異なるタイプの単一細胞の効率的な捕捉と十分な培養空間の提供は、複数のタイプの単一細胞1の単一細胞共培養実験に必要である。希釈を制限することは、必要な機器の低コストのために、このような実験のために単一細胞を調圧するために最も一般的に使用される方法です。しかし、ポアソン分布の制限により、最大単一細胞獲得確率はわずか37%であり、実験操作は手間と時間のかかる2を作る。これに対し、蛍光活性化細胞選別(FACS)を用いることは、単一細胞3を高効率に調製するポアソン分布の限界を克服できる。ただし、高額な計装費やメンテナンスコストにより、一部のラボではFACSにアクセスできない場合があります。微細加工デバイスは、最近、単一細胞トラッピング4、単一細胞ペアリング5、および単一細胞培養用途用に開発された。これらの装置は、単一細胞6を正確に操作する能力、ハイスループット実験を行う、またはサンプルおよび試薬の消費量を減らす能力に基づいて有利である。しかし、現在のマイクロ流体デバイスを用いて複数の細胞型を用いて単一細胞共培養実験を行うことは、ポアソン分布1、7、8、または不能の制限のために依然として困難である。2種類以上の単一セル4、5、6、9、10をキャプチャするデバイス。
例えば、ユンらは細胞相互作用研究11のためのマイクロ流体装置を報告した。この装置は1つの部屋の細胞を対一にするために確率的な方法を使用する。ただし、デバイス構造の幾何学的制限により、2 つの異なるセル タイプのペアリングのみが実現できます。Leeらからの別の報告は、単一細胞12を捕捉し、対にする確定的方法を実証した。この装置は、確定的方法によってペアリング効率を高めるが、細胞のペアリングに必要な長時間の操作時間によって制限される。具体的には、第2の細胞捕捉は、24時間後に最初に捕捉された細胞が表面に取り付けられた後にのみ行うことができる。液滴系マイクロ流体装置は依然としてポアソン分布に限定されており、非接続細胞でのみ使用でき、栽培過程で培養液を変更することはできないことが分かります。
以前は、決定論的な流体力を利用して複数のタイプの単細胞を培養室に捕捉し、その後細胞共培養実験を行って分析できるマイクロ流体「流体力学シャトルチップ」を開発しました。細胞相互作用における個々の細胞移動行動14.流体力学シャトルチップは、それぞれが蛇行バイパスチャネル、キャプチャサイト、および培養室を含むユニットの配列セットで構成されています。蛇行バイパスチャネルと培養室との間の流れ抵抗の差を使用することにより、特別に設計された操作手順により、異なるタイプの単一細胞を培養室に繰り返し捕捉することができる。特に、培養室の十分なスペースは、細胞の捕捉中に細胞がフラッシュされるのを防ぐだけでなく、細胞が広がり、増殖し、移動するのに十分なスペースを提供し、生きている単一細胞相互作用を観察することを可能にする。この記事では、このデバイスの生産と詳細なプロトコル手順に焦点を当てます。

細胞の挙動に対する細胞間相互作用の影響を理解するために、キャプチャ効率と動的なライブ細胞画像観察を効果的に組み合わせることが重要です。このチューブは、より大きなチャンバ内の複数の単一細胞を非常に効率的に捕捉し、十分な培養ベースを提供し、複数の単一細胞相互作用行動の動的追跡を行うことができます。PDMS デバイスを準備して開始します。100マイクロリットルのトリクロロシランを付した計量皿にウエハーモールドをデシケータに入れ、15分間真空を塗布します。真空を止め、デシケータのウエハーモールドを37°Cで少なくとも1時間はアルカリ化します。PDMSベースとPDMS硬化剤を10対1の比率で混合し、15センチメートル皿のウエハーモールドに合計20グラムの混合物を注ぎます。15センチメートルの皿をデシケータに入れ、1分半真空を塗ります。次に、デシケータから皿を取り出し、窒素ガスを用いてPDMS内の残留気泡を除去します。2〜4時間摂氏65度のオーブンに皿を置くことによってPDMSを治します。完了したら、ウエハ金型からPDMSレプリカを取り出し、PDMS上で1.5ミリメートルの入口と0.5ミリメートルの出口をパンチします。