このプロトコルは、マイクロピペット吸引技術を用いて、合成および実ポリマー脂質小胞の膜機械的特性の信頼性の高い測定を容易にする。これは、単一の実験で評価される膜の柔軟性と不安定性を引き伸ばすことができる唯一の技術です。このプロトコルには、小胞の調製から機械的評価までの一連の手順が含まれます。忍耐強く、技術的な問題が発生したときに解決することは非常に重要です。これらの手法の視覚化は、毛細管表面を適切に処理する方法と必須のプレストレスステップを実行する方法を理解するために重要です。欠陥のない小胞を表示します。デモンストレーション手順は、ハイブリッドポリマー脂質小胞の開発に取り組んでいる研究室の博士課程の学生、マーティン・ファウキニョンです。手順を開始する前に、冷たいガラスのマイクロピペット毛細血管をホルダーに垂直に置きます。そして、先端が一晩で作りたてのブドウ糖牛血清アルブミン溶液に浸漬されるまでホルダーを下げる。翌朝までに、溶液は毛細血管作用によって先端に約1センチメートル上昇するはずです。柔軟な融解シリカ毛管を装備した500マイクロリットルガラスシリンジを使用して、各ピペットにグルコース溶液を充填します。次いで各ピペットから溶液を吸引し、ピペットを新鮮なグルコース溶液で補充する前に、すべての血清が除去されるまで数回再生する。電気構造チャンバを準備するために、まず適切な有機溶媒でITOスライドを洗浄し、オーメータで導電面を識別します。各スライドの導電性側に、粘着テープで電線を取り付けます。約5マイクロリットルの溶液が毛管作用によって回収されるまで、1本の毛細管を円形溶液に浸す。1つのガラスITOプレートの中心に接触して、そっとスライド全体に溶液を広げます。溶剤が完全に蒸発したら、ちょうど実証したように、溶液をさらに2回塗布します。堆積の領域の周りに開いたOリングスペーサーの両側にシリコンフリーグリースの層を追加する前に。次に、スペーサーの上部に第2のITOガラス板の導電面を置き、電気構造チャンバを真空下に3時間置き、有機溶媒の痕跡を除去する。ジャイアントユニラメラベシクルの電気構造のために、発電機に電線を差し込みます。ジェネレータ周波数を10ヘルツに、振幅を2ボルトに設定し、ピークをピークにします。電圧が設定されたら、0.8ミリメートルの内径の針を装備したシリンジを使用して0.1モルショ糖溶液を1ミリリットル注入し、適用された電圧と周波数の下に75分間チャンバーを残します。電気の終わりに、発電機をオフにします。そして、空気泡がチャンバー内で生成されるまで、溶液の少量を吸引するために1ミリリットルのシリンジを使用してください。チャンバーに泡を移動し、小胞がスライド表面から切り離されるのを助けるために、チャンバーをわずかに傾けます。そして、溶液の全容を注射器に吸引する。次いで、ベシクル溶液を1ミリリットルのプラスチックチューブに移します。マイクロマニピュレーション用の材料を設定するには、吸引して水の流れ、純水のタンクを1つのホルダーに作り出す。そして、任意の気泡を排除し、陽圧を作成するために、タンクを上げながら軽くタップします。先端に滴が形成されるまで、新鮮なグルコース溶液で血清コーティング毛細管を充填します。金属ホルダーからシリンジチューブを取り外してホルダーの端にわずかな水の流れを作り、キャピラリーを逆さまにしてグルコースドロップを水の流れに接続します。次に、キャピラリーとホルダーを一緒にねじ込みます。ピペットを配置するには、カスタムメイドのアルミニウムステージから2つのガラススライドを真空グリースと一緒に接着します。そして顕微鏡の段階にスライドを取付ける。1ミリリットルのピペットを使用して、0.1モルグルコースで2つのスライド間に半月板を形成する。ピペットとそのホルダーをマイクロマニピュレーターのモーターユニットに置きます。クランプノブを締め、コントロールパネルのジョイスティックを粗いモードで使用します。グルコース半月板付近のマイクロピペットを下げます。細かいモードを使用して、半月板の中心に先端の位置を調整します。先端をブドウ糖に浸して、その外面と内面をきれいにします。数分後、半月板から毛細血管を引き出し、ブドウ糖を新鮮な半月板に置き換えます。0.1モルショ糖のジャイアントユニラメラベシクルの2マイクロリットルを20ミリリットルのマイクロピペットチップに吸引する。半月板に小胞を導入します。顕微鏡を使用して、スライドチャンバーの底部の小胞を観察します。小胞がわずかに収縮したら、吸引ピペットを再挿入し、ピペットの先端に焦点を当てます。次に、水タンクのベースライン高さを、パーティクルの運動が停止する圧力に設定します。蒸発を防ぐためにミネラルオイルで半月板を囲みます。マイクロピペット吸引実験を行うために、ピペットチップを半月板に下げる。小胞を吸引するために少量の吸引圧力を作成します。選択された小胞の膜はわずかに変動する必要があり、目に見える欠陥を提示すべきではありません。マイクロマニピュレーターを使用してピペットを高いレベルに上げて、吸引した小胞を他の小胞から分離します。水槽を約10センチメートルに下げて小胞をプレストレスしてから、タンクを上げて圧力を初期値に戻します。0.5センチメートルの高さから、膜が変動する圧力に達するまで、ゆっくりと吸引圧力を下げます。次に、圧力を高くして、先端の舌を明確に視覚化し、数ミクロンの投影長を示す。曲げ弾性率を決定するには、吸引圧力を段階的に1マイクロメートルずつ増加させる。1メートル当たり0.5〜0.8ミリニュートンに達するまで。5秒待って、各ステップの後に舌のスナップショットを撮る。面積圧縮率を決定するために、溶解の緊張とひずみは、破裂緊張に達するまで1メートル当たり0.5ミリニュートンから吸引圧力を増加させ続ける。この代表的な実験では、POPCの領域圧縮率と溶解株は、文献から期待されるものと完全に一致していた。この表では、得られたポリマーソームの典型的な値が観察できる。なお、ジブロックコポリマーから得られる膜の靭性は、三重重合体から得られるものよりもはるかに大きい。興味深いことに、ジブロックコポリマーを使用して、リポソームについて測定したものよりも強靭性を発揮するジャイアントハイブリッドユニラミラー脂質ポリマーベシクルを得ることができる。このプロトコルは、例えば、アマティックショックを通して膜の透過性を測定するために、ピペットでベシクルを測定するのに役立ちます。特にトリビューン接続を設定する際には、各ステップを厳格かつ正確に完了してください。この技術は、改変小胞の主な境界での線張力の測定を通じて、膜分裂の物理的起源を理解するために利用されてきた。
