SNX-BAR 是膜重塑蛋白，在人类疾病中起着关键角色。使用我们的方法，我们可以确定其精确的生物物理特性，以促进脂质结合和膜重塑。从酵母细胞中纯化 SNB-BAR 蛋白，可以获取原生同源和异质 SNX-BAR 二丁体，同时最大限度地减少非原生表达系统经常遇到的毒性和溶解性。我们的方法可用于了解所有酵母 SNX-BAR 的脂质特异性，或从任何其他生物体中生成重组的 SNX-BAR 蛋白质。正确的挤出机组件对于防止挤出过程中脂体损失至关重要。对于初来的学生或那些新进入这个领域的学生来说，目视观察最关键的步骤将大大提高你成功的机会。首先将大量酵母细胞接种到烧瓶中，其体积至少是含有50毫升标准YP培养基的四倍，并辅以2%的拉芬糖和0.1%的葡萄糖作为碳源，并在30摄氏度的摇床中过夜生长。第二天早上，在一升标准YP培养基中接种50毫升预培养物，在2.8升体积中接种2%的拉芬糖和0.1%的葡萄糖，使Fernbach烧瓶在摇晃的培养箱中培养4到5个小时的培养。当600纳米的光学密度达到0.2时，将乳糖加入到细胞中，最终浓度为2%，在摇动的培养箱中进行过夜培养。第二天早上，通过离心收获细胞，将酵母颗粒转移到50毫升锥形管中。将颗粒重新悬浮在 15 毫升的纯化缓冲液中，以达到约 30 毫升的最终体积，并将细胞悬浮液加载到冷却至 4 摄氏度的均质器上。将样品以每平方英寸20至25，000磅的2至3轮，将落酸转移到冰上新的50毫升锥形管中。立即通过离心清除细胞，并小心地将上清液转移到新管中。接下来，平衡300微升IgG Sepharose珠子与三个再悬浮在纯化缓冲液中，然后将珠子加入清除细胞，在4摄氏度下旋转两小时孵育。在孵育结束时，将珠子添加到 10 毫升色谱柱中，让未绑定的乳酸盐流经。每次洗涤时用一毫升的洗涤缓冲液清洗珠子 10 次，允许每次洗涤完全流过，然后再添加下一个卷。最后一次洗涤后，使用移液器和新鲜缓冲液将珠子转移到微离心管中。用新鲜洗涤缓冲液将珠子的总量降至500微升，用每毫升10毫克的两微升将珠子从麦芽中去除，在4摄氏度的温度下旋转过夜孵育。第二天早上，使用27表针完全去除上清液，并评估蛋白质纯度10%聚丙烯酰胺SDS页。然后根据标准协议使用布拉德福德蛋白质测定来量化浓缩蛋白质，并在四摄氏度下储存蛋白质长达一周。为了准备脂质体，使用玻璃注射器将库存脂质转移到干净的玻璃培养管中，以实现1%磷脂酰胺-3-磷酸盐、20%ergosterol、30%磷脂酰胺、磷脂酰胆碱的最终脂质混合物，如表中所述。根据每个脂质重新排放的溶剂，混合物在添加每种脂质后可能会变得浑浊。通过循环运动将低流量氮气引导到混合物处，小心地干燥脂质混合物，以均匀处理玻璃管底部的脂质。然后用铝箔包裹玻璃培养管，使开口未覆盖，并在真空中进一步脱水脂质一小时。在孵化结束时，在每瓶中加入400微升的结合缓冲液，以最终脂质浓度2.5毫摩尔完全补充脂质，然后以中等速度在室温的涡流上重新补充脂质30分钟。当脂质重新暂停时，缓冲液应变为多云。将重新吸收的脂质体转移到微离心管中，将液氮中管冻结，随后在 37 摄氏度的水浴中解冻 7 至 8 次。在化学烟罩中，用每支注射器三次清洁两个一毫升玻璃注射器，以清除任何残留脂质，并平衡每个注射器与两卷超纯水和两卷结合缓冲液。根据制造商的建议组装微型挤出机。然后在结合缓冲液中将两块过滤器支撑淹没在一个200纳米膜中。组装迷你挤出机，使之完全密封，这一点很重要。检查泄漏的一个好方法就是通过设备传递缓冲区。将滤芯支架之间的膜夹入，将膜放入迷你挤出机中。使用一个平衡的一毫升注射器，通过迷你挤出机将一个与脂质体混合物体积相似的结合缓冲液通过迷你挤出器，并使用另一个注射器将脂质体混合物倒置微离质管，将最后一个脂质体收集到管盖中，供其收集。然后通过200纳米膜挤出脂质体19至21次，将挤出的脂质体收集到新的微离心管中。在进行 SNX-BAR 脂体结合和输卵管检测之前，通过超离心预清除纯化蛋白质，然后将上清液转移到新的微离心管中，而不会干扰颗粒。接下来，在20微升的总反应量中孵育4个纯化SNX4-ATG20和2.5毫摩尔的脂质体，在30摄氏度下孵育30分钟。要可视化和量化脂体管，在孵化结束时，立即将样品发现到碳涂层的铜网网格上，并用 1%的醋酸碱状染色。然后在200千伏的透射电子显微镜上分析样品。对于脂质体结合和沉积，将反应的20微升转移到聚碳酸酯离心管中，并在超离心机中用兼容的路由器旋转样品。在离心结束时，小心地将上流水根转移到新的微离心管中，并在 40 微升 SDS 页面样品缓冲液中重新填充颗粒。将颗粒悬浮液转移到新的微离心管中，并在上流液中加入 20 微升样品缓冲液。然后将等效的颗粒和上总比样品量加载到 10%聚丙烯酰胺 SDS 页面凝胶上，并执行 Coomassie 染色，以可视化与脂质体绑定的 SNX-BARs。为了检查 SNX-BAR 表达的正确诱导，可以使用串联亲和力纯化标签的西部印迹作为 SNX-BARs 的蛋白质水平，可能无法通过 Coomassie 污渍检测到。纯化 SNX-BAR 异质器后，两个 SNX-BAR 的波段应以一对一的计量比出现，并且应很少或没有污染波段。在这项具有代表性的分析中，SNX-BAR异质剂被表达、纯化，并如所证明的与合成脂质体有关。请注意，MVP1 形成同源体，并按预期在细菌中表达。SNX-BAR 与不同脂体组合物的结合可以通过密度测定来量化，以评估磷脂素对脂体结合的影响。电子显微镜成像允许测量SNX4-ATG20脂体管，大多数管状通常直径在14至26纳米之间。请注意，纯化的 SNX-BA 可以与脂质体上的货物捕获复合物重组，以了解全组件如何影响膜重塑。在比较纯化SNX-BAR二丁二剂与由不同脂质组成的合成脂质体的不同组合结合时，发现SNX-BAR蛋白具有不同的脂质结合偏好。处理氯仿时，始终在发动机罩中工作，在处理利器或玻璃材料时，请务必佩戴适当的 PPE。
