このプロトコルでは、安定した連続歩行ラットが運動学的分析の最も重要な要素である。トレッドミルの速度は20cm/sに設定されました。この歩行速度は、16のスペース制約なしにラットが動く場合、決して「高い」とは考えられない。それにもかかわらず、この速度は訓練されていないラットがトレッドミルを安定して歩くのには速すぎて、異常な歩行と不均一な動きをも?...

坐骨神経機能性指数(SFI)は、機能的な坐骨神経評価を行うためのベンチマーク法である1。SFIは広く採用されており、ラット坐骨神経損傷に関する様々な機能評価研究で頻繁に使用されています2,3,4,5,6.その人気にもかかわらず、SFIには、オートマチネーション7、共同拘縮リスク、および足跡8の塗抹など、いくつかの問題があります。これらの問題は、その予後の値に深刻な影響を与えます9.したがって、SFIの代用として、代わりのエラーが発生しやすい方法が必要です。
そのような代替方法の1つは、運動学的分析である。これには、骨のランドマークやジョイントに取り付けられたトラッキングマーカーを使用した包括的な歩行分析が含まれます。キネマティック解析は、機能評価9にますます使用されています。この方法は、SFI11,12に起因する欠点を持たない機能評価10のための信頼性の高い敏感なツールとして徐々に認識されている。
このプロトコルでは、トレッドミル、4つの120 Hz充電式結合デバイス(CCD)カメラ、およびデータ処理ソフトウェアからなる3Dモーションキャプチャ装置を使用する一連の運動学的解析について説明します(資料表を参照)。この運動学的分析方法は、一般的なビデオ歩行または歩行分析13、14とは異なります。2 台のカメラは、1 つの側面から後肢の動きを記録するために、異なる方向に配置されます。続いて、後肢の3Dデジタルモデルがコンピュータグラフィックス9を使用して構築される。実際の四肢の寸法を厳密に再現することで、股関節、膝関節、足首関節、つま先関節などの指定関節角度を計算できます。また、ストライド/ステップ長や、スイング相に対するスタンス相の比率など、さまざまなパラメータを決定できます。これらの再構成は、2組のカメラによって送信されたデータから生成された後肢の完全に再構築された3Dデジタルモデルに基づいています。想像上の重心(CoG)軌道でさえ、自動的に計算することができます。
この3Dモーションキャプチャ装置を使用して、ラット坐骨神経クラッシュ傷害モデルの文脈における時間の経過に伴う機能的変化を明らかにする複数の運動学的パラメータを導入し、評価しました。
