ミトコンドリア機能は造血幹細胞の自己再生に重要です。ミトコンドリア活性は、ミトコンドリア膜電位の反射体であり、細胞蛍光性、細胞透過性カチオン色素であるTMRMによって測定することができる。異種性流出ポンプはHSCにおいて高活性であり、TMRM押出を引き起こす。このプロトコルの主な利点は、この効果の補正、エフラックスポンプの阻害剤であるベラパミルを利用することである。マウス大腿骨から骨髄を分離することから始めます。鉗子と鋭いはさみを使用して、2つの後足を切ります。その後、マウスの腹側の皮膚に小さな切り取りを行い、それを伸ばします。大腿骨と脛骨を抽出し、大腿骨の頭部を外さないで、骨を6ウェルプレートに入れ、1.5ミリリットルの染色バッファーで満たします。骨から筋肉を取り除き、骨の端を切り取り、骨髄を出ることができます。洗浄された骨を1.5ミリリットルの染色バッファーで、新しい井戸に入れます。次に、骨が白くなるまで、3ミリリットルの注射器と25ゲージの針で骨髄を洗い流す。1.5ミリリットルチューブ内のすべての細胞を収集し、180倍Gで5分間遠心分離し、上清を捨てます。慎重に氷冷ACKライジングバッファーの300マイクロリットルにペレットを再中断し、1分間氷の上に細胞を置きます。その後、すぐに1ミリリットルの染色バッファーを添加することによって、ライシスを失活させる。180倍のG.180倍で5分間細胞を遠心分離し、白く見えるはずの細胞ペレットを1ミリリットルの染色バッファーに再懸濁させる。細胞ストレーナーキャップを用いて細胞をフィルターし、35マイクロメートルのナイロンメッシュで、単核細胞を得る。原稿の指示に従って、系統カクテル、およびフルオロフォア共役抗体を準備することから始めます。氷の上に抗体を保持し、光から保護します。次いで、試料をGの180倍で5分間遠心し、上清を捨てる。400マイクロリットルのリネージュカクテルを加え、4マイクロリットルのCD135ビオチン化抗体を細胞ペレットに加え、混合物をボルテックスします。その後、氷の上で30分間インキュベートします。インキュベーション後、染色バッファーを3ミリリットル加えて、Gの180倍で5分間回転させて上清を捨ててサンプルを洗います。その後、400マイクロリットルの抗体溶液を加え、混合物をボルテックスする。さらに30分間氷の上でインキュベートし、染色バッファーで洗浄を繰り返します。TMRM染色液を調製するには、2.2マイクロリットルのTMRMストック溶液と1.1マイクロリットルのベラパミルを、血栓ポエチンおよび幹細胞因子を用いて1.1ミリリットルの無血清造血培地に添加する。TMRM染色液の1ミリリットルで骨髄を再懸濁し、素早く渦を、摂氏37度で1時間インキュベートする。35マイクロメートルのナイロンメッシュを使用して、フローサイトメーターを詰まらせないように、セルストレーナーキャップを使用してサンプルをフィルター処理します。次に、DAPIを1マイクロリットル加え、フローサイトメトリーを進めます。骨髄サンプルを実行し、少なくとも100万個のイベントを取得します。次に、生きた骨髄単核細胞を太平洋青色のプロットで表示し、CD135 Lin陰極およびLin-陽性画分を同定する。APC/Cy7 対 PE/Cy7 の Lin-負の分率をプロットして、多能性前駆子または MPP 分率を識別します。次に、造血幹細胞またはHSC画分を識別するために、APC対PerCP/Cy5.5のMPP分数をプロットします。FITC または CD34 の HSC 分数を表示して、CD34 マイナス HSC および CD34 正の HSC 分数に分割します。最後に、各集団のTMRM強度を取得します。TMRM染色が正しく機能したことを確認するには、1ミリモルの最終濃度のためにサンプルに1マイクロリットルのFCCPを加え、5分間摂氏37度でインキュベートします。その後、100万のイベントを取得します。FCCPの追加後、TMRM強度を大幅に増加する必要があります。このプロトコルは、TMRM色素を用いて、様々な細胞集団におけるミトコンドリア膜電位の定量化に成功しています。培養培地中のPBSおよびFBSの存在がTMRMシグナル強度に影響を及ぼすことが実証された。そのため、無血清膨張培地でTMRM染色を行うことが重要です。造血幹細胞は、TMRM色素を押し出す異種性流出ポンプの高い活性レベルを発現する。したがって、TMRMプロファイルは、ポンプをブロックするベラパミルまたはシクロスポリンHの存在においても異なります。TMRM染色の精度を検証するために、FCCPはミトコンドリアを脱分極し、TMRM強度を低下させることができる。このプロトコルは、TMREまたはJC-1を含む異なるミトコンドリア膜電位色素を用いてHSCを研究するために容易に適応することができる。ミトトラッカーやミトソックスなどの他のミトコンドリア色素と同様に。骨髄集団の流出ポンプの異なる活性によって受け継がれてきた重大な問題は、つい最近指摘された。したがって、このプロトコルは、以前の知見間の不一致を減らすことを目的としています。
