Острый коронарный синдром (АКС) является ведущими причинами смерти в западном мире1,2. Острая окклюзия коронарных артерий приводит к активации ишемического каскада и некроза пораженной сердечной ткани3. Поврежденный миокард постепенно заменяется неконтрактной рубцовой тканью, которая клинически проявляется как сердечная недостаточность4,5. Несмотря на последние достижения в лечении ACS, распространенность ACS и ACS связанных сердечной недостаточности растет, и терапевтические варианты ограничены6,7. Поэтому разработка моделей животных для изучения ACS и его осложнений представляет огромный интерес.
На сегодняшний день наиболее широко используемой моделью животных для изучения ACS и ВЫЗВАННОй ACS миокарда ремоделирование является перевязка левой нисходящей коронарной артерии (LAD). Лигация ЛАД приводит к острой ишемии миокарда, подобно ткани миокарда человека во время АКС.  Тем не менее, непоследовательные нефарктные размеры остаются ахиллесовой пятой перевязки LAD. Хирургические вариации и анатомическая изменчивость LAD приводят к несовместимым недопустимым размерам и препятствуют воспроизводимости и релицируемости этой процедуры8,9,10. Кроме того, лигация LAD имеет высокую внутрихирургическую и послеоперационную смертность. Несмотря на недавние усилия по улучшению воспроизводимости и снижению смертности11,12, большое количество животных по-прежнему необходимо правильно оценить анти-ремоделирования терапии.
Альтернативные модели ACS были предложены и изучены в течение последних лет, в том числе радиочастоты13, тепловые14 или криогенных травм15,16,17,18. Текущие методы криотравмы применяют металлический стержень, предварительно охлажденный в жидком азоте, чтобы повредить сердечную ткань субъекта15,16. Тем не менее, эта процедура должна быть повторена несколько раз, чтобы создать достаточный размер инфаркта. Из-за высокой проводимости и низкой тепловой емкости стержня по сравнению с тканью, зонд нагревается быстро, и ткань охлаждается (и, таким образом, infarcted) неоднородно. Чтобы преодолеть эти ограничения, мы описываем здесь модель криоинфаркции с использованием ручной системы доставки жидкого азота. Эта модель воспроизводима, проста в исполнении и может быть установлена быстро и надежно. Генерируется воспроизводимое трансмурльное инфарктное утранжное, независимое от коронарной анатомии, что в конечном итоге приводит к сердечной недостаточности. Гистологическое окрашивание трихромом Массона выявляет образование трансмуральной фиброзной ткани в месте травмы.