PDMSレプリカとスライド面を取り外し可能なテープでクリーニングします。酸素プラズマで表面を処理します。次に、PDMS レプリカをスライドに手動で合わせて、相互に接触させます。PDMSスライドを65度の摂氏オーブンに1日放置します。翌日、PDMSデバイスをPBSで満たされた容器に浸します。デシケータに入れ、15分間真空を適用します。装置を細胞培養フードに移し、30分間UV光で殺菌します。PDMSデバイスバッファを培地に交換し、摂氏4度で1日インキュベートします。細胞が70〜80%の合流性を達成したら、培養培地を取り除き、5ミリリットルのPBSで3回軽く洗います。1つのマイクロモル蛍光色素を含むDMEM/F-12培地をWNT5B sh4およびpLKO-GFP細胞に1ミリリットル加え、室温で30分間インキュベートします。インキュベーション後、5ミリリットルの滅菌PBSで細胞を3回静かに洗浄し、PBSを取り除き、0.25%トリプシン-EDTAの2ミリリットルを加えます。室温で4分間細胞をインキュベートし、培養皿を軽くタップして細胞剥離を促進します。WNT5B sh4およびpLKO-GFP細胞を分散させるためにDMEM/F-12培地を4ミリリットル加え、細胞を15ミリリットルチューブに移し、300 xgで3分間遠心分離します。上清を取り除き、DMEM/F-12培地の1ミリリットルで細胞ペレットを静かに再中断します。1ミリリットル当たり10~5番目の細胞の濃度で1ミリリットルの細胞懸濁液を調製し、氷の上に保管して細胞凝集を防ぎます。デバイスの出口とシリンジポンプの間にポリテトラフルオロエチレンチューブを接続します。培地を取り出し、調製したセル懸濁液の1マイクロリットルをPDMS装置の入口に加える。次に、1分間に0.3マイクロリットルの流量でシリンジポンプを使用して、セル懸濁液をデバイスにロードします。DMEM/F-12培地を1マイクロリットルのデバイスの入口に1マイクロリットル追加し、シリンジポンプを使用してデバイスにメディアをロードします。0.3マイクロリットルのDMEM/F12媒体を、毎分10マイクロリットルの流量でシリンジポンプを備えた装置に積み込みます。細胞がロードされたら、チューブを取り出し、入口と出口をポリオレフィンテープで密封して閉じた培養システムを作ります。PDMSデバイスを10センチメートルの培養皿に移動し、10ミリリットルの無菌PBSをデバイスの周りに加えて培地の蒸発を避けます。培養皿を三重単細胞培養用インキュベーターに移す。PDMS装置は、培養チャンバとバイパスチャネルに接続するキャプチャ部位を有する3層構造を有する。培養チャンバとバイパスチャネルの間の流れ抵抗の違いにより、セルはキャプチャ位置に流れ込み、次のセルはバイパスチャネルを通過して次の捕獲部位に向かう。還流ステップは、捕捉部位から細胞を放出し、培養チャンバーに送るために使用される。このプロトコルの細胞捕捉効率は、3種類の異なる細胞タイプで評価した。ヒトリンパ管内皮細胞、または、PIN、WNT5B特異的短ヘアピンRNAを発現するヒト口腔扁平上皮癌T2.6細胞、およびpLKO-GFPベクターコントロール細胞。実証された3つの細胞の単細胞捕捉効率は、LECsで約71%、WNT5B sh4細胞で74%、pLKO-GFP細胞で78%であった。この方法は、リンパ内皮細胞と扁平上皮癌細胞の複数の単細胞共培養に使用でき、細胞の増殖と形態を顕微鏡で観察することができる。細胞凝集体は単一のペアリング効率を低下させるだけでなく、マイクロチャネルを詰まらせることができるので、準備された細胞懸濁液が最小限の細胞凝集体を持っていることを確認することが重要です。個々の細胞間の細胞間相互作用は、細胞の挙動を制御する上で重要な役割を果たします。我々の方法は、研究者が単一の細胞レベルで細胞と細胞の相互作用を研究するのに役立つ効率的なツールを提供することができ、人口ベースの研究から容易に得ることができない新しいメカニズムの発見につながる可能性があります。
