1. 蚊子

</ol>2. 蚊子喂食
<ol><li>人工液体饮食的制备<ol>
<li>在层流柜中无菌条件下准备人工液体饮食。通过在初始液体饮食（i-liq_diet中添加以下内容，准备丰富的液体饮食（r-liq_diet）;Dulbeco 的改性 Eagle 的中度 [高葡萄糖与 L-谷氨酰胺]，见表 1： 0.55 g/L ATP，1 g/L 胆固醇和 200 g/L 牛血清白蛋白 （BSA）。彻底混合所有成分，并使用 0.45 μm 微过滤器过滤。 注意：不要储存饮食;准备饮食新鲜从股票解决方案为每个实验，因为他们失去质量时存储。表1描述了饮食的组成部分。</li>

除了提供必要的营养外，人工餐还需要吸引和刺激雌性蚊子以与脊椎动物新鲜血液为食的方式进食。本文描述的人工无血饮食，与脊椎动物血液喂养组相比，完全感染雌性蚊子的繁殖率增加了20%。这种间接的吸引力测量可以通过使用人工测量仪来进一步澄清，以确认人工饮食比新鲜血液更有吸引力，对蚊子更有吸引力。
观察到饮食对幼虫死亡率的影响最大，来自以血液喂养的蚊子的幼虫，这表明与新鲜血液相比，稳定成分的人工饮食有助于降低死亡率，提高蚊子繁殖的成功。血粉的较不可预测的结果可能来自成分17的宿主变异和血液中可能干扰蚊子生理22的分子的存在。上述事实强调了高质量蚊子饲养新鲜无血饮食的优点。
总体而言，我们研究中产卵的平均数量与一些昆虫中报告的卵子数量相比较低，但卵子的平均数量与以人类血液为食的A.gambiae实验室培育的菌株（22.6 ~ 5.5卵/母）23相当。以新鲜血液或人工膳食为食的实验组之间没有观察到显著的统计差异（表2），表明采用我们配制饮食的人工膜喂养系统足以维持和繁殖圈养的阿诺菲蚊菌群。
人工无血膳食可维持艾蚊菌群22，但当应用于阿诺菲斯蚊子时，它们有限或没有成功11。最近，一种以血浆为基础的人工餐为阿诺菲勒蚊子描述了24，但喂养率和繁殖潜力很低。我们的研究结果代表了最先进的（由冈萨雷斯和汉森11审查）的实质性进步，因为我们的配方r-liq_diet有类似或更好的性能比标准的脊椎动物血粉。进一步改进存储稳定性和成本应扩大其应用范围。

病媒传播的疾病影响全世界数百万人，每年造成数百万人死亡。它们由感染产生疾病的微生物（原生动物、病毒）的昆虫传播，这些微生物在以受感染宿主的血液为食时获得。随后，受感染的病媒在下一次血餐期间将病原体传染给新的宿主。疟疾是最致命的病媒传播的疾病，由几种不同种类的阿诺菲勒蚊子传播，影响世界人口的40%。疟疾原虫寄生虫每年造成40多万人死亡，其中大多数是5岁以下的儿童（世界卫生组织）。雌性疟原虫在以脊椎动物血液为食时，在人类和其他动物之间传播疟原虫，这是卵子生产发育的必要步骤。
目前消灭疟疾和其他新出现的致命蚊媒传播疾病的策略，有赖于制定创新的蚊虫控制策略3、4、5，其中包括将大量在昆虫中繁殖的蚊子释放到野外。然而，一个关键的限制因素是依赖新鲜血液供应，以便有效地饲养和繁殖蚊子。脊椎动物血液的可变成分会对蚊子的生育能力和后代健康产生负面影响，并可能限制圈养繁殖地的可靠性和可持续性。蚊子的释放和控制计划需要大规模的蚊子生产系统和定期供应大量的脊椎动物血液。这是蚊子生产的主要障碍，并引发了一系列与使用活体动物相关的道德问题，以及相关要求苛刻的安全法规造成的后勤限制。这使得蚊子群落的维持和安全费用很高，对目前蚊子饲养做法的可持续性提出了挑战，特别是在疟疾威胁大得多的低收入国家。
最近研究的重点是开发模仿脊椎动物血粉的血液替代品，但到目前为止，只有有限的成功取得了6，7，8，9。成功的人工饮食需要 （1） 引发全雌性蚊虫的侵放，（2） 触发维特洛根宁生产，（3） 生产大批量的活卵，（4） 产生热的后代10。此外，人工饮食具有标准成分，因此对于用于研究和控制目的的蚊子生产更可靠。已经为伊蚊开发了成功的无血饮食（由冈萨雷斯和汉森11审查），但不包括阿诺菲尔斯，现有的人工饮食含有一种噬菌体（例如ATP10），一种蛋白质来源，用于卵子成熟6，12，碳水化合物作为能量来源，氨基酸（aa）13是卵子生产的基础，是蚊子生育能力的主要限制因素。人工无血饮食也需要提供胆固醇15，从而改善卵子的产生。在这里，我们描述了一种针对雌性阿诺菲斯蚊子的人工无血饮食，并证明它具有与高品质脊椎动物血粉一致且同等的性能。

a blood-free diet to rear anopheline mosquitoes

用r-liq_diet喂养的雌性蚊子数量（89%）明显高于用血喂养的雌性（56%）（图8）
生育力和生育力
第一个淋病周期的女性生育力和生育能力用于评估i-liq_diet和r-liq_diet的营养质量。雌性以新鲜脊椎动物血液喂养，平均产下24~11个卵子，而以r-liq_diet喂养的雌性平均产下25~5个卵子（表2）。在i-liq_diet喂养的雌性没有观察到产卵。
F1 死亡率
F1蚊子的适应性在以脊椎动物血液或r-liq_diet喂养的菌落之间被评估。有记录到幼虫、幼虫和成人死亡率。与喂食r-liq_diet的蚊子相比，用血喂养的蚊子的变异性（平均[SEM]的标准误差）较高（图9和表2）。以血液或r-liq_diet喂养的F1代蚊子的死亡率和存活率相当。
F1 预期寿命
据美国疾病控制和预防中心估计，野生成年雌性蚊子活一个月，但存活时间可能超过1⁄2周，雄性蚊子活一周左右，只靠花蜜和其他糖源进食。值得注意的是，父母食物摄入量的差异可能会影响蚊子后代17的生存。在我们的实验中，成年女性和男性血液（女性24.5~6.8;男性18.5~6.9）和r-liq_diet（女性：22.5~8.1;男性：11.9~6.9）组平均预期寿命平均值（表3），女性预期寿命与男性相比增加。
F1 车身尺寸
翼长被用作成人体型的指标。与其他物种相比，阿诺菲勒成年蚊子是中小型蚊子，翅膀长度在2.8至4.4毫米之间，翼长18。喂食r-liq_diet的F1厌食蚊子的成年体型在预期范围内，与用血喂养的昆虫蚊子相似（图10）。
统计分析
提供的数据代表至少三个独立实验的平均值（除非另有说明）。错误条表示 SEM。当数据跟随高斯分布时，使用学生 t 测试对独立组进行比较，否则应用曼-惠特尼测试。使用费舍尔的精确测试对人工饮食喂养组之间的差异进行了分析，在P = 0.05下被认为是显著的。
