遺伝子機能と疾患の研究は、人間の集団の外来性によって妨げられている。患者固有および制御iPS細胞株の異なる遺伝的背景は、所定の機能的アッセイに見られる分化効率および表現型に影響を及ぼす可能性がある。唯一の違いが関心の特定の突然変異である遺伝的に同一または同一の線の使用は、これらの問題を克服するために重要である。多くのヒト疾患は、CRISPR-Cas9システムでは生成が困難なヘテロ接合性突然変異によって引き起こされます。これは、非常に高い頻度で起こり得る非標的性アリールにおけるインデル突然変異の生成によるものです。私たちの技術は、1つだけが関心の突然変異を抱えている2つの修復テンプレートを使用して、この問題を克服します。この技術を試みる研究者は、ヒト多能性幹細胞培養に熟練すべきである。コロニーピッキングのビデオデモンストレーションは、正しいサイズと形態だけでなく、ピッキングやスクリーニングに使用される技術を示すために役立ちます。この手順をデモンストレーションするレオ・カルデナスです。まず、照射されたMEF上のヒトESCを6ウェルプレートにプレートします。トランスフェクションマスターミックスを準備します。ピペットで混ぜ、室温で15分間インキュベートします。細胞が70〜80%の合流度に達したら、トランスフェクションマスターミックスを細胞と共に各ウェルにドロップして、摂氏37度で48時間インキュベートします。メディアを 24 時間ごとに変更します。細胞を収穫するには、まず、トリプルEで細胞を室温で3分間インキュベートし、MEFを酵素的に除去する。10マイクロモルロック阻害剤とHESC培地の0.5ミリリットルを追加します。ペレット細胞は3分間300回gで、10マイクロモルROCK阻害剤を有するヒトESC培地の0.5ミリリットルで再懸濁した。35ミクロンの細胞ストレーナーキャップを通して5ミリリットルチューブに細胞懸濁液をろ過します。蛍光活性化細胞選別を使用して、生細胞にゲートを付け、緑色蛍光タンパク質陽性細胞を選別します。次に、4つの選別された細胞に最大1.5倍の10を直接10センチメートルの皿に移し、1〜3基膜マトリックスを塗り、ROCK阻害剤を含むMEFおよびヒトESC培地を照射した。ROCK阻害剤を使用せずにヒトESC培地を使用して毎日培地を変更します。10〜15日後、コロニーは直径約1〜2ミリメートルである。顕微鏡下で、P200ピペットを使用して1つのクローンを慎重に削り取り、ピペットに細胞を引き込みます。コロニーで作成された培地中で3~4回軽くピペットを入れて、96ウェルプレートのウェルに細胞を分散させます。次に、各ガイドRNAに対して20個のコロニーを選び、スクリーニングのためにPCRストリップチューブに分配します。ペレット細胞は、10,000回Gで5分間遠心分離して細胞をペレット化する。現在、タンパク質処理Kバッファーの20マイクロリットルで細胞ペレットをインキュベートし、DNAと渦を激しく分離します。5分間Gの10,000倍で遠心分離機。次に、編集したDNAのスクリーニングPCRを行う。目的の領域を増幅するように設計されたフォワードとリバースプライマーを含むマスターミックスのチューブ20マイクロリットルだけでなく、プロテアーゼKダイジェストの5マイクロリットルを追加します。スクリーニングPCRを行う際に、コントロールiPSCラインから単離したゲノムDNAを使用して、クリーンアンプリコンを確認します。70~90ボルトで1時間経ったPCR産物のサイズ変化を、2.5%の重量で2.5%のアガロースゲルで評価します。どんなサイズの違いも切断を示す。一本鎖オリゴDNAとCRISPR-Cas9プラスミドをトランスフェクトするには、照射されたMEFの6ウェル皿にターゲット細胞株をプレートし、一晩のインキュベーション後に70〜80%のコンフルエンスに達する。記載に従ってトランスフェクション反応を設定する。反応をピペットで混ぜ、室温で15分間インキュベートします。次に、トランスフェクション反応混合物を細胞にドロップワイズ添加する。48 時間後、セルの並べ替えに使用するセルを以前のように準備します。単一細胞をめっきしてから約10日後、200マイクロリットルのピペットを使用してコロニーをPCRストリップチューブに選び、ゼラチンとMEFで事前にコーティングされた12ウェルプレートの井戸に入ります。細胞の100マイクロリットルを24または48ウェルプレートの各ウェルに移し、以前にゼラチンでコーティングし、ROCK阻害剤を用いたヒトESC培地中のMEFを照射した。残りの100マイクロリットルをDNA分離に使用します。一本鎖オリゴDNAの正常な統合を確認するには、各コロニーから分離した5マイクロリットルのDNAを採取し、以前に設計したスクリーニングプライマーを使用してPCRを行います。PCR産物を精製し、一本鎖オリゴDNAに作成されたユニークな酵素サイトを使用して、制限酵素消化の40マイクロリットルを調製します。ピペットで反応を混ぜ、メーカーの推奨温度で1〜3時間インキュベートします。次に、1.5%重量の負荷色素を含む消化PCR産物を、80~100ボルトで40分間、80~100ボルトで表した量のエチジウム臭化物を可視化します。一本鎖オリゴDNAの積分が成功した場合、入れ子になったプライマーを用いて配列特異的変異が起こる。本研究では、ガイドRNA構築のために、1.5%アガロースゲルから100塩基対バンドを切除した。細胞トランスフェクション後、ガイドRNA切断の検証を2.5%アガロースゲルを用いて可視化した。180塩基対PCR産物は、インデル形成を示すバンドシフトを有するノーカット制御および異なるクローンを示した。異なるガイドRNAは、異なる切断効率を持っていました。編集された対立遺伝子のCRISPR-Cas9再切断を避けるために、PAM配列はGからAにサイレント突然変異を使用して改変された。PAMシーケンスのサイレント突然変異は、1つまたは2つの対立遺伝子への正常な統合をスクリーニングするのに役立つユニークな制限サイトを生成しました。ゲノム編集幹細胞株は、下流の分化や機能アッセイに使用して、ヒトの発達や疾患に与えられた突然変異の影響を研究することができます。この方法論は、ヒト疾患の多種多様に関与する特定のヘテロ接合性またはホモ接合性突然変異を有する等元性細胞株の生成を可能にする。これらの疾患モデルは、新しい細胞療法の開発への道を開くだけでなく、創薬のためのプラットフォームを提供します。
