この拡散画像化プロトコルを用いて、解剖学的MRIでは見えない軽度の外傷性脳損傷を有するラットの海馬の微細構造変化を調査することが可能である。この技術は、CTまたは解剖学的MRIでは検出できない軽度および拡散性外傷に続く脳内の変化を検出することができる。この技術は、客観的かつ定量的な方法で軽度の外傷性脳損傷を持続した後、回復プロセスを監視することが容易になります。この拡散イメージングおよび分析技術は、前臨床試験だけでなくヒトにおいても、認知症や多発性硬化症などの脳に影響を及ぼす他の障害にも適用できる。このプロトコルでは、拡散スキャンと補正ステップの品質が高いということが重要であるため、経験豊富な技術者やアナリストからの指導が推奨されます。生後12週の雌のウィスターHラットでつまみつまみへの応答の欠如を確認した後、動物を37°Cの加熱パッドに置き、カテーテルを側面尾静脈に挿入します。2%リドカインの100マイクロリットルを剃り、分解した頭皮に局所的に注入し、中線切開を行い、頭蓋骨を露出させます。小さなはさみを使用して余分な膜を取り除き、骨膜が存在しなくなるまで頭蓋骨を横切って綿棒をこすり、ティッシュ接着剤の滴を使用して直径10ミリメートル、厚さ3ミリメートル、金属ディスクを約1/3の前に、ブレグマの後ろに2/3を取り付けます。外傷性脳損傷誘導のために、ラットを特定のばね定数のフォームマットレスでカスタムメイドのベッドに置き、ラットをできるだけ水平にヘルメットを持つ450グラムの真鍮の重量を持つ透明なプラスチックチューブの下に直接置きます。重量を1メートルまで引き上げろ。2番目の実験者が存在すると、体重を解放し、2番目の実験者は、2番目の損傷を防ぐために、衝撃の直後にラットをチューブから離して移動させます。頭蓋骨からヘルメットをそっと引っ張り、ガーゼを使って出血を止めます。縫合糸で皮膚を閉じ、切開部に局所鎮痛ゲルを塗布します。ラットをCTスキャナーのベッドに置き、頭蓋骨骨折を排除するために汎用の低用量CTスキャンを行い、動物をケージに戻す前に完全な再発まで監視して37°Cの加熱パッドのきれいなケージにラットを置きます。外傷誘導の前および1日後に、実験動物におけるつまみつまみへの応答の欠如を確認し、MRスキャナーベッドの上に動物を頭の最初に置き、起こりやすい位置に置く。直交容ボリュームコイルを頭の上にスライドさせ、スキャナーベッドをスキャナーボアに進めます。正しい位置を確認するには、デフォルトの 3 平面スカウトスキャンを取得します。スキャンが終了したら、画像ディスプレイにスキャンをロードし、頭がまっすぐに横たわっていて、脳が磁石とコイルの中心に配置されていることを確認します。フィールド、ビュー、およびマトリックスサイズを除き、デフォルト設定を使用して T2 加重画像を取得します。ジオメトリ エディタを開き、脳の球根や小脳などの正しい位置にスライス パッケージを配置し、B_diffusion フォルダから 3 つの新しいエコー平面、拡散重み付け、スピン エコー シーケンスをスキャン制御プロトコルにロードします。デフォルト設定を使用して拡散ウェイト画像を取得し、[スキャンの編集]タブを開きます。スライスの方向を軸に、スライスの数を 25 に設定して、スライスの厚さ 500 マイクロメートルと内側のスライス距離を 600 マイクロメートルにし、読み出し方向を左から右に修正します。[ジオメトリ]タブで、幾何学パラメータを調整し、視野とマトリックスのサイズを105 x 105に調整して、333 x 333 マイクロメートルの解像度を確保します。3 つの拡散シェルのそれぞれについて[研究]タブの[拡散]タブをクリックし、拡散方向の数を第 1 シェルでは 32、2 番目のシェルの場合は 46、3 番目のシェルの場合は 64 に調整します。B0 イメージの数を、最初のシェルでは 5、2 番目のシェルでは 5、3 番目のシェルでは 7 に変更し、カスタム グラデーション方向ファイルでグラデーションの方向を調整します。