سابقا ، تم إنشاء بروتوكولات للثقافة الخلايا الجذعية الظهاريه من اي جهاز الهضمي في شكل ثلاثي الابعاد (3D) الهيكل التنظيمي13. ويمكن الاحتفاظ بهذه العضوية المعوية في الثقافة لفتره طويلة من الزمن13، والسماح للدراسة الوظيفية للمريض-والأمعاء خصائص الظهاريه الموقع المحدد14. فهي مستقره وراثيا وظاهريا ويمكن تخزينها ، مما يسمح التوسع علي المدى الطويل و بيوانكينج13.
وقد أظهرنا مؤخرا ان تشكيل LD يمكن قياسه بسهوله في الأمعاء المعوية البشرية في الفحصالسادسلتشكيل الهوية العامة (df). عند التعرض للزراعة العضوية لمده 16 ساعة ، تولد الorganoids LDs لحماية الخلايا من السمية المستحثة بالدهن. عندما تركيزات الزراعة العضوية مرتفعه جدا ، تموت الخلايا من قبل المبرمج بوساطة caspase6. وقد تبين سابقا ان الفحص الذي قامت به هذه المبيدات يعتمد إلى حد كبير علي DGAT1 علي النحو المشار اليه في الغدد العضوية المشتقة من المرضي DGAT1 المتحولين وباستخدام مثبطات DGAT1 الخاصة6.
لفحص DF الموصوفة بالتفصيل هنا ، يتم استزراع العضو العضوي ثلاثي الابعاد من الخزعات المعوية ويتم المرور أسبوعيا عن طريق التشويش في خلايا واحده تشكل بسهوله التشكيلات العضوية الجديدة. لتشغيل فحص DF ، يتم طلاء ~ 7,500 الخلايا الاحاديه المشتقة organoid في كل بئر من لوحه 24 بئر. تتشكل organoids علي مدي عده أيام ، حضنت بين عشيه وضحيها مع 1 مم الزراعة العضوية وملطخه مع LD540 ، وهي صبغه الخلية الفلورية-نفاذيه LD الخاصة التي تسهل التصوير. ثم يتم قياس تشكيل LD عن طريق المجهر البؤري ، قارئ لوحه الفلورسنت ، أو تدفق الخلوي.
