科利耳表面制剂允许使用免疫histo化学和共生成像对科蒂器官的整个透镜进行可视化。它已被广泛用于调查感兴趣的特定耳蜗病理学。我们将修改耳蜗微切除方法。该技术的主要优点是将一块耳蜗上皮粘附到 10 毫米圆形盖玻片，用于免疫标记程序，同时避免在多个洗涤步骤期间组织丢失。此外，耳蜗病理学，耳蜗表面制剂也可用于评估表达和头发细胞再生。这种技术需要基本的显微镜技能，该方法的视觉演示有助于确保每个步骤都正确执行。演示这个程序的将是我实验室的研究生方乔军。对于时间骨提取，立即验尸后拉皮前露出头骨骨，并使用剪刀沿头骨中心线从后部向前切割骨骼。在使用拇指和食指手动取出三个月或三个月大的小鼠的时骨之前，使用钳子切除脑组织。将骨骼放入直径为 30 毫米的培养皿中，在解剖显微镜下放入含有新鲜冰冷 4%PFA 和 PBS 的培养皿中，然后从椭圆形窗口取出石块。用五号钳刺穿圆形窗膜，并使用 27 表针连接到含有新鲜 4%PFA 的 1 毫升注射器，使小孔进入耳蜗的顶点。通过圆形和椭圆形的窗户，轻轻缓慢地用固定溶液将耳蜗固定化，直到溶液从顶点的小孔中洗出。然后将多达两个带水耳蜗转移到单个20毫升闪烁小瓶中，每瓶含有10毫升4%PFA的光小瓶，并在室温下轻轻搅拌样品两个小时，然后在旋转器上隔夜储存在4摄氏度的温度下。第二天早上，用三个五分钟的洗涤用新鲜的PBS洗耳蜗。上次洗涤后，在每瓶中加入20毫升4%EDTA，并在4摄氏度下以温和搅拌旋转样品48小时。在孵育结束时，用钳子触摸每个耳蜗的骨面庭部分，以评估样品的弹性。如果骨骼是弹性的而不是坚定的，耳蜗已经脱钙。用新鲜的 PBS 更换每个样品的 EDTA。对于耳蜗上皮的微解，用钳子抓住时间骨的前庭部分，用手术刀以 45 度角切割角。沿着圆形窗口和椭圆形窗口之间的模糊线垂直切割，将耳蜗与前庭部分分离，并定向耳蜗部分，基底转向底部，中间转向 Petri 盘的顶部。从这个位置，切割骨胶囊和中间的横向壁转向从其中去除的面体部分，并继续切割，以完全分离中间部分从基底和钩区域。将基底和钩部分与罗勒膜位点朝向菜底，垂直切开钩区域的介质，以去除介质，并切割基底和钩区域以分离钩部分。为避免组织变形，在分离基底转弯和钩区域之前，将介质从钩区域切开。切开骨胶囊和中间转弯的侧壁组织相对较大的部分，用钳子握住侧壁，使骨胶囊和侧壁与培养皿底部对齐，从罗勒膜侧切下这些组织。接下来，将试样平展，使感觉性毛细胞表面朝上，修剪掉骨胶囊和侧壁的其余部分。然后使用钳子去除电膜，以完全分离中间区域，并删除骨胶囊，横向壁组织和剩余转弯的电体区域，正如刚刚演示的。当所有耳蜗转动被解剖后，将0.5微升的细胞和组织粘合剂扩散到一轮10毫米盖玻片的中心，让胶粘剂在室温下干燥3至5分钟。将干燥的盖玻片放入培养皿中，将每个感觉上皮样品的所有四块粘在一个盖玻片上。然后使用钳子抓住每个盖玻片的一个边缘，将标本转移到四孔细胞培养皿的单个孔中，用于免疫标记。对于表面人工耳蜗突触制剂的免疫标记，每次洗涤用新鲜 PBS 清洗每个感觉上皮三次，每次洗涤五分钟，然后用两毫升2%非离子表面活性剂在旋转器上用两毫升非离子表面活性剂洗涤30分钟。在孵化结束时，从每孔中吸出表面活性剂溶液，并阻止任何非特异性结合，每孔有100微升的阻滞溶液，在旋转器上用温和搅拌在室温下一小时。在阻断孵育结束时，用PBS清洗样品三次，如在温和搅拌下证明，然后用100微升的原抗体标记每个上皮，在37摄氏度的高温下24小时。第二天，用新鲜的PBS和每次洗涤的温和搅拌洗涤样品三次，并在37摄氏度的温度下用100微升的有关二次抗体标记样品两小时。在孵化结束时，每次洗涤用三个五分钟的新鲜 PBS 洗涤样品，然后将每个盖玻片放在单独的玻璃显微镜幻灯片上，样品朝上。接下来，小心地将8个适当安装介质的微升添加到每个盖玻片的中心，并使用钳子将另外10毫米盖玻片安装到每个样品上。然后用清明的指甲油密封每个盖玻片三明治的两侧，将样品放入纸板幻灯片夹中，并在四摄氏度处存放幻灯片。虽然对成年小鼠耳蜗进行表面准备的解剖并不简单，但新接触该技术的研究人员在练习10到15只耳朵后应该能够学习出这种方法。CTBP2和GluA2的未治疗的10至12周大CBAJ小鼠表面制剂的免疫标记表明，两个前突触带和后突错端子分别位于内毛细胞核下方，并并列指示功能突触。对 myosin 7A 进行免疫标记，用 phaloidin 和 DAPI 进行反染，揭示了感觉性毛细胞的存在，包括外发细胞和内毛细胞及其核。无论使用细胞和组织粘合剂，对米奥辛7A的免疫标记和用 phaloidin 进行反污表明免疫反应性或均匀性没有差异。此外，扫描电子显微镜的表面制剂从未经处理的6至8周大的C57黑色6小鼠显示一个组织良好的V形外毛细胞在三行样本。请记住，在圆窗和椭圆形的窗户中轻轻、缓慢地填充人工耳蜗，直到溶液从顶点的小孔中洗出。在将钩区域与基底转分离之前，注意将介质从钩区域切断，以方便去除介质。