第 1 シェルの場合は 1,500 秒/ミリメートル、3 番目のシェルでは 1,000 秒/ミリメートル、3 番目のシェルでは 2,000 秒/ミリメートルに 2,000 秒の方向の B 値を調整し、次にジオメトリ エディタを開き、大脳との間に視野を置いてアーティファクトとスキャン時間を短縮します。スキャンプロトコルの完了時に、スキャナーベッドから37°Cの加熱パッドを備えたきれいなケージに動物を転送し、完全な債務まで監視します。拡散MRI画像処理の場合は、MRtrix3に画像をロードし、ソフトウェアプログラムの拡散加重画像にノイズ補正とギブリング補正を実行します。T2 イメージ内の補正された拡散重み付けされたイメージを、示されている NIFTI 形式に変換します。エコー平面イメージング、モーション、渦電流歪みの補正を実行するには、ExploreDTIのプラグインメニューで、被験者モーションのEC/EPI歪みの補正を選択し、前処理された拡散データファイルを選択します。各ラットの拡散テンソルイメージングメトリックを計算するには、[プラグイン]と[ものをNIFTIにエクスポート]をクリックし、拡散テンソルイメージングモデルのパラメトリックマップを選択して、尖度および白質トラテグリティモデル用のパラメトリックマップをエクスポートします。各ラットの海馬のマスク ファイルを作成するには、MRtrix ビューアでラットの分数異方性画像を読み込み、プラスボタンをクリックして新しい関心領域を作成します。ラットの海馬の拡散メトリックを抽出するには、作成したマスクファイルをAMIDEソフトウェアにインポートし、ラットのパラメトリックマップとマスク画像を開きます。マスクファイルの対象領域を AMIDE に追加するには、マスクファイルイメージを選択し、「編集」「対象領域を追加」「3D 等角」をクリックして、対象領域に意味のある名前を付けます。マスクイメージに表示されている対象領域をクリックし、このボリュームに1の値を持つボクセルのみが含まれていることを確認します。海馬の拡散メトリックの平均値を計算するには、[ツール] と [対象地域統計の計算] をクリックし、含める画像と対象地域を指定します。[実行] をクリックすると、さらに統計分析に使用できる計算値を含むポップアップ ウィンドウが表示されます。この代表的な実験では、CTイメージングによって評価された頭蓋骨骨折の証拠はなく、T2画像は外傷の翌日に消耗品サイトで異常を示さなかった。T2画像と拡散データセットとの間の非剛性共登記ステップの品質を調べるために、T2画像のオーバーレイを色エンコードされた分数異方性マップに追加した。分数異方性、平均拡散率、軸拡散率、および放射状拡散度パラメトリックマップのパラメトリックマップを計算できます。対象領域内では、軸方向、平均値、放射状尖度値の平均値、軸水分率、軸軸および半径軸外拡散率、および白物位トラクトの耐音性の計算も行うことができる。本代表的な実験では、拡散テンソルイメージングメトリックの分析により、軽度外傷性脳損傷群の影響に続く分数異方性値の有意な増加および拡散率値の減少が明らかになった。また、拡散尖度測定基準は、軸方向または平均尖度の変化は認められなかった一方で、衝撃後の放射状尖度の有意な減少を示した。白質管完全性モデルを用いて、放射状軸外拡散率は有意な減少を示し、トルツオシティは衝撃の翌日に軽度の外傷性脳損傷群の有意な増加を示した。画像解析では、MRtrix のデータ形式が適切に変換されて ExploreDTI にインポートされ、各修正手順が正しく実行されたかどうかを確認することが重要です。ROI ベースの分析の代わりに、ボクセルによるボクセル分析を適用して、脳全体の変化を調査できます。この技術は、神経イメージング分野で非常に貴重であり、同様に他の脳障害、例えば、認知症および多発性硬化症に適用することができる。
