针,穿过心脏的顶点。将针头固定在左心室,同时缓慢地将鼠标注入10 mL冰冷的磷酸盐缓冲盐水(PBS),至少2分钟。 注:有些方案使用甲醛灌注,但这干扰了几种下游应用,如淋巴细胞的免疫性化学分析。因此,本协议中不执行甲醛灌注固定。</li>
	<li>使用解剖钳和解剖剪刀解剖感兴趣的器官(如淋巴结、脾脏、肝脏、肠道、内脏脂肪垫、肾脏等)。</li>

	<li>取出覆盖胸主塔的脂肪组织。仔细解剖胸腺的斜端,以释放主动脉拱与分支。继续在胸腔中尽可能不分泌胡萝卜动脉。在特殊情况下,可以进行颈部解剖,以包括胡萝卜分叉。</li>
	<li>在实际切割主塔之前,在去离子水、RNase 去污溶液、70% 乙醇和 PBS 中连续冲洗,清洁仪器。用钳子将心脏提起顶点。切主塔靠近心脏,把整个心脏放在一个管与PBS。心脏可以在冰上储存几个小时,然后继续处理和冷冻主动脉根。</li>
	<li>根据图1A切割主动脉拱门。将主动脉拱门放入含有1mL 4%甲醛的管中过夜,温度为4°C。标本可以以这种方式存储几年,然后再固定和分析。 警告:甲醛可能导致癌症,过敏性皮肤反应,如果吞咽是有害的。根据需要使用个人防护设备。</li>
	<li>解剖剩余的下降主方,并将其放入RNA稳定溶液中,或将其卡扣冷冻,以便后续的RNA分析或其他应用。优化工作流程,尽量减少解剖时间,对于避免RNA过度降解至关重要。</li>
	<li>将采血管(在步骤 1.3 中收集)放入离心机。在室温下,以1,500 x g将单独的血浆和血清管旋转15分钟。小心地将血浆和血清转移到微离心管中,并储存在-80°C。收集 EDTA 和肝素化血浆或血清可为多种下游应用留下可能性。</li>
	<li>将心脏放在软木床上,腹腔侧朝上。用针把心脏固定在软木塞上。用解剖钳子握住心脏底部。

2. 主动脉拱门和胸动脉的面分析
<ol><li>准备固定床,用于主动脉拱门的面部分析。折叠一段石蜡蜡膜 8 次,使表面平坦 25 mm x 25 mm。用黑色电绝缘胶带包裹,为主塔制作深色背景。在固定床的背面放置一个标签,并使用铅铅笔写鼠标识别号(正常的笔墨在染色过程中将消失)。</li>

	<li>准备苏丹四国的工作解决方案。将 1 g 苏丹 IV 粉末、100 mL 70% 乙醇和 100 mL 丙酮混合在深色瓶子中,轻轻搅拌 10 分钟。无需过滤溶液,如果在室温下保持黑暗,可以使用几个月。如果染色颜色不令人满意,可以做出新的解决方案,使标本再次染色。 警告:丙酮是一种易燃液体,可引起严重的眼部刺激。存放在通风良好的地方,处理时采取预防措施。</li>
	<li>在实验室的长凳上安排五个培养皿:一个装满了70%乙醇,一个装满苏丹IV工作溶液,两个装满80%乙醇,一个装满PBS。
	<ol>

	<li>小心地固定亚冠和胡萝卜动脉,使病变在胸腔头动脉中进行定量(图2A)。亚环大动脉和普通胡萝卜动脉的病变定量通常非常具有挑战性,没有意义。

3. 主动脉根的冷冻剖分
<ol><li>将低温温度设定在-20°C,截面厚度设置为10μm.将含有主动脉根的OCT块安装在试样支架上,心室组织朝外。开始切割时,微调截面表面的对齐方式,以便与试样支架平行。</li>
	<li>移除过多的周围 OCT,以便更轻松地收集没有折叠的截面。主动脉根现在应垂直于刀刃,因为心脏的底座正确放置在模具中。</li>
	<li>收集普通显微镜幻灯片上的初始控制部分,这些部分将被丢弃。第一部分应只包含心肌组织。将切片进度时前进 200 μm。收集部分,用光学显微镜检查进度。</li>
	<li>接近左心室流出区时,在显微镜下检查每100μm。当观察到血管壁的初始指示时,将速度减慢到50μm。 注:当出现第一个主动脉尖时,这将是收集部分的零点。很难看到尖峰何时准确出现,但准确的定位对于对同一区域的病变进行比较至关重要。</li>
	<li>
将试样倾斜至点零尖,使截面平面与另外两个尖点对齐。这对获得主塔的真正横截面至关重要。绘制主动脉根,指示其出现时出现的尖尖,并计算从零点开始切割的每 10 μm 截面。

<li>当出现第二个尖时,将试样再次从尖点稍微倾斜,使试样与第三个尖点对齐。尖尖之间的水平差不应超过50 μm。开始从90μm水平起收集幻灯片上的部分。</li>

4. 主动脉根动脉粥样硬化的油红O染色和定量
<ol><li>通过在 100 mL 异丙醇中溶解 1 克油红 O 溶液,制备饱和油红 O 溶液。在室温下将溶液在暗瓶中搅拌1小时。饱和溶液可以保存数月。 注:建议指定实验室设备用于油红 O 染色,因为很难清洁与溶液接触的设备。</li>
	<li>通过将 75 mL 的饱和油红 O 溶液与 50 mL 的去离子水混合,准备工作溶液。在室温下站立10分钟,通过定性滤纸过滤。</li>
	<li>将幻灯片放入油红色 O 工作溶液中 20 分钟,在自来水中冲洗 5 分钟。</li>
	<li>为协助组织可视化,用迈尔的血氧素染色1分钟,在温水自来水中冲洗5分钟。所有核现在应该以蓝色染色,调整染色时间以优化结果。</li>
	<li>将滑轨安装在水性安装介质中(例如,凯撒的甘油明胶)。热凯撒的甘油明胶到40°C,使其在使用前流动。无需干燥滑轨,因为安装介质是水基的。添加盖玻璃时,小心避免气泡形成。 警告:Kaiser的甘油明胶含有苯酚,怀疑引起遗传缺陷。根据需要使用个人防护设备。</li>

quantification of atherosclerosis in mice

油红O是一种脂溶性鲜红的二氮染料,它污渍中性脂质。细胞膜中的极性脂质没有染色。油红O染色可以在新鲜、冷冻或正式固定样品上进行,但不能在石蜡嵌入样品上进行,因为在所需的脱蜡过程中去除脂质。病变脂质积累可以通过颜色阈值对总病变区域的油红O正面积进行定量(图4C)。血氧林产生细胞核的蓝色染色,有助于形象化斑块形态。左右冠状动脉通常偏离主动脉约250 μm从主动脉27,这往往与最突出的病变大小重合。来自此区域的横截面通常显示为具有代表性的结果 (图 4D)。

Quantification of Atherosclerosis in Mice
