Primary Culture of Neurons Isolated from Embryonic Mouse Cerebellum

初代ニューロン細胞培養は、解剖ニューロンを体外培養で研究する理想的なモデルシステムである。この方法を使用する利点は、生体内状態にできるだけ近い数週間のインビトロ条件で解体細胞の細胞特徴を維持できることです。バイオセーフティキャビネットの下の24ウェルプレートに丸いカバースリップを入れます。各カバースリップに90マイクロリットルのポリL-オルニチンを加えます。キャップを閉じ、プレートを37度インキュベーターに2日間そっと入れなさい。培地と播種培地を準備し、摂氏4度に保ちます。DMEM/F12で動作するトリプシン溶液を準備し、4度に保ちます。培養培地と播種培地をインキュベーターに37度に入れる。24ウェルプレートをインキュベーターから取り出します。カバースリップを二重蒸留水で3回洗います。カバースリップを少なくとも2時間は完全に乾燥させます。冷たいPBSで満たされた3つのペトリ料理、冷たいHBSSで満たされた3つのペトリ料理、氷の上の冷たい解剖媒体で満たされた5つのペトリ料理を準備します。子宮の角を切り出し、氷の上で冷たいPBSで3回洗います。ここからすべてのステップは、組織の損傷を最小限に抑えるために氷上で行われるべきです。最後の洗浄ステップの後、HBSSの子宮から子犬を分離し、冷たい解剖媒体に移します。解剖媒体では、はさみで子犬を切断します。角のマグナムから軌道空洞の下の境界までカルバリウムを開きます。細かい鉗子を使用して、頭蓋骨のベースを取り除き、頭蓋骨を脳から剥がします。中小脳ペダンクルとポンの側面から始まる小脳の髄膜を慎重に取り除く。小脳のペダンクルを両方カットし、脳の残りの部分から小脳を分離します。すぐに氷の上にDMEM / F12で満たされた無菌15-mL円錐管に集めたセレベラを置きます。DMEM/F12で1分間、4度で1,000gのチューブを遠心します。これを3回繰り返し、ピペットで上清を静かに取り除き、新鮮なDMEM/F12でペレットを再懸濁します。2ミリリットルのプリ温めたトリプシンとピペットを加えて混ぜ合わせます。チューブを37度の水浴に12分間入れます。インキュベーション後、チューブを安全キャビネットに持ち込み、10 mLのDMEM/F12を加えてトリプシンを不活性化します。混合物を1,200gで5分間遠心分離する。上清を捨て、新鮮な培地で再中断し、その後3回遠心分離します。DMEM/F2で滅菌プラスチックピペットを事前に濡らします。同じチューブに3.5ミリリットルのDNAse作業溶液を加えます。液体が均質な乳白色になるまで、ピペットで組織を数回トリチュレートします。冷たいDMEM/F12を10mL加え、遠心分離機に進みます。サンプルを1、200gで4度で5分間遠心する。ペレットを邪魔することなく上清を慎重に除去します。インキュベーターから播種培地を取り出します。500マイクロリットルの前温めの播種培地を加え、ピペットを使用してよく混ぜます。ヘモサイトメーターを使用して細胞を数えます。細胞懸濁液を、1ミリリットル当たり500,000細胞の密度に播種培地で希釈する。カバースリップの中央にある各ウェルに90マイクロリットルの混合物を加えます。プレートをインキュベーターに37度3~4時間置きます。インキュベーション後、各ウェルに500マイクロリットルのプリウォーム培養培地を加える。プレートを37度でインキュベーターに交換します。7日後、古い培地を新鮮な培養培地II.に交換し、対応する数値組織で別々の24ウェルプレートを準備し、各ウェルにPFA 4%の100マイクロリットルを追加します。培養で所望の日の後、元の培養プレートのウェルからカバースリップをそっと取り除き、前のステップで説明したPFAプレートの対応するウェルに入れる。PFAをウェルに加え、カバースリップを完全に浸します。PFAプレートを30~120分間4度に保ちます。インキュベーション後、プレートを室温に戻します。PBSで蓋のスリップを5分間3回やさしく洗います。図2は、E18から始まる小脳細胞培養を示す。カルビンビンの免疫蛍光は、3日間の軸索拡張を有するプルキンジェニューロンをインビトロで示す。インビトロで7〜10日までに、樹状プロセスが明らかである。21日で、複雑な樹状枝が存在する。図3は、様々な胚期から始まる小脳細胞培養を示す。カルビンジンの免疫蛍光検出は、インビトロで18日後にのみ初期軸索を有するプルキンジェニューロンを示す。プルキンジェニューロン培養はE14とE15で始まり、樹状突起を発症しますが、E18が出発点であるときに発症するものほど複雑になることは決してありません。図4は、インビトロで21日後に異なる細胞タイプがE18培養物から発達することを示している。カルビンビングリーン免疫蛍光はプルキンエニューロンを示す。BおよびCにおけるパルブアルブミンに対する赤い免疫蛍光は、阻害性インターニューロンを示す。EおよびFにおけるナトリウム電圧ゲートチャネルの赤い免疫蛍光は、顆粒ニューロンを示す。現在のプロトコルは費用対効果が高く、実施しやすいものです。このビデオの最後に、小脳ニューロンを収集し培養し、所望のDIVでそれらを修正することができます。
