ラーバルゼブラフィッシュは、インビボ8でマクロファージの機能と可塑性を研究するための理想的なモデル生物である。幼虫ゼブラフィッシュの光学的透明性は、特にマクロファージマーキングトランスジェニックレポーターラインと組み合わせた場合に、マクロファージの挙動を直接観察するウィンドウを提供します。生画像化の可能性と幼虫ゼブラフィッシュの実験のトラクチャビリティを利用することは、ヒト疾患9、10、11、12、13、14、15に直接関連するマクロファージ機能に関する多くの重要な洞察を導いた。これらの研究の多くは、ゼブラフィッシュにおける薬物活性の高い保全(動物創薬プラットフォーム16、17、18)としての使用を支える属性)を利用して、薬理学的にマクロファージ機能を操作するための化学的介入を利用している。現在までに、これらの薬理学的治療は、主に水溶性である薬剤を必要とする浸漬、または遊離薬物の直接マイクロ注射によって提供されてきた(図1A)。これらの受動デリバリー戦略の限界には、マクロファージ機能への影響を評価することを妨げる可能性のあるオフターゲット効果および一般的な毒性が含まれる。さらに、 マクロファージに対する薬物の影響を調べる場合、薬物がマクロファージ自体に作用しているのか、それともより間接的なメカニズムを通じて作用しているのかは不明である。マクロファージ機能を調べるのに同様の化学的介入研究を行う際に、マクロファージに特異的に薬物を標的とする安価で簡単な送達方法を開発する必要はないと認識した。
リポソームは、顕微鏡、生体適合性、脂質二重層小胞であり、タンパク質、ヌクレオチドおよび薬物貨物19をカプセル化することができる。リポソームのユニラメラまたはマルチラメラ脂質二重層構造は水溶性薬物を組み込むことができる水性内腔を形成し、一方で疎水性薬物を脂質膜に組み込むことができる。さらに、サイズ、電荷および表面修飾を含むリポソームの物理化学的特性は、それらの標的を特定の細胞20、21に合わせて調整するように操作することができる。リポソームのこれらの特徴は、彼らに薬剤を提供し、現在の治療レジメン20の精度を高めるための魅力的な車両を作りました。リポソームはマクロファージによって自然に貪食される(アブレーション実験22のためにマクロファージにクロドロナートを送達する際の日常的な使用によって利用される特徴)、マクロファージ特異的薬物送達のための魅力的な選択肢として提示される(図1B)。
このプロトコルは、親水性ポリマーポロクサマー188でコーティングされた青色蛍光リポソームへの薬物の製剤を記述し、リポソーム表面に保護層を形成し、薬物保持性を高め、優れた生体適合性23を有することが示されている。ポロクサマーは、我々の以前の研究としてリポソームの表面コーティングのために選ばれた、ポリエチレングリコール修飾リポソームと比較すると、ポロクサマー修飾リポソームは、点滴注入後のウサギの注射におけるラット足および類似の薬物動態の皮下に続くより良い生体適合性を示した23。また、プロトコルは、幼虫ゼブラフィッシュへのマイクロインジェクションとライブイメージングについても説明され、リポソームの分解や細胞質学的薬物送達に必要な細胞内コンパートメントへのマクロファージターゲターゲの能力と局在性を評価します。概念実証として、我々は以前に急性痛風炎症の幼虫ゼブラフィッシュモデルで彼らの活性化を抑制するためにマクロファージに2つの薬物を標的にこの技術を使用しました24.この薬物送達技術は、関心のある薬物のマクロファージターゲを確実にしたいゼブラフィッシュの研究者が利用できる化学的「ツールキット」を拡大する。
