급성 관상 동맥 증후군 (ACS)은 서방 세계1,2에서사망의 주요 원인입니다. 관상 동맥의 급성 폐색은 영향을받는 심장 조직의 허혈성 캐스케이드 및 괴사의 활성화로 이어진다3. 손상된 심근은 점차 적으로 비 수축성 흉터 조직으로 대체되며, 이는 임상적으로 심부전으로 매니페스트4,5. ACS의 처리에 있는 최근 어드밴스에도 불구하고, ACS 와 ACS 관련 심부전의 보급은 상승하고, 치료 선택권은 제한됩니다6,7. 따라서 ACS와 그 합병증을 연구하기 위한 동물 모델을 개발하는 것은 엄청난 관심사입니다.
현재까지 ACS 및 ACS 유도 심근 리모델링을 연구하기 위해 가장 널리 사용되는 동물 모델은 좌측 내림차순 관상 동맥(LAD)의 결찰이다. LAD의 결찰은 ACS 동안 인간의 심근 조직과 유사한 심근의 급성 허혈을 유도합니다.  그러나, 일치하지 않는 경색 크기는 LAD 결찰의 아킬레스건 발 뒤꿈치 남아있다. LAD의 외과적 변이 및 해부학적 가변성은 일관되지 않은 경색 크기로 이어지고 이 절차의 재현성 및 복제성을 저해하는8,9,10. 또한, LAD 결찰은 높은 수술 후 사망률을 가지고 있다. 재현성을 향상시키고 사망률을 줄이기 위한 최근의 노력에도 불구하고11,12,많은 수의 동물이 여전히 리모델링 방지 요법을 제대로 평가할 필요가 있다.
ACS의 대체 모델은 무선 주파수13,열14 또는 극저온 상해15,16,17,18을포함하여 최근 몇 년 동안 제안되고 연구되었다. 현재 의 극저온 손상 방법은 액체 질소에 미리 냉각된 금속 막대를 적용하여 피험자의 심장 조직을 손상시키는15,16. 그러나 이 절차는 충분한 경색 크기를 생성하기 위해 여러 번 반복되어야 합니다. 조직에 비해 막대의 높은 전도도 및 낮은 열 용량으로 인해 프로브가 빠르게 따뜻해지고 조직이 이질적으로 냉각됩니다(따라서 경색)됩니다. 이러한 한계를 극복하기 위해, 우리는 휴대용 액체 질소 전달 시스템을 활용하는 극저온 절색 모델을 설명하기 위해 여기에 기술한다. 이 모델은 재현 가능하고 수행하기 쉬우며 빠르고 안정적으로 확립할 수 있습니다. 관상 동맥 해부학의 독립적인 재현가능한 경색 병변은 결국 심장 실패로 이끌어 내는 생성됩니다. 이 방법은 신규한 치료 적 약리학 및 조직 공학 기반 전략의 평가를 위한 리모델링 공정을 연구하는 데 특히 적합하다.
