ヒト被験者の深度分解能を有する水、タンパク質、脂質の直接測定は、皮膚関連疾患において、スキンケア製品の性能を特性化するために非常に重要である。この方法は、その後の分析と共に、化学測定を利用して化学情報を抽出します。この技術の主な利点は、臨床ラマンデータセットを、分光アーティファクトのすべてのソースを特定、除外、修復するための技術的専門知識を欠く訓練を受けた機器オペレータによって収集できることです。結果のデータ・セットを処理して、分析前の日付から除外する必要がある外れ値を識別することができます。データ分析の際に重要な課題は、外れ値の除去と、データ・セット内の主要コンポーネントの数の識別です。このビデオのアプローチは、臨床データセットと化学メトリックスアプローチの予備知識を活用して、深度分解能で水、タンパク質、脂質を正常に抽出します。この手順のデモンストレーションは、C&Tラボの技術者である李陽です。まず、in vivo共焦点ラマン器具の撮像窓と密接に接触して、印の病変体部位または制御部位を被験者に配置させる。部屋の光がイメージングに与える影響を避けるために、ウィンドウ全体を覆っていることを確認してください。次に、ソフトウェアを開き、ここに示したものと同様のスペクトルが見えるまでフォーカスを移動します。その後、焦点を皮膚表面から10ミクロン離します。ここに示す周波数領域で2ミクロンのステップサイズを持つ26ステップのデータを収集し、1秒間の露光時間を使用します。各領域に対して 8 回の反復を測定し、合計で最大 15 分間持続します。まず、コマンド ウィンドウと MATLAB を使用して、収集したデータのファイル拡張子を ric から mat に変更します。次に、MATLAB ソフトウェア プラットフォームに MATLAB ファイルをロードします(次に示すように)。自動加重最小二乗法を使用して、PLS_Workspaceウィンドウに移動し、インポートされたデータセットを右クリックし、分析までスクロールし、[その他のツール]を選択して[前処理]をクリックして、データ セットのベースラインを修正します。ポップアップ表示されるウィンドウで、[表示] をクリックします。次に、[使用可能なメソッド] ツールバーをスクロールダウンして、[自動加重リース平方基線] フィルタに移動し、[追加] を選択します。次に、[OK] をクリックしてオプションを設定し、データに前処理を適用します。これをSpectra_baselineとして保存します。次に、コマンド ウィンドウに戻り、ベースライン修正結果を使用してデータを置き換えます。次に、テキスト エディタに移動し、次に示すようにプログラムを実行します。これにより、2910 ~ 2965 の逆センチメートルの値を合計して、26 個の連続したステップの測定から各ラマンスペクトルの強度値を取得し、Excel ファイルに格納します。MATLAB で、次に示すように、ワークスペースに移動し、Depth_saveのデータのパスを設定します。次に、MATLABの linspace 関数を使用して、計測器のオフセット値を 26 から 260 に補間するために、ここで説明するプロセスを使用します。このプロセスでは、スプライン法を使用して 26 ~ 260 の強度値を補間し、新たに生成された 260 の位置値を利用します。さらに、260 の位置と強度の値をそれぞれ、ポリフィット関数の x および y 入力として使用し、度値を 20 に設定します。次に、出力係数と260の拡張位置値をポリバルの入力として使用し、最終的な260の強度値を得る。次に、平均強度を計算し、平均強度に最も近い曲線の点を見つけます。また、既知の2ミクロンのステップサイズで、皮膚表面に応じて深さの値を変更します。次に、プログラムを実行します。外側のスキンスペクトルを取り外した後にラマンスペクトルデータセットをPLS_Toolboxソフトウェアにロードし、データセットを右クリックして[分析]を選択し、[PCA]を選択します。次に、[前処理の選択] をクリックします。前処理の方法として [正規化] を選択します。次に、クロス検証に [なし] を選択します。次に、PCA 分解解析用の 3 つのコンポーネントを使用してモデルを構築します。in vivo ラーメン計器コレクションウィンドウのカバーを取り外し、参照材料のデータ収集に使用したのと同じパラメータを使用して、高周波領域の室内光スペクトルを収集します。部屋のライトの背景と比較して、部屋のライト効果係数を特定します。ここで、スコアを確認し、通常よりもかなり高い対応するスコア値を持つスペクトルを削除します。つまり、データ セット全体の 99.8% を超えるスコア値を削除します。結果のキャリブレーションXブロックデータを保存します。最後に、PLS_Workspaceブラウザに移動し、新しいファイルを編集します。[行ラベル] を選択し、[除外されたハード削除] に移動して、除外されたデータを完全に削除してから、ファイルを再保存します。まず、ラマンスペクトルベースラインを同じ方法で修正します。次に、前処理されたデータ・セットに対して PCA 分析を実行します。成分選択ボタンをクリックして成分の数とともに対数スケールで固有値をプロットし、y値として log(固有値)を選択します。多変量曲線分解能解析を実行するには、まずデータ選択ボタンを使用して、MCR_mainソフトウェアにデータセットをロードします。手動でコンポーネントの数を選択し、コンポーネント番号を 3 ~ 8 に設定します。次に、[初期推定]タブで、[純粋]ボタンをクリックします。次に、[濃度] を選択し、[実行] ボタンをクリックします。画面が更新されたら、[OK] ボタンをクリックし、[続行] をクリックして次のページに移動します。次に、[続行] をクリックし、[実装] の下で fnnls を適用します。次に、非否定性プロファイルを持つ種の数のドロップダウン メニューから 6 を選択し、[続行] をクリックします。次のページで、同じパラメータを選択して[続行]をクリックします。皮膚表面の位置を決定するために、タンパク質のラマンピーク下の領域を統合し、タンパク質シグナルの深さプロファイルを得た。スキンサーフェスは、補間された深さプロファイルからの強度値が平均強度に最も近い場所として定義されました。皮膚表面の正確な位置は、実験データポイントと一致する必要はありません。このビデオで説明したデータ収集プロトコルを使用して、合計30,862ラマンスペクトルを収集しました。この大きなスペクトル データセットには、20% のスペクトル外れ値が含まれています。適切なデータ セットを実現するには、外れ値スペクトルを適切に識別して除去することが重要です。ここでは、ルームライトの基準スペクトルに重ね合わせたルームライトからの寄与を見ることができます。前処理された共焦点ラマンデータセットに対して主成分解析を行い、固有値と使用する因子の数をプロットします。因子9に対して固有値の有意な減少が認められた。この観測は、多変量曲線分解能モデルに含める3~8因子の間で異なる原理成分の数を持つモデルを調査することを示唆している。この手順を試みる際には、スキン表面の上に深さプロファイルを開始し、皮膚表面の位置を正確に決定することが重要です。この方法論により、スキンケア製品が水、タンパク質、脂質などの主要な皮膚成分に及ぼす影響を調査できます。
