Le micro-modelage est un outil très puissant car il permet d’étudier les réponses cellulaires pour définir les signaux biochimiques et biophysiques que les cellules rencontrent chez les organismes vivants. Contrairement à d’autres méthodes, le décrit est plus flexible pour la génération de motifs. Par exemple, il permet la production de gradients et de micro-modèles de protéines multiples avec une reproduisabilité élevée.
Notre groupe utilise LIMAP non seulement pour étudier le pathfinding neuronal, mais aussi comment d’autres cellules réagissent aux signaux qui ont une influence sur la prolifération cellulaire, la différenciation et la migration. Le micro-modelage est utilisé pour répondre à des questions fondamentales en biologie cellulaire. Les connaissances acquises peuvent être utilisées pour modifier les dispositifs existants en médecine régénérative, par exemple, la réparation nerveuse après une lésion nerveuse périphérique.
Commencez par concevoir des modèles pour le modelage sur un logiciel de dessin numérique. Sélectionnez une taille d’image de 1 824 pixels de longueur et de 1 140 pixels de largeur. Concevez le modèle et enregistrez le modèle sous la forme d’un fichier TIFF 8 bits.
Avant de modeler, nettoyez et activez la surface avec un nettoyeur à plasma à l’aide d’une pression de processus de 1 000 à 1 300 millitorr et d’une puissance de 29,6 watts pendant une à cinq minutes. Utilisez un plat à fond de verre avec une taille de puits intérieur de 20 millimètres et une épaisseur de verre de 0,16 à 0,19 millimètre. Choisissez un plat de six puits ou un plat à un seul puits, selon le nombre de conditions testées.
Couper les pochoirs PDMS dans des conditions stériles pour s’assurer qu’ils s’insufflent bien dans le fond intérieur en verre. Retirer les garnitures intérieures de microwell avec des forceps stériles, et coller les pochoirs sur le puits de verre. Préparez une solution PLL-PEG de 0,1 milligramme par millilitre dans PBS et ajoutez 20 microlitres à chaque microwell.
Ensuite, incuber le plat à température ambiante pendant une heure. Après l’incubation, retirer 15 microlitres du PLL-PEG de chaque puits et les laver cinq fois avec 20 microlitres de PBS sans le laisser sécher. Préparez un microwell pour créer un modèle de référence en enlevant 18 microlitres de PBS d’un seul puits et en ajoutant cinq microlitres de photoinitiateur, laissant PBS dans les microwells restants.
Ensuite, utilisez un surligneur fluorescent pour marquer un puits de verre intérieur vide. Placez le puits au-dessus de l’objectif, et ajustez la mise au point objective jusqu’à ce que le logo PRIMO et le slogan prennent soin de vos cellules " sont au point. Ensuite, enregistrez la position Z du plan focal.
Pour créer un motif de référence, visualisez le bord de la microwell avec le photoinitiateur à l’aide de la lumière transmise à travers la caméra, et choisissez le symbole roi dans le menu de droite. Sélectionnez PRIMO pour télécharger le modèle de modèle désiré, qui sera projeté sur le roi en tant qu’unité de conception. Naviguez dans une région périphérique du microwell, sélectionnez Lock et ajustez la mise au point à la position Z de l’étalonnage.
Ensuite, sélectionnez Jouer pour commencer à photopatterning le modèle de référence. Lavez le microwell de modèle de référence trois fois avec 20 microlitres de PBS pour enlever le photoinitiateur. Ensuite, ajoutez 20 microlitres de matrice extracellulaire étiquetée fluorescente, ou ECM, solution protéique.
Incuber le plat à température ambiante pendant 10 à 20 minutes, en s’assurant de bien protéger la référence de la lumière. Après l’incubation, retirer 18 microlitres de solution protéique et laver le puits trois fois avec 20 microlitres de PBS. Utilisez un microscope à épifluorescence pour visualiser le modèle de référence.
Ajustez la mise au point sur les bords du motif à travers le chemin de la caméra, et enregistrez la position Z en fonction de la meilleure mise au point. Cette position sera la mise au point laser optimale utilisée pour les modelages ultérieurs. Dans des conditions stériles, retirer 18 microlitres de PBS de tous les microwells.
