Hovedfokus i vores undersøgelse var at undersøge respiratoriske virkninger af Leptin signalering i CB. Flere protokoller er blevet udviklet til at vurdere rollen af leptin i en mekanisk måde. En fedtcellerne produceret hormon Leptin handlinger i hypothalamus at undertrykke fødeindtagelse og øge stofskiftet. Undersøgelser udført i vores laboratorium1,2 og af andre investigatorer3,4 viste, at Leptin øger hypercapnic ventilatorisk respons (HVR) forebygge fedme hypoventilation i leptin mangelfuld fedme. Men, et flertal af overvægtige personer har høj plasma leptin niveauer og demonstrere resistens over for de metaboliske og respiratoriske virkninger af hormonet5,6,7,8. Resistens over for Leptin er multifaktoriel, men begrænset permeabilitet af blod-hjerne barrieren (BBB) til Leptin spiller en stor rolle. Vi foreslår, at Leptin virker under BBB i et nøgle organ af perifer hypoxisk følsomhed, carotis organer (CB), for at forsvare vejrtrækning i overvægtige individer. CBs udtrykker den lange funktionelle isoform af Leptin receptor, leprb, men cb's rolle i respiratoriske virkninger af Leptin er ikke tilstrækkeligt belyst9,10.
Målet med vores metode var at undersøge effekten af Leptin signalering i CB på HVR. Vores rationale var at udføre (a) tab af funktions eksperimenter ved infusion af leptin i mus med intakte carotis-kroppe og denerverede carotis-kroppe efterfulgt af HVR-målinger; b) øgning af funktions eksperimenter i DB/DB -mus, der mangler leprb, hvor vi målte HVR ved baseline og efter ekspression af leprb udelukkende i CB. Fordelen ved vores teknikker var, at vi udførte alle vores eksperimenter i uhæmmet ubedøvet mus under søvn og vågenhed. Tidligere investigatorer enten udført deres eksperimenter under anæstesi9 eller ikke måle virkningerne af Leptin under søvn10. Desuden er vores undersøgelse den første til at udnytte en unik gevinst ved funktions tilgang med selektiv LepRb -ekspression i CB beskrevet ovenfor.
I den brede sammenhæng kan vores tilgang generaliseret til andre receptorer udtrykt i CB og deres rolle i hypoxisk følsomhed. Investigatorerne kan indgyde en ligand til en receptor af interesse og måle HVR ved baseline og efter CB denervation. Som en supplerende tilgang kan en receptor af interesse over udtrykkes i CB-og HVR-målinger kan udføres før og efter overekspression ved hjælp af vores teknologi, der er beskrevet i dette manuskript.

En adipocyte-producerede hormon Leptin er en potent respiratorisk stimulerende, som kan spille en vigtig rolle i forsvaret respiratorisk funktion i fedme. Kontinuerlig infusion af Leptin øgede HVR signifikant i Lean C57BL/6J-mus fra 0,23 til 0,31 mL/min/g/ΔFiO2 (P < 0,001, figur 2)11.
