Hematopoietische stamcellen (hscs) zijn zelfvernieuwend, multi-potent, en kunnen aanleiding geven tot alle cellen van het bloed1,2. Cellulaire metabolisme is een belangrijke regulator van HSC-onderhoud, samen met transcriptionele factoren, intrinsieke signalen en de micro-omgeving3,4,5. De juiste controle van de mitochondriale functie en kwaliteit is daarom van cruciaal belang voor HSC-onderhoud6,7.
Mitochondriale membraanpotentiaal (ΔΨm) is een belangrijke parameter in de beoordeling van mitochondriën als het direct weerspiegelt hun functionaliteit, die voortvloeit uit het evenwicht van proton pomp activiteit in de elektronentransport keten en de Proton stroom door F 1/FO ATP-synthase. Deze zijn beide nodig (afhankelijk van de genexpressie en beschikbaarheid van het substraat) voor de zuurstof afhankelijke fosforylering van ADP naar ATP8,9. Door gebruik te maken van de electronegativiteit van het mitochondriale compartiment zijn verschillende potentiometrische kleurstoffen ontwikkeld om ΔΨm te meten. Een van hen is tetramethylrhodamine methyl ester perchloraat (TMRM), die uitgebreid is gebruikt voor het meten van ΔΨm doorstroming cytometrie in een verscheidenheid van cellen10, met inbegrip van hematopoietische stam en voorlopercellen11.
Mitochondriale kleurstoffen moeten worden gebruikt met enige voorzichtigheid in HSCs, echter, omdat de hoge activiteit van de xenobiotische Efflux pompen van deze cellen kan resulteren in kleurstof extrusie12. Inderdaad, de extrusie van mitochondriale kleurstoffen zoals Rhodamine 123 heeft onderzoekers toegestaan HSCs13 te isoleren of te identificeren HSC "side populaties" door het benutten van de differentiële extrusie van de kleurstoffen Hoechst blauw en Hoechst rood14, 15. ook is aangetoond dat Fumitremorgin C, een specifieke blokkade van de ATP-bindende cassette subgroep G member 2 (ABCG2), geen invloed heeft op het kleurings patroon van MitoTracker in HSPCs16. Na de publicatie van deze resultaten, meerdere studies werden uitgevoerd met behulp van mitochondriale kleurstoffen in de afwezigheid van xenobiotische Efflux pomp remmers, wat leidt tot de wijdverbreide indruk dat HSCs slechts een klein aantal mitochondriën met lage ΔΨm hebben16 , 17 , 18.
Onlangs werd echter aangetoond dat verapamil, een breed spectrum remmer van effluxpompen, het kleurings patroon van de mitochondriale kleurstof MitoTracker Green19aanzienlijk wijzigt. Dit verschil is waarschijnlijk te wijten aan het feit dat Fumitremorgin C zeer selectief is voor Abcg2, terwijl HSCs ook andere transporters uitdrukken, zoals Abcb1a (die slechts zwak gevoelig is voor Fumitremorgin C)19. We hebben ook gemeld dat andere mitochondriale kleurstoffen, zoals TMRM, nonyl acridine Orange en Mitotracker Orange (MTO) dezelfde patronen vertonen als Mitotracker Green. Wat nog belangrijker is, we hebben geconstateerd dat de Flow cytometrische patronen van HSPCs weerspiegelen hun ΔΨm in aanvulling op mitochondriale massa11.
De inname van TMRM kleurstof is strikt afhankelijk van de negatieve lading van de mitochondriën, maar de resulterende accumulatie van kleurstof is in constante balans tussen de inname en de klaring door Efflux pompen20. Het verschil in xenobiotische effluxpomp expressie tussen HSCs en volwassen celpopulaties beïnvloedt dit evenwicht en kan leiden tot bevooroordeelde resultaten. Het gebruik van speciale remmers zoals verapamil moet worden overwogen bij de analyse van ΔΨm door potentiometrische kleurstoffen. Hier beschrijven we een gewijzigd protocol voor nauwkeurige ΔΨm meting door TMRM gebaseerde flow cytometrie die corrigeert voor xenobiotische Transporter activiteit door het gebruik van speciale remmers.