Ajouter cinq microlitres du photoinitiateur à chaque puits, et homogénéiser en pipetting de haut en bas. Si vous modélisez plusieurs protéines dans le même microwell, téléchargez simultanément tous les ensembles de modèles de motifs désirés. Sélectionnez ensuite le nombre de lignes, de colonnes et de doses.
Enregistrez la configuration du modèle en tant que fichier dans le logiciel. Accédez à la zone où le modèle de référence a été produit, et ajustez la mise au point à la position Z optimale. Ensuite, sélectionnez Jouer pour lancer le photopatterning.
Les unités de conception tournent du rouge au bleu au fur et à mesure que le photopatterning est terminé. Retirez le photoinitiateur des microwells en les lavant trois fois avec 20 microlitres de PBS. Après le dernier lavage, retirer 18 microlitres de PBS de chaque microwell, et ajouter 20 microlitres de solution protéique ECM.
Incuber le plat à température ambiante de 20 à 30 minutes, en s’assurant de le protéger de la lumière. Ensuite, retirez 15 microlitres de la solution protéique ECM et lavez les microwells trois fois à l’aide de 20 microlitres de PBS. Pour éviter la liaison croisée, ajouter 20 microlitres de PLL-PEG, ou BSA, aux microwells, et incuber le plat à température ambiante pendant une heure, dans l’obscurité.
Ensuite, retirez 15 microlitres de l’agent bloquant, et lavez tous les microwells trois fois avec 20 microlitres de PBS. Retirez 18 microlitres de PBS et ajoutez 5 microlitres du photoinitiateur à chaque microwell, en veillant à ce que le photoinitiateur soit homogène sur toute la surface des microwells. Naviguez vers le premier microwell et chargez la configuration du modèle précédemment enregistrée.
Ensuite, sélectionnez les actions à modeler pour le deuxième tour de photopatterning, en vous assurant de désélectionner les actions du premier tour. Après la photopatternation, retirer le PLPP en le lavant avec du PBS. Ensuite, incuber pendant 20 minutes avec la deuxième solution protéique ECM, et laver les microwells trois fois avec 20 microlitres de PBS.
Utilisez un microscope à épifluorescence pour visualiser et visualiser les motifs imprimés. Lorsqu’il est prêt à plaquer les cellules, ajouter un millilitre de milieu avec des cellules au verre intérieur bien, en s’assurant de couvrir tous les microwells. Et placer le plat de culture dans un 37 degrés Celsius, et 5% incubateur de dioxyde de carbone.
Afin de s’assurer que les modèles générés sont une représentation précise du modèle, des mesures d’intensité de fluorescence sont obtenues le long des bandes, et à partir d’une région d’arrière-plan correspondante au-dessus de chaque bande. Des micro-modèles définis reproductibles sont réalisés allant de 20 à 2 micromètres de largeur, mais un effet de bord peut être observé lors de l’impression de 20 micromètres ou plus. Les concentrations de protéines définies sont obtenues soit en couveant avec des concentrations variables de protéines, soit en variant la dose laser utilisée pour fendre le film antiadhésif.
L’intensité laser est proportionnelle au niveau de gris du modèle, générant des gradients d’éclairage UV. La mesure du profil d’intensité de fluorescence le long de la bande de gradient, est linéaire dans le modèle de modèle, et dans le modèle de gradient généré. Ce protocole peut être utilisé pour créer des micro-modèles avec plusieurs protéines dans le même microwell, tels que des motifs croisés avec des rayures horizontales de fibronectine, et des bandes verticales de laminine.
Mais une étape de blocage est nécessaire pour empêcher la liaison croisée des protéines. Les modèles croisés générés peuvent ensuite être utilisés pour des analyses cellulaires avec des lignées cellulaires neuronales, ou des cellules CAO, ou des neurones primaires, rats DRGs. Lors de l’exécution de ce protocole, les aspects les plus importants à retenir sont de faire un étalonnage du système approprié, ne pas laisser les microwells sécher pendant les multiples étapes de lavage, et en utilisant les bons paramètres pour l’incubation et le blocage des protéines.
L’analyse du comportement cellulaire implique généralement une microscopie, comme la fluorescence, le time-lapse, l’AFM. D’autres caractérisations peuvent comprendre d’autres méthodes biochimiques, telles que le profilage de l’expression génétique.
