緑膿菌は、ヒト、特に免疫不全において生命を脅かす疾患を引き起こす可能性のある日和見細菌病原体である。P.エルギノーサの病原性は、プロテアーゼおよびリポ多糖を含む多くの毒性因子の発現、ならびに保護バイオフィルム1を形成する能力に起因する。病原性因子を産生し、ヒトに疾患を引き起こす能力のために、P.アルギノーサを使用して市販品を安全上の懸念を提示する。大腸菌の非病原性株は、伝統的に人間の使用のためのバイオエンジニアリング医療および商業製品に使用されてきました。しかし、大腸菌が作るのが難しい製品もあれば、封入体に包装されている製品も多く、抽出に手間がかかるものもあります。特定の製品を作り、分泌する能力を持つ操作された細菌株は、分泌が収率を増加させ、精製プロセスを容易にする可能性が高いので、非常に望ましいです。したがって、細菌の他の種の非病原性株(例えば、より多くの分泌経路を利用する種)は、大腸菌に有用な代替手段を提供し得る。我々は最近、生物の病原性及び毒性が高く減衰するP.アルギノーサ、PGN5の株の開発を報告した2.重要なことに、この株はまだ多糖アルギン酸塩を大量に生成し、P.エルギノーサバイオフィルムの商業的に興味深い成分である。
PGN5株は、pEX100T-NotIプラスミドとの2段階のアレル交換手順を用いて生成され、生物の病原性に寄与することが知られている5つの遺伝子(toxA、plcH、phzM、wapR、aroA)を順次削除した。 pEX100T-NotIは、ハーバート・シュバイツァーの研究室3、4で開発されたプラスミドpEX100Tの多重クローニング部位内のNotI制限酵素認識部位にSmaIを変更することによって生成された。制限酵素NotIの認識部位は、SmaIに比べて稀なDNA配列であり、クローン化される配列中に存在する可能性が低いため、クローニングの目的でより便利である。プラスミドは、β-ラクタマーゼをコードし、カルベニシリンに対する耐性を付与するブラ遺伝子や、スクロースに対する感受性を付与するB.サチリサッカB遺伝子を含む、選択を可能にする遺伝子を運ぶ(図1A)。プラスミドはまた、大腸菌と互換性のある複製(ori)の起源、および結合を介して大腸菌からシュードモナス種へのプラスミド移動を可能にする転写(oriT)の起源を運ぶ。しかし、プラスミドはシュードモナスと互換性のある複製の起源を欠いているため、シュードモナス種内で複製することはできません(すなわち、シュードモナス自殺ベクターです)。これらの特性は、シュードモナス染色体からの遺伝的欠失を標的とするのにpEX100T-NotIを理想的にする。プラスミドクローニング工程は大腸菌を用いて行われ、得られたプラスミドは形質転換または結合によってシュードモナスに移される。そして、相同組換えイベントおよび選択的ステップを介して、標的化されたフレーム内欠失が生成され、マーカーフリーである。P.アルギノーサの染色体からゲノム領域を順次削除するこの方法は、PGN5のような高度に減衰したシュードモナス株を生成するために、または他の特定の用途のための株を設計するために使用することができる(例えば、プラスミド伝播のためのエンドヌクレアーゼの株欠乏またはプロテアーゼの株欠乏)。
細菌株の全体的な毒性は、突然変異が頻繁に起こる増殖条件および相の影響を受ける。したがって、遺伝子組み換え株の安全性を測定することは困難な場合があります。全身性毒性に対する細菌分離株を評価するために、C57BL/6マウス5の腹腔内注射による感染の以前に公表されたプロトコルを適応させた。注射に冷凍細菌ストックを使用するようにこの手順を変更し、正確な検査と使用する株の簡単な検証を可能にしました。このモデルでは、バイオ医薬品の製造に対してFDA承認されている大腸菌株BL21を、株6、7、8の相対病因を決定するための制御安全基準として用いた。この方法を使用する主な利点は、感染株が感染の前後の両方の細菌細胞数、表現型、および遺伝マーカーについて検証されるため、再現性があり、変動の原因を最小限に抑えるということです。これらの制御されたステップによって、必要な動物の数は減少する。このモデルでは、P.アルギノーサ株は、腹腔内注射時にC57BL/6マウス死亡率が大腸菌BL21以下をもたらすと考えられ得る。この単純なマウスモデルの感染は、FDA承認大腸菌株を基準として、他の種由来の遺伝子操作株の減衰病原性を評価するためにも使用され得る。ステップ1~7は、P.アルギノーサにおける逐次ゲノム欠失の生成を詳細に説明し(図1)、ステップ8~12は、P.アルギノーサ株の病原性を試験するためのマウスモデルの使用について詳述する。
