pan-myeloid differentiation of human cord blood derived cd34+ hematopoietic stem and progenitor cells

このプロトコルは、ヒトCD34陽性造血幹細胞および前駆細胞の正常な骨髄異分化中に起こる分子および細胞変化を研究するために使用することができる。このプロトコルの主な利点は、巨核球、赤血球、顆粒球、および単球を含む4つの骨髄系すべてに沿ってCD34陽性細胞の分化を可能にすることです。手順を実証するのは、私の研究室のポストドクターであるアディティ・バパットです。ヒト臍帯血試料からの単核細胞分離のために、70%エタノールで臍帯血の袋の外表面を殺菌してから、生物学的安全キャビネット内の無菌250ミリリットルのペットボトルに袋内容物を移す。室温ハンクの緩衝生理食塩水の等量で血液を希釈し、5つの無菌50ミリリットル円錐形チューブのそれぞれに15ミリリットルの単核細胞分画培地を加える。穏やかな混合の後、慎重に各チューブの単核分画媒体に希釈臍帯血の30ミリリットルを注ぎ、層を混合するのを避けるために角度で50ミリリットルのチューブを保持する。細胞を密度勾配遠心分離で分離し、単核細胞層の上方の血漿の上に15〜20ミリリットルを除去する。ピペットを使用して、露出した単核細胞層を新しい50ミリリットルチューブに慎重に移し、単核細胞を2〜3本のチューブにプールし、各チューブの最終体積を冷たいPBEバッファーで最大50ミリリットルにします。ペレットは遠心分離により単核細胞試料を、上清を吸引する。各チューブを軽くタップして細胞ペレットを緩め、ペレットを5ミリリットルの氷冷PBEバッファーで希釈します。3~4本のチューブを1本の50ミリリットル円錐形チューブに組み合わせ、5ミリリットルの新鮮なPBEバッファーですべてのチューブを洗浄して残りの細胞を採取します。カウント後、新鮮な氷冷PBEバッファーでチューブ内の最終体積を最大50ミリリットルにし、遠心分離によって細胞を収集します。CD34陽性単核細胞を単離するには、ペレットを新鮮なPBEバッファーの50ミリリットルで再懸濁してから、別の遠心分離で細胞を採取する。上清を捨て、穏やかにタップしてペレットを緩め、氷冷PBEバッファー濃度の300マイクロリットルあたり8個の単核細胞に10回で細胞を再中断します。Fc受容体遮断試薬100マイクロリットルで非特異的結合をブロックし、穏やかに混合し、磁気活性化細胞選別ヒトCD34マイクロビーズキットから10回10回から8細胞に穏やかな混合で100マイクロビーズを加え、セルを摂氏4度で30分間インキュベートします。細胞がインキュベートしている間、LSカラムと分離前フィルタをMACSセパレータの磁場に入れてください。2ミリリットルの氷冷PBEバッファーと1ミリリットルのバッファーで分離前フィルターでカラムを平衡化します。列を15ミリリットルの円錐管に空にし、フロースルーを破棄します。インキュベーションの最後に、単核細胞サンプルの最終体積を新鮮な氷冷PBEバッファーで最大15ミリリットルにし、遠心分離によって細胞を沈沈める。氷冷PBEバッファーの1ミリリットルにペレットを再中断し、カラムの分離前フィルターに細胞をロードします。新鮮な氷冷PBEバッファーの3ミリリットルでチューブをリンスし、分離前フィルターにバッファーを追加します。フィルターなしで1回の洗浄ごとに3ミリリットルの氷冷PBEバッファーでカラムを4回洗浄し、各洗浄の間にカラムを完全に排出できるようにします。最後の洗浄の後、新しい15ミリリットルチューブにカラムを移動し、5ミリリットルの新鮮な氷冷PBEをカラムからチューブに突き落とします。