この方法の潜在的な制限は、非変換された根の残留成長に基づいて行われます。これは、使用されるプラスミドが変換された根の選択を可能にするレポーター遺伝子を欠いている場合に特に重要である。この問題を解決するために、我々は、健康な抗生物質耐性形質転換根の成長を可能にしながら、非形質化された根の成長を阻害する抗生物質選択に基づく代替方法を開発した。A.根生は芽の変態を誘発しないので、抗生物質の影響を受けやすく、したがって、抗生物質含有培地から分離しておくべきである。

このプロトコルは、科学者がトマトの細菌性萎れ性疾患の簡単な遺伝子解析を行うことを可能にするので、重要です。この技術の主な利点は、遺伝子発現の操作とその後のアッセイが短時間で、設備や植物の成長空間の小さな要件で行うことができるということです。これは、非常に汎用性の高い方法であり、病原体接種および他の生理学的アッセイの両方を行うために他の作物植物に適用され得る。インビトロで苗の操作中に無菌性を維持するためには、それらがフローフードの外にあるときにプレートを閉じておくことを重要です。これは簡単な手法ですが、操作には複数の手順が必要です。この方法の視覚化は、原稿で説明するのが難しい小さなトリックを見るのに役立ち、他の研究者が研究室でこの方法を実装するのに役立ちます。この手順を実証するのを手伝うのは、大学院生のアーヘン・ジャオです。トマトの種を殺菌して洗った後、スクロースを使わずに半強度の村重・スクーグ培地に移します。3日間摂氏25度から28度の温度で暗闇の中で種子を保ちます。半強度のムラシゲとスクーグ培地を含む9平方センチメートルのペトリ皿の中にオートクレーブされた8.5平方センチメートルのフィルターを置き、各プレートに6つの発芽トマトシードを置きます。プレートをマイクロポアテープで密封し、発芽した種子を摂氏25~28度で3~4日間インキュベートします。植物の形質転換前に28°Cで2日間適切な抗生物質を用いて固形LB培地でアグロバクテリウム根虫MSU440を成長させます。無菌メスを使用して、トマト苗のラジクルと低血糖の底を切断します。プラスチックチップまたはメスブレードを使用して、LB培地の表面からA.rhizogenesバイオマスを収穫し、カットトマトの苗を細菌バイオマスに慎重に浸します。この後、トマトの苗を2センチメートル/4センチメートルの半円形のフィルターペーパーで覆い、高湿度を維持し、生存と新しい根の開発を容易にします。蒸散が可能な環境で、高湿度で苗を保つことが重要です。これを行うには、ろ紙で苗を覆い、マイクロポアテープでプレートを閉じます。25〜28°Cの温度で成長室に6〜7日間変換されたトマトの苗を保存します。その後、新しい新興毛深い根をカットするために無菌メスを使用し、苗が新しい毛深い根を生成できるようにします。新しい毛深い根の第二世代が現れたら、苗の上にフィルターペーパーを取り除き、再びマイクロポアテープでプレートを密封します。DsRed蛍光または植物インビボイメージング用の他の機器を可視化するには、赤い蛍光によって識別することができる正の形質転換された根をマークします。メスを使用して、赤い蛍光の欠如によって識別できる陰性非形質の根を除去する。赤い蛍光を示す苗を半強度のムラシゲとスクーグ培地を含む新しいプレートに移し、変換された根を主根として開発しやすくする。同じプレートに赤色蛍光を示さない苗を保管し、後の時点で蛍光根の出現を確認します。苗を2センチメートル/4センチメートルの半円のろ紙で覆い、プレートを密封し、苗をインキュベートして新しい毛深い根を開発できるようにします。最初に接種ポットに水を浸し、余分な水を注ぎ、プラスチック製の植栽トレイに入れることで、根の表面が細菌の接種に曝される接種ポットを準備します。ピンセットを使用して、選択した苗を、形質転換した根を入れ替えた苗を接種ポットに移します。ラップまたは透明な蓋でトレイを覆い、湿度65%で摂氏25~28度に保ちます。5~6日後にカバーを取り外します。28°Cの軌道シェーカーで5つのLB液体媒体でラルストニアを成長させ、静止期まで200rpmで揺れる。600ナノメートルで光学密度を測定し、細菌数を決定します。1のOD600に水で細菌培養を希釈します。変身したトマト植物を含む16〜20個の接種ポットを接種トレイに入れます。その後、希釈した細菌接種物の300ミリリットルをトレイに注ぎ、植物が接種物に20分間浸漬できるようにします。この後、ポッティング土壌の層で新しいトレイを準備します。接種されたポットを新しいトレイに移動し、湿度75%、摂氏26~28度の温度、12時間の光と12時間の暗闇の光周期を備えた成長室にトレイを置きます。ここでは、トマトの根を変換し、細菌性萎れ病の研究のための簡単な遺伝子解析を行うためにラルストニアを接種する。病気の症状の発症は、空のベクターで形質転換された根を持つトマト植物と、トマトCESA6を標的とするRAI構造で形質転換されたもので追跡される。疾患指数データは、ゼロから4までの任意のスケールに従って同じ実験単位から時間の経過とともに収集され、ガウス分布に従わない、パラメトリックデータの標準的なテストの使用を除外する。標準的なアプローチとして、コントロールと感染曲線の両方を比較するUマンホイットニー、両側の非パラメトリックテストが使用されます。この分析では、両方の曲線の中央値の差は有意ではないようです。また、疾患進行曲線下の領域を定量化することも可能であり、疾患進行の複数の観測を単一の値に組み合わせることが可能である。病気進行曲線の下の領域は、感染プロセスの終わりにトマトCESA6 RNAi植物と比較して対照植物の方が高い値を示し、トマトCESA6沈黙植物は対照植物よりもラルストニア感染に対してより耐性があることを示している。信頼区間は、高い確率で、特定の変数の母集団値が見つかる値の範囲を推定する方法を提供します。ここで見られるように、制御および感染曲線の95%信頼区間の面積は、CESA6が沈黙したときの抵抗の高い確率を推定する。疾患指数値は、ゼロに対応する2未満の疾患指数と1つに対応する2つ以上の疾患指数を考慮して、バイナリデータに変換することができる。これは、ラルストニア接種後の生存曲線の表現を可能にする。対照と感染した植物の生存率の差は、ゲハン・ブレスロー・ウィルコクソン統計検定によると統計的に有意ではない。次に、CESA6の発現を接種工程の前にランダムに選択した2つの形質転換根で解析し、RALSTONIAに対する抵抗力の増強がCESA6の発現低下と相関することを示す。2回の選択の後、35~40%の変換速度を得ることができ、追加の選択ラウンドを行うことでこの値を増やすことができます。この手順を実行している間、高湿度を維持するために濾紙で覆われた苗を維持し、ガス交換を確実にするためにマイクロポアでプレートを密封することが重要です。根形転換後、根の生理学を観察したり、分子生物学、細胞生物学、生化学を用いて植物の反応を研究するために、多くの治療を応用することができます。主に、この技術は、限られたリソースを持つ研究者がトマトの根の細菌感染の遺伝子解析を行うことを可能にする。