Als cellulaire metabolisme is een belangrijke regulator van hematopoietische stamcel (HSC) zelf vernieuwing, de verschillende rollen gespeeld door de mitochondriën in hematopoietische homeostase zijn uitgebreid bestudeerd door HSC onderzoekers. Mitochondriale activiteit niveaus worden weerspiegeld in hun membraan mogelijkheden (ΔΨm), die kan worden gemeten door cel-perbetekende kationische kleurstoffen zoals tmrm (tetramethylrhodamine, methyl ester). Het vermogen van effluxpompen om deze kleurstoffen uit cellen te extrumaken, kan echter hun bruikbaarheid beperken. De resulterende meet bias is met name van cruciaal belang bij de beoordeling van HSCs, omdat xenobiotische transporters hogere niveaus van expressie en activiteit vertonen in HSCs dan in gedifferentieerde cellen. Hier beschrijven we een protocol met behulp van verapamil, een effluxpomp remmer, om nauwkeurig ΔΨm te meten over meerdere beenmerg populaties. De resulterende remming van de activiteit van de pomp is aangetoond dat de intensiteit van de TMRM in hematopoietische stam en voorlopercellen (Hspc's) te verhogen, terwijl het verlaten van het relatief onveranderd in rijpe fracties. Dit benadrukt de nauwe aandacht voor kleurstof-Efflux activiteit die nodig is wanneer ΔΨm-afhankelijke kleurstoffen worden gebruikt, en zoals geschreven en gevisualiseerd, dit protocol kan worden gebruikt om nauwkeurig te vergelijken ofwel verschillende populaties binnen het beenmerg, of dezelfde populatie over verschillende experimentele modellen.

Het hierboven beschreven protocol maakt het eenvoudig isoleren van BM-Mnc's van een muismodel mogelijk. Figuur 1 vat de belangrijkste stappen van het protocol samen: botisolatie, blozen uit het beenmerg, rode bloedcel lysis, en antilichaam kleuring gevolgd door tmrm kleuring om Mitochondriale membraanpotentiaal in een specifieke hematopoietische populatie te meten .

Improving the Accuracy of Flow Cytometric Assessment of Mitochondrial Membrane Potential in Hematopoietic Stem and Progenitor Cells Through the Inhibition of Efflux Pumps

Xenobiotic Efflux pompen zijn zeer actief in hematopoietische stam en voorlopercellen (Hspc's) en veroorzaken extrusie van TMRM, een mitochondriale membraan potentiële fluorescerende kleurstof. Hier presenteren we een protocol om het mitochondriale membraan potentieel in Hspc's met TMRM nauwkeurig te meten in aanwezigheid van verapamil, een effluxpomp remmer.