新たに単離されたCD34陽性造血幹細胞および前駆細胞における幹細胞および前駆細胞集団の画分を決定するために、遠心分離によって細胞を収集し、ペレットをフローサイトメトリーバッファー濃度の50マイクロリットル当たり6細胞に1回10回再懸濁する。次に、実験フローサイトメトリー染色プロトコルに従って5ミリリットルの蛍光活性化細胞選別チューブ当たり50マイクロリットルのアリコートで細胞を移動させる。適切な抗体カクテルと生きた/死んだ染色を細胞に加え、合計100マイクロリットルの体積に調整します。光から保護された暗闇の中で20分後、チューブあたり1ミリリットルの新鮮なフローサイトメトリーバッファーで細胞を洗浄し、チューブ当たりの新鮮なフローサイトメトリーバッファーの500マイクロリットルで細胞を再中断します。次に、標準的なフローサイトメトリックプロトコルに従ってフローサイトメーター上の細胞を解析します。微生物分離CD34陽性幹細胞および前駆細胞を骨髄系細胞に分化するために、まず、PBS中の組換えヒトフィブロネクチン溶液のウェルあたり200マイクロリットルの無菌、コーティングされていない48ウェルプレートのウェルを室温で2時間コーティングする。インキュベーションの終わりに、フィブロネクチン溶液を各ウェルから捨て、2%ウシ血清アルブミンの200マイクロリットルでウェルをブロックします。室温で30分後、1回の洗浄につき500マイクロリットルの無菌PBSで井戸を2回洗浄します。新たに単離したCD34陽性細胞を、温かい刺激媒体濃度のミリリットル当たり5つの細胞を5倍10回で再懸濁する。次に、フィブロネクチンフラグメント被覆板の各ウェルに200マイクロリットルの細胞をシードし、48時間細胞培養インキュベーターにプレートを入れる。インキュベーションの終わりに、穏やかなピペットで細胞を収集し、500マイクロリットルの加温されたダルベッコの改変イーグル培地でウェルを洗浄し、細胞懸濁液で洗浄をプールします。カウント後、細胞を2.5倍10回で、骨髄分化培地濃度の1ミリリットル当たり4細胞に再懸濁した。層200マイクロリットルのCD34陽性細胞をMS-5間質細胞で播種した48ウェル組織培養プレートに含まれる。次いで、プレートを細胞培養インキュベーターに入れ、各ウェルの上澄み液100マイクロリットルを、3〜4日ごとに100マイクロリットルの新鮮な2X骨髄分化培地にそっと置き換える。21日目に、ちょうど実証したようにフローサイトメトリーによる骨髄系マーカー発現の分析のために細胞を収穫する。80~90%の免疫平ノティック分析の範囲として、コードビーズ分離によって臍帯血から分離された全CD34陽性細胞の典型的な割合は、これらのCD34陽性細胞が典型的に約20%の系統陰性CD34陽性CD38陽性CD38陽性幹細胞集団および72%リネージ陰性CD34陽性CD38多増殖細胞集団で構成されることを明らかにした。多強力な前駆細胞集団内では、細胞の約25%が一般的な骨髄性前駆腫マーカーを発現し、約15%が顆粒球単球前駆細胞マーカーを発現し、約56%が赤血球前駆体マーカーを発現する。免疫フェノタイピングは、CD34陽性細胞の割合が、21日目に単離時の約90%から25%未満に減少することを示している。CD34陰性集団の分析は、成熟した骨髄系マーカーを発現する細胞の割合の増加を示している。さらに、ライト・ギムサ染色細胞は、1日目と21日目に、大きな丸い核と非常に少ない細胞質を示す未分化培養物と、単球、顆粒球、および赤芽球を含む系分細胞の形態学的特徴を示す分化培養物との明確な形態学的特徴を示す。層を混合することなく、単核細胞分画培地に希釈された臍帯血を注ぎ、培地を交換する際にMS-5およびCD34陽性細胞を乱さないことを忘れないでください。この分化プロトコルに従って、CD34陽性細胞培養は、形態学的変化、アポトーシス効果、細胞周期パターンの変化、および分化の程度について分析することができる。
