Skyll platen tre ganger med PBS og la den stå under ultrafiolett lys over natten.Før cellesåing, pre-varm spredning medium, PBS løsning, og trypsin EDTA løsning til 37 grader Celsius. Kast det overnaturlige fra cellekulturkolben og vask det samtidige periodontale ligamentet, eller PDL-cellene, med 10 milliliter PBS. Kast PBS og tilsett en milliliter trypsin EDTA-løsning i kolben.Inkuber kolben ved 37 grader Celsius i tre minutter, og legg deretter til tre milliliter spredningsmedium for å avslutte trypsinfordøyelsen. Overfør cellesuspensjonen til et konisk rør på 15 milliliter og sentrifuger den i fem minutter ved 300 ganger g. Kast de overnaturante og re-suspendere cellene i 500 mikroliter av spredningsmedium.Tell cellene med et hemocytometer og frø dem på platen på fem, 10, 30 og 60 tusen celler per kvadratcentimeter. Kultur PDL-cellene i en inkubator ved 37 grader Celsius i 5% karbondioksid og fuktet luft, og endre kulturmediet to ganger i uken. Observer cellemorfologi daglig i fem dager via et omvendt fasekontrastmikroskop.Levedyktige PDL celle spheroider er vellykket dannet ved hjelp av denne protokollen. Sfæroider observeres sjelden for lavere såetetthet på dag en og tre. Men ved de to høyere tetthetene er ulike størrelser av sfæroider tilstede fra dag én.Den optimale såetettheten er 30 tusen celler per centimeter kvadrert fordi det resulterer i homogene sfæroider. Etter fem dager med kultur observeres større sfæroider for alle såetettheter. En levedyktighetsanalyse viser at de fleste PDL-celler er i live på dag én, tre og seks, men antall døde celler øker på dag seks.En begrensning av denne protokollen er redusert spredning av celler etter sfæroiddannelse. Så ytterligere studier for å fremme spredning av celler etter sfæroiddannelse er nødvendig.
