パラスポニアは、根茎と窒素固定共生を確立することができる大麻ファミリーの熱帯の木です。パラスポニアと豆類の比較研究は、根茎共生の根底にある中核的な遺伝的ネットワークに関する洞察を提供することができる。このプロトコルにより、パラスポニア・アンダーソンイの安定したトランスジェニック突然変異株は、2~3ヶ月の期間で生成することができる。これらのラインは、インビトロ伝播を通じて効率的に伝播することができる。このプロトコルは、この熱帯樹木の生物学の他の側面と同様に根茎の共生を研究するための実験的なプラットフォームを提供します。組織培養および分子クローニングにおける基礎的なスキルは、このプロトコルを実装するのに十分である。これにより、あらゆる研究室がパラスポイアを研究モデルとして適応させることができます。この手順のデモンストレーションは、博士課程の学生であるティティス・ワルダニとユダ・ロスワンジャヤと、私たちの研究室の技術者であるマリケ・ハートグです。パラスポニア・アンダーソンイの木を育てるには、種子を発芽させることから始めます。パラスポニアベリーから種子を取り除き、茶ふるいの内側にこすったり、ティッシュペーパーの上に押しつぶしたりします。4%次亜塩素酸漂白剤を使用して種子を15〜20分間消毒し、殺菌水で種子を6回洗浄します。種子を滅菌200マイクロリットルPCRチューブに移し、チューブに殺菌水を充填して種子を水没させます。原稿の指示に従って、サーモサイクラーで10日間チューブをインキュベートします。10日間のインキュベーションの後、種をSH-0プレートに移し、2層の弾性シールホイルで閉じて乾燥を防ぎます。16時間、8時間の夜のサイクルで摂氏28度で3〜4週間インキュベートします。苗木が真の葉の最初のセットを開発したら、商業ポッティング土壌で満たされたポットにそれらを転送し、乾燥を防ぐために半透明のプラスチックカップでそれらをカバーしています。16時間、8時間の夜のレジメンの下に28度摂氏、85%の相対湿度気候室または温室にポットを置きます。1週間後、プラスチックカップを取り外します。定期的にポットに水を与え、木の成長を維持するために肥料で補います。温室で栽培された木から5〜8センチメートルの若い枝を収穫し、健康で感染していない枝のみを使用することで、変革に備えます。葉を切り落とし、各ペティオールの端に葉組織の1センチメートルの正方形を残します。ポリソルベート20の数滴を含む2%次亜塩素酸漂白剤で15分間組織を消毒し、オートクレーブ水で8回洗い流します。25ミリリットルの浸潤培地でアグロバクテリウム細胞を光学密度約5に再懸濁する。ステムとペティオール組織を細胞懸濁液の中の1センチメートルに切ります。葉組織を取り除き、幹細胞とペチオール片を細胞懸濁液中に10~30分間インキュベートします。ティッシュピースを無菌のろ紙に乾かして、原稿の指示に従って準備した根付き培地に置きます。2日間摂氏21度で暗闇の中でプレートをインキュベートします。2日後、プレートの真菌や細菌の汚染を検査し、汚染されたプレートを廃棄します。原稿の指示に従って調製したSH-10培地の10ミリリットルに組織片を移す。少なくとも10分間、培地に組織を残し、2〜3分ごとに穏やかに攪拌して洗浄します。原稿の指示に従ってセフォタキシムとカナマイシンで根付き培地を準備し、プレートを注ぎます。濾紙の滅菌片に組織を乾燥させ、プレートに移します。16時間、8時間の夜のレジメンの下で摂氏28度で7日間プレートをインキュベートします。2日ごとにプレートに真菌や細菌の汚染がないか確認してください。汚染が発生した場合は、非感染組織片を新鮮なプレートに移します。7日後、原稿の指示に従って準備された伝播培地に組織片を移し、16時間、8時間夜のレジメンの下で摂氏28度でインキュベートする。トランスジェニック芽が発生するまで週に1回プレートをリフレッシュし、感染していない組織片を新しいプレートにのみ転送します。推定トランスジェニック芽が少なくとも1センチメートル長い後、芽を切断し、抗生物質で伝播培地で独立して培養する。シュートが独立した形質転換体を表していることを確認するには、外植の両側から1回の撮影のみを行います。伝播媒体の新鮮なプレートに約10のシュートを置き、弾性シールホイルの2つの層でプレートを閉じることによって、組織を伝播します。16時間、8時間の夜のレジメンの下で28度のプレートをインキュベートし、4週間ごとにこのステップを繰り返します。撮影の長さが1センチメートルに達したら、ベースに切り、根付き媒体に置きます。撮影の基礎先端を媒体に挿入して、シュートを直立位置に配置します。1枚のプレートに約10本のシュートを配置できます。根茎菌を準備することで始めます。MESorhizoium PLURIFARIUM BOR2の単一コロニーから10ミリリットルの液体YEM培地を接種し、28°Cで2日間インキュベートする。この培養液を使用して、より多くの液体YEM培地を接種する。一旦増殖し、培養物を3、500倍gで10分間遠心し、細胞を収穫する。細菌ペレットを液体EKM培地に再懸濁し、光学濃度を決定する。約20ポットで十分な3リットルのEKM培地を用意し、根茎懸濁液で接種します。1.25キログラムのパーライトと培地を混合し、無菌半透明のポリプロピレンポットにこの混合物の210グラムを追加します。各ポットに1〜3個のパラスポニア・アンダーソンイ・プランツを植え、コントロールコンストラクトで形質転換したプランレットを使用していくつかのポットを準備します。ポットの数を計量し、各ポットの底を覆って根を光の露出から保護します。16時間、8時間の夜のレジメンの下で摂氏28度の気候化された成長室でポットを4〜6週間インキュベートします。週に一度、水の損失を決定するためにいくつかのポットの重量を量る。水の損失が10ミリリットルを超える場合は、損失を補うために超純水で補います。インキュベーション後、パーライトから根をきれいにし、双眼鏡を使用して結節数を決定します。この技術は、アグロバクテリウム株AGL1でパラスポニア・アンダーソンii茎のペチオールおよびセグメントを正常に変換するために使用されています。まず、組織外植物を2日間アグロバクテリウムと共培養する。長期の共同培養は細菌の過剰コロニー形成をもたらし、予防すべきである。数週間後、小さな緑色のマイクロカリが元の創傷表面に沿って観察される。これらのcalliは、変換の開始から6〜8週間後に1つ以上の推定変換された芽を成長させ、発達し続けています。成熟した枝から成長する若い枝の組織外植の組織外植物の変換効率は、成熟した枝から65〜75%まで10〜30%変化します。トランスジェニック芽は、インビトロ伝播を介して乗算することができ、1ヶ月以内に数十の芽を生み出す。これらの芽は、約2週間で根形成を誘導し、実験に使用することができる植物を収量する根源培地に置くことができます。安定した変換実験を開始する際には、健全な分岐をソース材料として選択することが重要です。パラスポニアT0トランスジェニックラインは、インビトロで伝播される。したがって、パラスポニアトランスジェニックラインを徹底的に遺伝子型化することが不可欠である。実験モデルとしてのパラスポニアの確立により、2つの遠くに関連する結節系、パラスポニアとマツムの比較分析が可能になります。これは、根茎共生の根底にある遺伝的ネットワークの保全を特定することができる。パラスポニアはまた、木の形成、バイセクシュアル花の開発、またはカナバ科特異的二次代謝産物の生合成のような他の研究トピックを研究するための貴重なモデルであり得る。
