Epigenetiske modifikationer spiller vigtige roller under hjernens udvikling, modning og funktion. Som en stabil markør for DNA modifikation reagerer Dynamic 5-hmC på adfærds tilpasning, neuronal aktivitet og er positivt korreleret med genekspression; således er det involveret i den normale funktion af hjernen og neurologiske lidelse4. For at undersøge dets funktion i celler og væv, er det nødvendigt at påvise eksistensen af 5-hmC og sammenligne niveauet før og efter behandling. Her demons...

Epigenetiske modifikationer, herunder DNA modifikation, Histon modifikation og RNA modifikation, har vist sig at spille væsentlige funktioner i forskellige biologiske processer og sygdomme1,2,3, 4 , 5 , 6 , 7. i lang tid, DNA-methylering (dvs., 5-methylcytosin (5-mC)) er blevet betragtet som en meget stabil epigenetisk markør og kan ikke ændres yderligere i genomet. For nylig, det har været konstateret, at 5-mC kunne oxideres til 5-hydroxymethylcytosin (5-HMC) af tet (ti-elleve translocations) familie proteiner, herunder TET1, TET2,og TET3 8,9. Der er flere tilgange til påvisning af 5-HMC i celler og væv14,21,22,23,24. Her beskriver vi to metoder til at påvise eksistensen af 5-hmC og kvantificere det globale niveau af 5-hmC: immunofluorescens farvning og DNA dot-blot. Disse to metoder er bekvemme og følsomme, og er blevet anvendt med succes i tidligere undersøgelser25,26,27,28,29,30. De vigtigste trin i disse to metoder er DNA-denaturering. For immunofluorescens farvning af 5-hmC kræves forbehandling af prøver med 1 M HCl. For 5-hmC dot-blot er DNA-denaturering udført med NaOH-opløsning. Disse to metoder sammen med næste generations sekvensering er meget nyttige værktøjer til at undersøge funktionen af 5-hmC.

Her præsenterer vi en protokol til påvisning af 5-hydroxymethylcytosin i celler og hjernevæv ved hjælp af immunfluorescens, den nye metode og DNA-dot-blot. 5-hydroxymethylcytosin, dvs 5hmC, afbødes epigenetisk modifikation, spiller en vigtig funktion i reguleringen af genekspression og involveret i flere neurologiske lidelser.Faktisk kan vores metoder bruges til at detektere 5hmC i flere celler og væv. Det er praktisk og kan udføres med fælles udstyr i laboratoriet. Begynd med at placere musen med ansigtet på en plastik bord og fastsættelse af hver lemmer med tape.Brug kirurgisk saks til at skære gennem huden og musklen, og åbne brysthulen. Eksponere hjertet og afbrød en lille del af højre atrium med fine kirurgiske saks. Brug en 10-milliliter engangstændere, steriliseret sprøjte, perfuse musen fra venstre hjertekammer med kolde PBS.Derefter, gennemgå musen med 4% PFA, indtil det er stiv. Derefter åbne kraniet med knoglekræft og fjerne hjernen. Placer hjernen i en 15-milliliter centrifuge rør fyldt med fem milliliter 4% PFA og gemme det på fire grader Celsius.Efter 24 timer, overføre hjernen til en 30% saccharose opløsning og lad det ved fire grader Celsius for fuldstændig dehydrering. Før sektionsskæring, imbed hjernen i optimal skæretemperatur sammensatte og afkøles til minus 20 grader Celsius i mindst en time. Sektion hjerneprøver ved en tykkelse på 20 til 40 mikrometer ved hjælp af en kryostat mikrotom.Saml sektioner i PBS og opbevares ved fire grader Celsius. Afhente hjernen sektioner af interesse og læg dem i en 24-brønd plade med PBS. Sektionerne vaskes med PBS på en shaker i 10 minutter.Fjern PBS og behandle med forvarmet en-molar saltsyre ved 37 grader Celsius i 30 minutter. Saltsyre er ætsende og forberede det i den kemiske hold. Derefter vaskes prøverne tre gange med PBS i fem minutter, og de blokeres med PBS, der indeholder 3 % normalt gedeserum og 0,1 % Triton X-100.Lad prøverne blive på en shaker ved stuetemperatur i en time. Tilsæt 600 mikroliter chloroform til supernatanten, bland grundigt og centrifuge i henhold til manuskriptretninger.Overfør supernatanten til et nyt rør og tilsæt 500 mikroliter isopropanol. Efter centrifugering, kassér supernatant, og vask DNA-pellet med 70% ethanol i henhold til manuskript retninger.Lufttørre DNA-pellet helt og derefter opløse det i Tris-HCl buffer. Denature DNA-prøven ved at opvarme den til 100 grader Celsius i 10 minutter og derefter placere den på is.Skær den passende størrelse af en nylonmembran og skyl den med 6X SSC. Placer membranen på et dot-blot apparat og tilslut vakuumpumpen. Spot seks-mikroliter prikker på membranen og hybridisere i 30 minutter ved 80 grader Celsius.Derefter blokere prøvemembranen med fedtfri mælk og Tris-buffered saltvand, eller TBS, i en time. Tilsæt polyklonal kanin anti-5-hydroxymethylcytosin antistof til membranen og inkubere natten over ved fire grader Celsius. Den næste dag, lad prøvemembranen ved stuetemperatur i en time, derefter vaske membranen tre gange med TBS.Inkuber membranen med det sekundære antistof mod kanin i 30 minutter, og vask derefter tre gange med TBS. Visualiser chemiluminescenssignalerne og kvantificere deres intensitet. Denne protokol kan bruges til at visualisere 5-hydroxymethylcytosin, eller 5hmC, i hippocampus af voksne mus.Immunfluorescens med antistoffer mod neuronale celler, 5hmC afslører, at der er en berigelse af 5hmC i neuroner. Dynamikken i 5hmC under neuronal udvikling kan bestemmes med en dot-blot assay af DNA-prøver isoleret fra prolifererende og differentierede voksne neurale stamceller. Globale niveauer af 5hmC stige betydeligt under differentiering og niveauet af 5hmC i neuroner er højere end i neurale stamceller.Det vigtigste trin i denne metode er at sikre, at fuldstændig denaturering af DNA-prøver.
