1. 加罗斯微孔模的制备

</ol>3. 在阿加罗斯微井上将hPSC区分为3D肝球

<li>将2克聚HEMA溶解在100 mL的95%乙醇中。在55°C下使用热板搅拌溶液过夜。每孔24孔板加入250μL的聚HEMA溶液,使用烤箱在60°C干燥过夜。</li>

	<li>将肝球转移到多HEMA涂层井中。
	<ol>
<li>去除KSR/DMSO培养基后,用肝细胞成熟培养基清洗一次细胞,无需补充,并加入1mL肝细胞成熟培养基,辅以10纳克/mL HGF和20纳克/mL OSM。</li>
		<li>使用 P1000 移液器,通过多次上下移液,将肝球从胶合板中提起。</li>
		<li>将含有肝球的介质转移到涂覆的多 HEMA 井中。</li>

</ol>4. 长期培养3D肝球的功能分析
<ol><li>切换到肝细胞成熟培养基,辅以10μM HCC、L-谷氨酰胺、1%青霉素/链霉素(最终浓度分别为100IU/mL和100μg/mL)和10纳克/mL HGF 48h,然后进行功能分析。</li>

使用细胞色素 (CYP) P450 测定分析肝细胞代谢功能。

<li>用1mL新鲜肝细胞成熟培养基取代培养基,辅以50μM荧光素-6'-五氟化醚(荧光素-PFBE)基板,以检测CYP3A基底活性或100μM荧光素甲基醚(荧光素-ME)基板检测CYP1A2基础活动(复制次数 = 3)。使用组织培养培养作为负对照。 注:为了避免交叉反应,建议不要使用含有3D肝球的相同井来测试不同的CYP P450活动,而是在单个井中执行。</li>

胚胎干细胞系(H9)或诱导多能干细胞系(P106)的三维聚集体使用我们定义的程序对肝细胞系进行了分化(图1)。在肝细胞规范之前,多能干细胞首先被充为明确的内皮。之后,肝细胞成熟成3D肝球,可以在培养中维持长达一年23。
为了研究3D球体的结构,30天老的hESC-或iPSC衍生球体被固定、分割和染色,以检测肝细胞和等位细胞中表达的蛋白质的存在。肝细胞核因子4α(HNF4+)和中间体标记维门丁被采用,揭示了由肝细胞组成的外层的存在,如围绕一个中间体细胞核心的细胞(图2)。我们跟踪这些实验,分析肝细胞中表达的蛋白质的表达:白蛋白、CYP3A和E-cadherin。免疫染色表明,这些蛋白质的表达仅限于球体的外层(图3)。
在30天培养的月份对肝球进行了功能分析。CYP1A2和CYP3A是功能性肝细胞中的重要酶。使用已建立的测定评估他们的活动。H9衍生肝球表现出CYP3A活性220,375 ~ 74514 RLU/mL/mg蛋白和CYP1A2活性732,440 = 33,330 RLU/mL/mg蛋白(图4A)。P106衍生肝球中的CYP3A活性为132117~43,391 RLU/mL/mg蛋白,CYP1A2活性为409,907~121,723 RLU/mL/mg蛋白(图4B)。与两批人类原发性肝细胞相比,3D肝球表现出可观的CYP活性10水平。
对白蛋白和α-牙蛋白(AFP)的合成和分泌分析表明,H9衍生的肝球分泌683.9± 84和159 = 20纳克/mL/24 h/mg蛋白的白蛋白和α-胎蛋白蛋白(图5A)。而P106衍生的肝球分泌497×41和756~24纳克/mL/24 h/mg蛋白的白蛋白和α-胎蛋白(图5B)。
