当社のプロトコルは、細胞外の環境や軽度の疾患状態の下での天体細胞小器官またはタンパク質の運動性と局在性を調べるのに使用できます。タンパク質およびオルガネラ局在化の単一のスナップショットを提供するMHA固定製剤とは異なり、当社のライブイメージング技術は、個々の生きたアストロサイトにおける粒子ダイナミクスを分析するために使用することができます。トランスフェクションシードアストロサイトの前日は、トランスフェクション後24〜48時間でイメージングに適した密度で行う。リポフィクション試薬を適当な実験濃度に還元した血清培地で希釈する。250マイクロリットルの低血清培地で5マイクログラムの高純度DNAを希釈し、5マイクロリットルのリポフィケーションエンハンサー試薬をチューブに加えます。リポフィクション試薬を同量のDNAリポフィケーションエンハンサーミックスと混合し、室温で15分間インキュベートします。インキュベーションの終わりに、アストロサイト培養から上清を取り除き、細胞に滴下してトランスフェクションミックスの100マイクロリットルを加える。摂氏37度と5%の二酸化炭素で6時間後、トランスフェクション複合体を適切な量のアストロサイト培養培地に置き換え、細胞を細胞培養インキュベーターにさらに24〜72時間戻す。イメージングの30分前に、200マイクロリットルのアストロサイト培養培地で適切なリソソーム標識プローブを1マイクロモルの作業濃度に希釈し、37°Cで30分間プローブでアストロサイトに標識します。次いで、細胞を温熱アストロサイト培地で1回洗浄し、その洗浄をイメージング培地に置き換えます。プローブ標識の直後に、アストロサイト培養容器を顕微鏡ステージ上の適切なアダプターに入れ、EBI蛍光灯を用いて、蛍光タンパク質またはプローブを発現する細胞を見つける。デジタルカメラを使用して蛍光サンプルの照明を調整し、選択した細胞を視覚化し、焦点を調整して単一のセルにズームインします。次に、ズームと明確なフォーカス機能を使用して、300 ~ 500 秒の時間間隔で 2 秒ごとに 1 フレームの周波数で単一の Z スタック タイムラプス シリーズを取得します。タイムラプスイメージを AVI または TIFF スタックファイルとして保存してエクスポートします。タイムラプス画像を解析するには、ImageJ または Fiji でタイムラプス画像シーケンスを開き、スプリットチャンネルツールを使用してチャンネルを分割します。8 ビット緑のチャンネル画像では、セグメント線ツールを使用して、すべてのムービーに対して同じ方向規則を使用してパーティクルの軌跡に沿った線をトレースし、研究対象のすべてのセル内およびすべてのセル間で極性が一貫するようにします。ラインツールをダブルクリックして、線幅をトラックの太さに合わせて調整し、線幅10を使用してKymo ToolBoxプラグインの「Kymoを描く」マクロを実行します。時間と空間で画像のキャリブレーションを求めるプロンプトが表示されます。キャリブレーションが完了すると、カイモグラフが生成され、TIFFファイルとして保存できます。パーティクルの軌道を割り当てるには、[セグメント線]ツールを使用して、取得の全期間、カイモグラフ内の各パーティクルを手動で追跡し、ROI マネージャを使用して各パーティクル軌道を対象領域として記録します。さらに分析するために各タイムラプスビデオごとに関心のある領域のすべてを保存し、Kymoツールボックスプラグインの分析キモマクロを実行します。ウィンドウが開き、ドロップダウンメニューから目的のセルの核からパーティクルモーションの外側方向を定義します。制限速度は、対象となる貨物ごとにソフトウェアの感度に応じて定義する必要があり、ライン幅は各粒子軌道の厚さに合わせて調整する必要があります。[すべてのデータをログ記録] と [外挿されたデータのログ] 座標は、さまざまな貨物輸送パラメータの計算用にする必要があります。[はい] をクリックします。個々のトラックに対して計算されたデータは、キモグラフごとにプールされ、各画像に固有のテキストファイルに保存されます。シトシンアラビノシド処理と揺れベースの精製戦略を組み合わせることで、精製工程のみを含む従来のプロトコルに対するアストロサイト培養の純度が豊かになります。リポフェクションベースのトランスフェクションは、毒性を引き起こすことも、アストロサイトの生存可能性に影響を与えることなく、生細胞イメージングに最適なレベルでタンパク質の一過性発現を可能にします。同様に、蛍光プローブを使用することで、オルガネラダイナミクスやアストロサイトの追跡に対して、酸性の内皮小器官の迅速かつ効率的な標識が可能になります。画像から経過したデータを使用して、時間と空間で貨物の動きを追跡するカイモグラフを生成できます。これらのキモグラフでは、示された貨物の前向き運動は負の斜面を持つ軌道で表され、逆行運動は正の斜面を持つ軌道で表される。静止した小胞は垂直軌道として現れる。この分析では、アストロサイトの領域を通る特定の貨物のフラックスの定量化は、映画が取得されたアストロサイトの領域における通常の基線運動性を代表する可能性が高い貨物間のモーダル粒子の割合の差を明らかにした。キモグラフから得られた各粒子のフルタイムスケールに沿ったX、Y位置の正確なマッピングは、貨物速度や走行長さなどの他の運動パラメータを評価するためにも使用できます。このプロトコルは、高品質のアストロサイト培養と効率的な貨物ラベリングを必要とするため、トランスフェクション試薬のインキュベーション時間と取得タイムラインは、目的のプラスミドまたは貨物ごとに調整する必要があります。このプロトコルは、細胞の損傷、毒性、病原性突然変異、シナプス活性、または細胞内または細胞外環境における変化に応答して、アストロサイトにおける輸送事象を特徴付けるために変更することができる。