Mitochondriale functie is van cruciaal belang voor hematopoietische stamcel zelfvernieuwing. Mitochondriale activiteit is een reflector van het binnenste mitochondriale membraanpotentieel, dat kan worden gemeten door TMRM, een cel fluorescerende, cel-permeant kationische kleurstof. Xenobiotische efflux pompen zijn zeer actief in HSC, en veroorzaken TMRM extrusie.Het belangrijkste voordeel van dit protocol, is de correctie van dit effect, met behulp van Verapamil, een remmer van de efflux pompen. Begin met het isoleren van beenmerg van muisvrouwjes. Gebruik een tang en een scherpe schaar om de twee achterste poten te snijden.Maak vervolgens een kleine knip in de ventrale huid van de muis, en strek het uit. Haal het dijbeen en het scheenbeen eruit, terwijl je ervoor zorgt dat je de hoofden van het dijbeen niet los maakt en plaats de botten in een zes-put plaat, gevuld met 1,5 milliliter kleuringsbuffer. Verwijder de spieren van de botten, en snijd de uiteinden van de botten, waardoor beenmerg te verlaten.Plaats de gereinigde botten in nieuwe putten, met 1,5 milliliter kleuring buffer. Spoel vervolgens het beenmerg weg met een spuit van drie milliliter en een naald van 25 meter, totdat het bot wit wordt. Verzamel alle cellen in een buis van 1,5 milliliter en vercentrifugeer ze gedurende vijf minuten op 180 keer G en gooi de supernatant weg.Voorzichtig resuspend de pellet in 300 microliters van ijskoude ACK lysing buffer, en zet de cellen op ijs voor een minuut. Schakel vervolgens onmiddellijk lyse uit door een milliliter kleuringsbuffer toe te voegen. Centrifugeer de cellen gedurende vijf minuten bij 180 keer G.Resuspend de celpellet, die wit zou moeten lijken, in een milliliter kleuringsbuffer.Filter de cellen met behulp van een cel zeef dop, met een 35 micrometer nylon mesh, om mononucleaire cellen te verkrijgen. Begin met het bereiden van de stamcocktail, en fluorofofre geconjugeerde antilichamen, volgens manuscriptrichtingen. Houd de antilichamen op ijs en beschermd tegen licht.Vervolgens centrifuge het monster gedurende vijf minuten op 180 keer G en gooi het supernatant weg. Voeg 400 microliter van de lijncocktail en vier microliter cd135 biotinylhoudende antilichamen toe aan de celpellet en draaikolk het mengsel. Dan uitbroeden op ijs voor 30 minuten.Na de incubatie, was het monster door het toevoegen van drie milliliter vlekken buffer, spinnen het naar beneden voor vijf minuten op 180 keer G, en het weggooien van de supernatant. Voeg vervolgens 400 microliter antilichaamoplossing toe en draaikolk het mengsel. Broed het nog 30 minuten op ijs en herhaal de was met de kleurbuffer.Om de TMRM-kleuroplossing voor te bereiden, voegt u 2,2 microliter TMRM-voorraadoplossing en 1,1 microliter Verapamil toe aan 1,1 milliliter serumvrije hematopoietische kweekmedium, met trombopoietine en stamcelfactor. Resuspend het beenmerg in een milliliter van de TMRM kleuroplossing, vortex snel, en incubeer voor een uur op 37 graden Celsius. Filter het monster met behulp van een cel zeef dop, met een 35 micrometer nylon mesh, om te voorkomen dat verstopping van de stroom cytometer.Voeg vervolgens een microliter van DAPI toe en ga verder met stroomcytometrie. Voer het beenmergmonster uit en verwerf minstens een miljoen evenementen. Geef vervolgens de levende beenmergmonnucleaire cellen weer in een plot voor pacific blue, om CD135 Lin-negatieve en Lin-positieve breuken te identificeren.Plot de Lin-negatieve fractie voor APC/Cy7 versus PE/Cy7 om de multipotente voorloper of MPP-fractie te identificeren. Zet vervolgens de MPP-fractie voor APC versus PerCP/Cy5.5 in kaart om de hematopoietische stamcel of HSC-fractie te identificeren. Geef de HSC-fractie voor FITC of CD34 weer om deze te verdelen in cd34-negatieve HSC- en CD34-positieve HSC-fracties.Ten slotte verwerven van de TMRM intensiteit in elke populatie. Om ervoor te zorgen dat TMRM-vlekken correct werkten, voegt u een microliter FCCP toe aan het monster, voor een laatste concentratie van één millimolar en ontcuut het gedurende vijf minuten bij 37 graden Celsius. Dan verwerven een miljoen evenementen.Na fccp toevoeging, TMRM intensiteit moet drastisch worden verhoogd. Dit protocol is met succes gebruikt om mitochondriaal membraan potentieel te kwantificeren in verschillende celpopulaties, met TMRM kleurstof. Aangetoond is dat de aanwezigheid van PBS en FBS in het kweekmedium de signaalintensiteit van TMRM beïnvloedt.Het is dus belangrijk om TMRM-kleuring uit te voeren in serumvrij expansiemedium. Hematopoietische stamcellen drukken hoge activiteitsniveaus uit van xenobiotische effluxpompen die TMRM-kleurstof extruderen. TMRM-profielen variëren dus ook in de aanwezigheid van Verapamil of Cyclosporin H, die de pompen blokkeren.Om de nauwkeurigheid van TMRM-kleuring te verifiëren, kan FCCP de mitochondriën depolariseren en de TMRM-intensiteit verminderen. Dit protocol kan eenvoudig worden aangepast voor het bestuderen van HSC met een andere mitochondriale membraan potentiële kleurstof, met inbegrip van TMRE of JC-1. Evenals andere mitochondriale kleurstoffen zoals MitoTracker of MitoSOX.De kritische kwesties die zijn geërfd door de verschillende activiteit van effluxpompen onder de beenmergpopulatie, werden pas onlangs opgemerkt. Dus dit protocol is bedoeld om de discrepantie tussen eerdere bevindingen te verminderen.
