フェムト秒パルスレーザーは、多光子ミスクロスコピーに幅広い用途があります。このプロトコルは、小型で堅牢で安価なフェムト秒全正常分散ファイバレーザーを製造するために使用できます。市販の固体超高速レーザーと比較して、この技術で製造されたレーザーは、市販の部品だけで構成されているため、はるかにコストが低くなります。また、ファイバーレーザーは水冷を必要としませんので、システムのサイズが小さくなります。最後に、ファイバコンポーネントはアライメントを必要とせず、システムを振動に強くします。市販のシステムとは異なり、このレーザーは、不要なビームをブロックするためのカバーを持っていません。レーザーを組み立て、操作するには経験豊富な人員が必要です。一部の実験は、書面による指示に従う際に、呼び出されていない詳細を見逃す可能性が非常に高いため、再現不可能であるように見えます。ビデオデモンストレーションでは、視聴者は何も見逃すことはありません。より価値のある光ファイバー材料を使用する前に、スプライシング装置の適切な性能を確保するために、シングルモードファイバ(SMF)をスプライスして開始します。繊維除去ツールで約 30 ミリメートルの繊維を取り除きます。壊れやすい繊維で作業する場合は、カミソリの刃を使用してバッファーを慎重に剥がすことができます。エタノールまたはイソプロパノールとリントフリーの組織を使用して、剥がされた繊維を洗浄します。拭き取りながらブンブン鳴る音は、繊維が十分にきれいであることを示します。次に、ファイバ・ホルダーをファイバー・クリーバーの上に置き、ブレード、包丁のファイバークランプ、およびファイバーホルダーがすべてきれいであることを確認します。約25ミリメートルの除去されたきれいな繊維を包み込むため、ファイバホルダーに慎重に繊維をロードします。包丁の繊維クランプを静かに閉じます。ファイバの余分な張力を避けるために、クランプを再度開いて閉じます。カットボタンを押すと、クリーバーが自動的にファイバーを切断します。プラスチック丸みを帯びた先端を持つピンセットを使用して、切り取った部分を繊維からシャープ処理容器に移動し、繊維ホルダーをフュージョンスプライサーに移します。手順を繰り返して、2 番目のファイバーを切断します。一緒にスプライスされる2つの繊維は、繊維スプライサー内の繊維ホルダーによって互いに対向する端を切断する必要があります。スプライサのカバーを閉じ、コア直径、モード フィールド直径、クラッディング直径などのパラメータを設定します。整列方法を[クラッディング]に設定し、[設定]ボタンを押すと、スプライサーが自動的に整列します。各ストップで[設定]ボタンを押して、位置合わせの品質を確認します。スプライスは自動的に行われます。スプライサーの品質コントロールと、領域のカメラ ビューを使用して、スプライスの品質を確認します。良いスプライスは、均一なクラッディング境界と、スプライスの分岐点が見えないよう、繊維に沿って均一な明るさを有します。次に、スプライサーカバーとファイバーホルダーの1つを開きます。必要に応じて、スプライスを保護するために繊維スリーブを追加し、スプライサーのヒーターを繊維上に成形するために使用することができる。コンバイナー出力をイッテルビウムドープ活性繊維にスプライスします。コンバイナー出力ファイバを切断するには、前述の手順に従います。そのクラッディングの形状のために、アクティブな繊維は、最初に切断され、後で除去されるシングルモードファイバの一部でスプライスする必要があります。シングルモードのファイバーを、ワイヤーカッターでスプライシングポイントから約2センチカットします。次に、アクティブファイバをバッファレスシングルモードファイバの2センチメートルでキャップしたままにするシングルモードファイバの全長と0.5センチメートルを取り除きます。アクティブなファイバをクリーバーにロードして、シングルモードファイバのみがファイバクランプによってクランプされていることを確認します。この時点から、前述の手順に従って、ファイバを切断およびスプライシングします。安全は最優先事項です。鋭利なオブジェクトボックスにファイバーフラグメントをすべて入れておいてください。また、レーザー安全ゴーグルは、ポンプが動作しているときはいつでも警告する必要があります。オシロスコープをオンにし、トリガレベルを30ミリボルトに設定して、ACカップリングモードに設定します。光スペクトルアナライザフォトダイオード入力ファイバを単色入力に移動し、デバイスをOSAモードに設定します。次に、波板を調整してレーザーの位相をロックします。四半期波プレート 2 を数度前後に回転させます。モードロックスペクトルは、2つの安定したピークとそれらの間の高原で構成されています。一方、オシロスコープで安定したパルス列を観察します。モードロックスペクトルが観測されない場合は、クォーター-ウェーブプレート 1 を 1 方向に数度回転させて、前のステップを繰り返します。それでもスペクトルが観測されない場合は、複屈折フィルタを数度回転させて、プロセスを繰り返します。ファイバーレーザー加工完了時にモードロック動作を検証した。レーザー発振器からのパルススペクトル出力は、数値シミュレーションで予測されるモードロックを示す特徴的な猫の耳の形状で1070ナノメートル付近を中心とした。モードロックのさらなる診断として、パルス持続時間およびパルス繰り返しパワースペクトルを、それぞれ自動相関器と無線周波数スペクトルアナライザを用いて測定した。70フェムト秒のパルス持続時間を測定した。パルス安定性は、平均出力電力とパルススペクトルを連続的に監視することにより試験した。レーザーセットアップを振動減衰付きのフローティング光学テーブルに取り付けた場合、パワードリフトはアクティブな冷却なしで24時間にわたって3.5%未満でした。モードロックを検証した後、単純な試験対象と生物学的サンプルを用いてイメージング性能を試験した。蛍光は、信号がサンプル面に送達されるレーザーパワーに二次的に依存していることを確認したパルスパワーの調整中に測定した。染色された生物標本と染色されていない生物標本を、独自のファイバーレーザーを用いて画像化した。2光子励起の追加検証として、多色蛍光微小球の収集されたハイパースペクトル画像を、市販のダイオードレーザーを用いて線形励起によって撮影された画像と比較した。最後に、ダイオードレーザーとカスタムFSファイバーレーザーで励起された緑と赤のビーズの正規化スペクトルを比較した。空きスペースコンポーネントは、さらに堅牢性と移動性を高めることができる対応するファイバ部品に置き換えることができます。全繊維システムは臨床シナリオのためのカートに置くことができる。空きスペースコンポーネントは、さらに堅牢性と移動性を高めることができる対応するファイバ部品に置き換えることができます。全繊維システムは臨床シナリオのためのカートに置くことができる。この技術の影響はオープンな質問です。研究者にフェムト秒レーザー技術への新たなアクセスを提供し、新しい出版物を開発できるようになると予想しています。
