1. 试剂制备

</ol>2. 第1天：早期索米特阶段胚胎的解剖

<ol><li>颅神经板的解剖仅限于胚胎在8.5天后（dpc）。在5~8个胚接阶段选择胚胎。将子宫分解成PBS，切开中体以分离每个胚胎（图1A）。子宫的肌肉壁收缩和二元组织将变得可见（图1B）。 注：在冰冷的PBS中保持子宫内胚胎，同时进行一次一个胚胎的解剖。用玻璃巴斯德移液器将胚胎移入新鲜无菌PBS，以提高可见度并减少污染。</li>
	<li>在肌肉层和二元组织之间滑动钳子，用第二对钳子去除肌肉层（图1C）。</li>

	<li>将头部折叠在心脏上方，使用钳子和/或睫毛工具刮去底层的中皮，以获得干净的神经板 （NP）（图 1H）。 注： NP 可以保持整体或向下分离前方轴，以便每侧可以单独镀。神经板边界可以进一步修剪远离神经板，以尽量减少神经元对外植的贡献。</li>

</ol>3. 第2天：鼠颅神经峰细胞的活细胞成像
注：成像应在24小时后进行切除，以最佳图像和量化神经峰细胞迁移。在活细胞成像之前，不需要刷新NC感应介质。需要使用倒置显微镜，带有电动舞台和内部环境室。使用适合成像的多井组织培养皿（材料表）。

</ol>4. 成像分析：神经峰细胞迁移的定量
注：为了更好地定义迁移小鼠颅神经峰细胞所表现出的细胞行为，我们分析了一系列可量化的迁移参数，特别是迁移能力和细胞形状动力学。（1）迁移（累积距离）是像元 （μm） 获取的总路径长度;（2）迁移（欧氏距离）是单元（μm）初始位置和最终位置之间的直线距离;（3）迁移（细胞速度）是按单位时间（μm/min）的细胞行驶距离;（4）细胞形状（细胞面积）是细胞覆盖的总表面。根据成像显微镜将像素设置为微米刻度。（A =px x Npx，其中 Apx = 像素面积和 Npx = 像素数。单位： μm2;（5）细胞形状（细胞圆度）是细胞形状与完美圆的偏差，由 A = 面积和 P = 周长的圆度值 1.0 （4°（A/P2）表示。

为了达到一致的结果，我们发现在选择胚胎时特别注意分期。毫不奇怪，在颅控NC发育中，索米特的数量与不同的阶段相关。因此，在获取任何实验数据之前，胚胎解剖学的知识是非常重要的。然后，这种方法可以适应分离神经峰细胞的离散群体，这取决于生物学问题和目标细胞。
一旦选择和解剖胚胎，中皮细胞就很容易区分，应该去除，以便更好的可视化和减少污染。对于长期培养，神经板组织可以在24小时电镀时被去除，以防止神经组织污染。进一步改进可能是使用荧光系标记（例如，使用Wnt1：：：cre或Sox10：：：creERT驱动器与荧光报告器24、 25） 来区分神经峰细胞和其他组织如图2C所示。

dissection, culture and analysis of primary cranial neural crest cells from mouse for the study of neural crest cell delamination and migration

24小时也很明显，植物外培养物预迁移和完全迁移NC细胞的特征同心环不依赖或受矩阵选择控制（图3）。在ECM基水凝胶和纤维素上镀层的外植培养物形成了可比的外植结构，包括三个细胞群，NP，pNC和mNC（图3A，C）。神经峰细胞形态在ECM水凝胶和纤维素上镀层细胞形态也比较（图3B，D）。然而，在纤维素上镀的外植植物在细胞前缘产生具有更突出的拉梅利波迪亚的细胞，在迁移方向上似乎更加极化（图3B，D）。
一旦迁移神经峰细胞的种群明显，活细胞成像就可以完成。延时显微镜设置为 10 倍放大倍率（18 小时，1 帧/5 分钟），以便后续分析 NC 细胞迁移（图 4A）。ImageJ 手动跟踪插件在延时影片的所有帧上生成各个单元的 XY 坐标（图4B）。可以使用迁移软件处理这些坐标。该软件支持可视化单个细胞轨道随时间（图4B），并可用于量化累积和欧氏距离，以及细胞速度。
