小型RNA（sRNA）参与控制生物过程的多样性1。真核子病监管sRNA的大小一般在20至30NT之间;三种主要类型是微RNA（miRNA）、piwi相互作用RNA（piRNA）和小型干扰RNA（siRNA）。异常miRNA表达水平已牵连于各种疾病2。这强调了miRNA在健康和疾病中的重要性，以及准确、定量的研究工具来检测一般sRNA的要求。
下一代测序（NGS）是研究sRNA的一种广泛应用的方法。与其他方法（如定量PCR或微阵列技术（qPCR）相比，NGS的主要优点是，它不需要先验的sRNA序列知识，因此可用于发现新的RNA，此外，它遭受的较少背景信号和饱和度效应。此外，它可以检测单个核苷酸差异，并且具有比微阵列更高的吞吐量。然而，NGS也有一些缺点;测序运行的成本仍然相对较高，将样本转换为库进行测序所需的多步骤过程可能会引入偏差。在典型的sRNA库制备过程中，在没有ATP的情况下，使用截断版本的RNA连带酶2（RNL2）和预分3'适配器（图1），首先将3'适配器与sRNA（通常是从总RNA中凝胶纯化）连结。这提高了sRNA适配器结扎的效率，并减少了侧反应的形成，如sRNA循环或串联。随后，5' 适配器由RNA连带1（RNL1）连接，然后是逆转录（RT）和PCR扩增。所有这些步骤都可能引入偏差3，4。因此，读取数可能无法反映导致人工的、依赖于方法的表达模式的实际 sRNA 表达水平。特定 sRNA 在库中可能过多或不足，且代表性严重不足的 sRNA 可能逃脱检测。情况特别复杂，植物miRNA，昆虫和植物的siRNA，以及昆虫、线虫和哺乳动物中的piRNA，其中3'端核苷酸具有2'-O-甲基（2'-OMe）修饰1。这种修改强烈抑制了3'适配器结扎5，使得为这些类型的RNA准备库准备是一项艰巨的任务。
以前的工作表明，适配器结扎引入严重的偏差，由于RNA序列/结构效应6，7，8，9，10，11。适配器结扎的下游步骤（如逆转录和 PCR）不会显著导致偏差6、11、12。结扎偏差可能是由于具有给定序列的适配器分子与反应混合物中的 sRNA 分子相互作用，形成共褶，这可能导致连接的有利或不利配置（图 2）。来自Sorefan等人7的数据表明，RNL1更喜欢单一搁浅的语境，而RNL2则更喜欢双绞线进行结扎。适配器/sRNA 共折叠结构由特定的适配器和 sRNA 序列决定这一事实解释了为什么特定 sRNA 在给定适配器集中过度表示或表示不足。还必须注意，在要比较的一系列 sRNA 库中，应使用相同的适配器序列。事实上，以前已经观察到，通过引入不同的条形码序列来改变适配器会改变测序库中的miRNA配置文件9，13。
在结扎结附近随机化适配器序列可能会减少这些偏差。Sorefan 和同事7使用四肢有 4 个随机核苷酸的适配器，指定"高清晰度"（HD） 适配器，并表明使用这些适配器可产生更好地反映真实 sRNA 表达水平的库。最近的工作证实了这些观察结果，并表明随机区域不需要与结扎交汇点11相邻。这种新型适配器被命名为"MidRand"适配器。总之，这些结果表明，改进的适配器设计可以减少偏差。
通过优化反应条件，可以抑制偏置，而不是修改适配器。聚乙烯乙二醇（PEG），一种大分子挤出剂，已知能提高结扎效率14，已被证明能显著减少偏置15，16。根据这些结果，市场上出现了几个"低偏置"套件。其中包括在连接反应中使用 PEG 的套件，包括与经典适配器或 HD 适配器结合使用。其他套件完全避免结扎，并使用3'聚化和模板切换3'和5'适配器添加，分别12。在另一种策略中，3' 适配器连接后跟一个循环步骤，因此省略了 5' 适配器连接17。
在以前的研究中，我们搜索了一个sRNA库准备协议，其偏差程度尽可能低，并且对2'-OMe RNA12的最佳检测值。我们测试了上述一些"低偏置"试剂盒，与标准协议 （TS） 相比，对 2'-OMe RNA 的检测效果更好。然而，令人惊讶的是，在修改（使用随机适配器，PEG在结扎反应和去除多余的3'适配器通过纯化）后，后者优于其他协议检测2'-OMeRNA。在这里，我们详细介绍了基于 TS 协议的协议"TS5"，该协议对 2'-OMe RNA 进行了全面检测最好）。该协议可以使用TS试剂盒中的试剂和"Nf"试剂盒中的一种试剂进行遵循，或者为了节省资金，使用自制的试剂，性能相同。我们还提供了从总RNA中纯化sRNA和制备预化3'适配器的详细方案。
