人工内臓表面製剤は、免疫組織化学および共焦点イメージングを用いてコルティの器官の全レンズを可視化することを可能にする。それは関心のある特定の人工内科の病理学の調査のために広く使用されている。人工内流微細解剖法を変更します。この技術の主な利点は、複数の洗浄工程の間に組織の損失を回避しながら、免疫標識手順のための10ミリメートルのラウンドカバーリップに人工内皮上皮の一片の付着です。また、人工内毛病変に加えて、人工内毛表面製剤は、評価発現および毛髪細胞再生のために利用することもできる。この手法には基本的な顕微鏡のスキルが必要であり、この方法の視覚的なデモンストレーションは、各ステップが正しく実行されることを確実にするのに役立ちます。この手順のデモンストレーションは、私の研究室の大学院生であるチャオジュン・ファンです。側頭骨抽出の場合、直ちに死後、皮膚を前方に引っ張って頭蓋骨を露出させ、はさみを使用して頭蓋骨の中心線に沿って後部の側面から骨を切断する。鉗子を使用して脳組織を取り除き、親指と人差し指を使用して3ヶ月以上のマウスから側頭骨を手動で取り除きます。骨を直径30ミリのペトリ皿に入れ、新鮮な氷冷4%PFAとPBSを含む分割顕微鏡で、楕円形の窓からテープを取り除きます。5番鉗子で丸い窓膜を穿刺し、新鮮な4%PFAを含む1ミリリットルの注射器に取り付けられた27ゲージ針を使用して、小さな穴を小さな穴を作ります。円と楕円形の窓を介して固定液を使用して、角の角の小さな穴から溶液が開くまで、静かにゆっくりとコクレアを浸透させます。その後、バイアルあたり4%PFAの10ミリリットルを含む個々の20ミリリットルのシンチレーションバイアルに最大2つの浸透した内欠性バイアルに移し、室温で2時間サンプルを穏やかに攪拌し、続いてローテーターで摂氏4度で一晩貯蔵します。翌朝、1回の新鮮なPBSで5分間の洗浄を3回でこする。最後の洗浄後、各バイアルに4%EDTAの20ミリリットルを加え、穏やかな攪拌で摂氏4度で48時間サンプルを回転させます。インキュベーションの終わりに、各コクリーの骨前庭部分を鉗子で触れてサンプルの弾力性を評価します。骨がしっかりしているのではなく弾性である場合、内痛は脱灰されています。各サンプルの EDTA を新しい PBS に置き換えます。人工内耳上皮の微細解剖の場合は、側頭骨の前庭部分を鉗子でつかみ、メスを使って頭頂部の旋回を45度の角度で切ります。丸い窓と楕円形の窓の間のかすかな線に沿って垂直に切り取り、内臓を前庭部分から分離し、底面の旋回で人工内膜部分をペトリ皿の上方に向けます。この位置から、尖語部が取り除かれた端に向かって中旋回の骨膜と側面壁を切断し、中間部分を基底領域とフック領域から完全に分離するために切断を続ける。皿の底部に向いたバジラー膜部位と基底部とフック部を配置し、フック領域の内側を垂直に切断して内側を取り除き、基底部とフック領域を切断してフック部を分離する。組織の歪みを避けるために、基底旋とフック領域の分離前に、フック領域の内側を切り取ります。中回転の骨膜と側壁組織の比較的大きな部分を切り取り、側壁を鉗子で保持して、骨状のカプセルと側面壁をペトリ皿の底に合わせ、これらの組織をバジラー膜側から切断する。次に、試料を平らにして、感覚性のヘアセル表面を上に向け、骨状のカプセルと側面壁の残りの部分をトリミングします。次に、鉗子を使用してテクトリアル膜を除去し、真ん中の領域を完全に分離し、骨膜、側面壁組織、および残りのターンのテクトリアル領域を取り除きます。すべての人工内壁の回転が解剖されたら、1ラウンド10ミリメートルのカバースリップの中央に0.5マイクロリットルの細胞と組織接着剤を広げ、接着剤を室温で3〜5分間乾燥させます。乾燥したカバーリップをペトリ皿に入れ、各感覚上皮サンプルの4つすべてを1つのカバースリップに貼り付けます。次に、鉗子を使用して各カバースリップの1つの端をつかんで、検査体を免疫標識用の4ウェル細胞培養皿の個々の井戸に移します。表面人工内膜シナプス製剤の免疫標識の場合、各感覚上皮を1回の新鮮なPBSで5分間3回洗浄し、続いて各サンプルを2ミリリットルの非イオン性界面活性剤を2%の非イオン性界面活性剤で処理し、ローテーター上の室温で30分間処理します。インキュベーションの終わりに、各ウェルから界面活性剤溶液を吸引し、室温で穏やかな攪拌を伴う回転子上で1時間、1ウェルあたり100マイクロリットルのブロッキング溶液で任意の非特異的結合を遮断する。ブロッキングインキュベーションの最後に、穏やかな攪拌の下で示されるようにPBSでサンプルを3回洗浄してから、各上皮に100マイクロリットルの目的の一次抗体を24時間、光から保護して24時間標識します。翌日、新鮮なPBSと1回の穏やかな攪拌でサンプルを3回洗浄し、適切な二次抗体の100マイクロリットルを光から保護した摂氏37度で2時間ラベル付けします。インキュベーションの最後に、各カバースリップを個々のガラス顕微鏡スライドに置く前に、洗浄ごとに新鮮なPBSで3回の5分間の洗浄でサンプルを洗います。次に、適切な取り付け媒体の8マイクロリットルを各カバースリップの中央に慎重に追加し、鉗子を使用して各サンプルに別の10ミリメートルのカバースリップを取り付けます。次に、各カバースリップサンドイッチの側面を透明なマニキュアで密封し、サンプルを段ボールスライドフォルダに入れ、スライドを摂氏4度で保存します。表面製剤のための成体マウスのコクレアの解剖は簡単ではありませんが、この技術に新しい研究者は、10〜15の耳で練習した後に方法を学ぶことができます。CTBP2およびGluA2を有する未治療の10〜12週齢のCBAJマウスの表面製剤の免疫標識は、シナプス前リボンおよび心内末端の両方がそれぞれ内側の毛細胞核の下に位置し、機能的シナプスを示す並置されていることを示している。ミオシン7Aの免疫標識とファロイジンおよびDAPIによる対抗は、外毛細胞および内毛細胞およびそれらの核を含む感覚性毛細胞の存在を明らかにする。ミオシン7Aの免疫標識およびファロイジンによる対抗染色は、細胞および組織接着剤の使用の有無にかかわらず、免疫反応性または均一性に差を示さない。さらに、未処理の6~8週齢のC57黒6マウスの表面製剤の走査型電子顕微鏡は、外毛細胞のV字型立体体をサンプルの3列に整理した。溶液が頂点の小さな穴から開くまで、円形と楕円形の窓を通して静かにゆっくりと固定液で人工内緒を埋めることを忘れないでください。フック領域を基底方向から分離する前に、内側の内側領域を切り離して、内側の除去を容易にするように注意してください。
