De belangrijkste focus van onze studie was het onderzoeken van respiratoire effecten van leptine signalering in de CB. Verschillende protocollen zijn ontwikkeld om de rol van leptine op een mechanistische manier te beoordelen. Eerste, specifieke bijdrage van CB aan de HVR werd geanalyseerd door zorgvuldige kwantificering van de HVR tijdens de eerste 2 min van hypoxische blootstelling. Ten tweede, de relevantie van CB in leptine-gemedieerde up-regulering van de controle van de ademhaling werd onderzocht door twee compleme...

Een adipocyte geproduceerd hormoon leptine handelt in de hypothalamus te onderdrukken van voedselinname en verhoging van de stofwisseling. Studies uitgevoerd in ons laboratorium1,2 en door andere onderzoekers3,4 toonde aan dat leptine verhoogt de hypercapnic ventilatoire respons (HVR) voorkomen van obesitas hypoventilatie in leptine gebrekkige obesitas. Echter, een meerderheid van zwaarlijvige individuen hebben hoge plasma leptine niveaus en demonstreren weerstand tegen de metabole en respiratoire effecten van het hormoon5,6,7,8. Resistentie tegen leptine is multifactoriële, maar beperkte permeabiliteit van de bloed-hersen barrière (BBB) aan leptine speelt een belangrijke rol. Wij stellen voor dat leptine handelt onder BBB in een belangrijk orgaan van perifere hypoxische gevoeligheid, de carotis lichamen (CB), om de ademhaling in zwaarlijvige individuen te verdedigen. CBs Express de lange functionele isovorm van leptine receptor, leprb, maar de rol van CB in de Ademhalings effecten van leptine is niet voldoende opgehelderd9,10.
Het doel van onze methode was om het effect van leptine signalering in de CB op HVR te onderzoeken. Onze redenering was het uitvoeren van (a) verlies van functie-experimenten die leptine in muizen met intact carotis lichamen en denervated carotis lichamen gevolgd door HVR metingen; b) winst van functie-experimenten in db/DB -Muizen zonder leprb, waarin we de HVR op baseline en na uitdrukking van LEPRb uitsluitend in CB hebben gemeten. Het voordeel van onze technieken was dat we al onze experimenten uitgevoerd in ongefixeerde unanesthetized muizen tijdens de slaap en wakkerheid. Vorige onderzoekers ofwel uitgevoerd hun experimenten onder anesthesie9 of niet meten van de effecten van leptine tijdens de slaap10. Daarnaast is onze studie de eerste die gebruik maakt van een unieke winst van functie benadering met selectieve LepRb -expressie in CB zoals hierboven beschreven.
In de brede context, onze aanpak kan worden gegeneraliseerd naar andere receptoren uitgedrukt in CB en hun rol in hypoxische gevoeligheid. Onderzoekers kunnen een ligand aan een receptor van belang en meet de HVR op baseline en na CB denervation. Als een complementaire aanpak kan een receptor van belang worden overuitgedrukt in CB en HVR metingen kunnen worden uitgevoerd voor en na overexpressie met behulp van onze technologie beschreven in dit manuscript.

Een adipocyte-geproduceerde hormoon leptine is een krachtige respiratoire stimulerende middelen, die een belangrijke rol kunnen spelen bij het verdedigen van de ademhalingsfunctie bij obesitas. De carotis lichamen (CB), een belangrijk orgaan van perifere hypoxische gevoeligheid, uitdrukken de lange functionele isovorm van leptine receptor (leprb) maar de rol van leptine signalering in de controle van de ademhaling is niet volledig opgehelderd. We onderzochten de hypoxische ventilatorrespons (HVR) (1) in C57BL/6J-muizen voor en na leptine-infusie bij baseline en na de denervatie van CB; (2) in Leprb-deficiënte zwaarlijvige db/DB muizen bij baseline en na Leprb overexpressie in CBs. In C57BL/6J muizen, leptine verhoogd HVR en effecten van leptine op HVR werden afgeschaft door CB denervation. In db/DB -muizen verhoogde de leprb -expressie in CB de HVR. Daarom concluderen we dat leptine in CB werkt om reacties op hypoxie te vergroten.

Continue infusie van leptine significant toegenomen HVR in Lean C57BL/6J muizen van 0,23 tot 0,31 mL/min/g/ΔFiO2 (P < 0,001, Figuur 2)11. CSND afgeschaft de leptine-geïnduceerde toename in HVR (Figuur 2), terwijl geen verzachtende effecten van csnd op HVR werden waargenomen in Sham chirurgie groep na leptine infusie.

Ons protocol kan worden gebruikt om de ademhalingseffecten van leptine op het halsslagaderlichaam in de perifere chemoreflex ademhalingscontrole en fysiologische reacties op hypoxie te onderzoeken. We kunnen ook de effecten van verlies en winst van halsslagader lichaam functie met behulp van chirurgische en chemische denervation en adenovirus transfectie respectievelijk. Voor het begin van de procedure, meet de temperatuur in een op maat gemaakte hele lichaam plasthemagrophy kamer, en meet de relatieve vochtigheid van het lab.Meet vervolgens het lichaamsgewicht en de rectale temperatuur van de experimentele muis. Plaats de muis in de kamer en plaats een oximeter kraag rond de geschoren hals van het dier. Overwegende dat na transfectie met controle LacZ adenovirus, geen verandering wordt waargenomen.Deze technieken maken analyse van de rol van halsslagader lichamen in de perifere chemoreflex en de effecten van leptine op halsslagader lichamen in de controle van ademhalingen.
