我々の方法は、新しい非プロットおよび毛髪マトリックスにおけるコルチゾールを測定するための多くの将来の研究のためのプラットフォームとなり得る新しい代替ストレス指標を導入する。この技術の利点は、そのシンプルさと、殺処分された動物でもストレスレベルを評価するために使用される能力です。この技術を使用して高コルチゾールレベルと健康な動物や人間との関係を確立することは、いくつかの疾患の診断に役立ちます.この方法は、サンプル寿命内のストレスレベルを評価するために、これらの古い化石のような何世紀も前の生物学的サンプルを研究するためにも使用できます。コルチゾール抽出の場合、サンプルを個々の15ミリリットル円錐形チューブに入れる前に、0.0001精度のデジタル分析スケールで各サンプルの300プラスマイナス10ミリグラムを計量します。各サンプルチューブに3ミリリットルのイソプロパノールを加え、コルチゾールを洗い流し、潜在的な外部汚染を取り除くために、毎分80回転でチューブを2.5分間回転させます。最後の洗浄の後、空気は、洗浄ごとに新鮮な超純水を使用して実証されるように、サンプルをさらに3回洗浄する前に、室温で7日間サンプルを乾燥させます。75プラスマイナス5ミリグラムの乾燥フィンまたは顎骨サンプルを計量し、ビーズビーターを使用して最終的に50ヘルツで32分間サンプルを粉砕します。次に、1.5ミリリットルのメタノールを粉末フィンまたは顎骨の各チューブに送り、室温で18時間毎分40回転でチューブローテーターにサンプルを置きます。コルチゾール抽出後、遠心分離によりサンプル組織を沈下する。そして、各サンプルから個々の1.5ミリリットルのマイクロ遠心チューブに黄色がかった有機コルチゾール含有層のトップ1ミリリットルを移す。コルチゾールサンプルをヒュームフードで摂氏38度で乾燥させ、メタノールを蒸発させます。翌朝、各チューブと渦に400マイクロリットルのPBSを加え、サンプルを遠心分離してコルチゾールを採取します。ELISA分析の場合、抽出された各標準コルチゾールサンプルの25マイクロリットルをロードし、96ウェルELISAプレートの適切なウェルに重複して制御します。25マイクロリットルのアッセイ希釈液を2つのウェルに積み込み、ゼロとして機能し、各非特異的結合ウェルに入れる。15マイクロリットルの酵素コンジュゲートとアッセイ希釈液24ミリリットルを混合し、各ウェルに200マイクロリットルのコンジュゲートを加えます。回転器のプレートを5分間5分間混ぜます。その後、室温で1時間のインキュベーションを行う。インキュベーションの終わりに、ウェルあたり1xウォッシュバッファの300マイクロリットルでプレートを4回洗います。洗い物をするたびに洗い物の上にプレートを素早く反転させ、バッファーを捨て、ペーパータオルの積み重ねにプレートを消します。最後の洗浄の後、TMB基板溶液200マイクロリットルを各ウェルに加え、1分間に500回転で5分間回転器にプレートを置きます。混合後、プレートを光から保護した室温で25分間インキュベートします。インキュベーションの終わりに、各ウェルに50マイクロリットルの停止溶液を加えます。そして、1分間に500回転で、またはすべての井戸が緑色から黄色に変わるまで、回転器のプレート内容物を3分間混ぜます。次に、サンプルの光学濃度を読み取り、マイクロプレートソフトウェアを使用して、各サンプルのコルチゾールレベルを1ミリグラムあたりのピコグラムに変換します。この代表的な分析では、コルチゾールレベルは、水で洗浄されたものよりもイソプロパノールで洗浄されたフィンサンプルでは高い傾向があったが、チョウザメ種の間ではフィンコルチゾールレベルに有意な差は認められなかった。洗浄溶媒とチョウザメ種との間に有意な相互作用はなかった。ハメオの顎骨がフィンの代替マトリックスとして使用されるかどうかを判断するためのH.Huso顎骨からのコルチゾールの検査は、洗浄溶媒がH.Husoのチョウザメで測定されたコルチゾールレベルに有意な影響を及ぼさないことを明らかにした。さらに、このデータは、テストされた3種のチョウザメ種とH.Huso顎骨のフィン間で高い類似性を明らかにした。ビーズビーターを使用してサンプルを微粉末に粉砕し、適切な商用ELISAアッセイキットを使用してコルチゾール測定を成功させるために重要です。この方法は、コルチゾールレベルの測定が動物生物学にどのような影響を与えるかに関する重要な情報を提供するので、歯および他の硬質マトリックス中のコルチゾールを検出するためにも使用することができます。この方法は、ヒトの小児乳歯および小児内分泌学、心理生物学、行動、考古学、および法医学研究におけるコルチゾールレベルを評価するための新しいアプローチとして使用することができる。
