現代医学の風景は、遺伝子および細胞ベースの治療法(GCT)の最近の発展によって急速に変化しました。翻訳研究で最も急速に成長している分野の1つとして、GCTセクターはユニークで前例のない課題にも直面しています。堅牢な臨床成果に加えて、GCTの商業的成功には効率的で費用対効果の高い製造プロセスが不可欠であり、小規模製造業では特に達成が困難である1.時間、労力、品質保証のコストは、細胞の各バッチが数百または数千の代わりに1人の患者に対して数回の用量しか生成する場合に拡大されます。製造プロセスが抗体および組換えタンパク質の産生に近い同種細胞療法とは異なり、自家細胞療法は、典型的には小規模な操作1として産生される。バイオ医薬品製造における比較的新しい現象として2は、小規模な細胞処理の選択肢が非常に限られている。
細胞製造には緩衝液交換が不可欠です。これは、細胞が培養媒体から取り出され、凍結保存または注入のために濃縮される下流のプロセスの一つです。現在、小規模な細胞製造は、多くの場合、学術研究の設定と同様のプロセスを適用し、不妊3を維持するために専門のクリーンルームに依存しています。手動下流プロセスは、多くの場合、ボリューム削減とバッファー交換のために細胞をペレット化し、再サスペンドするためにベンチトップ遠心分離機を使用します。これらのオープンプロセスは高価であり(すなわち、労働およびクリーンルームのメンテナンス)、商業生産2、3に理想的ではない限られた製造能力を有する。

自動化を既存の製造プロセスに導入できるさまざまな戦略があります。これは、完全に統合されたプラットフォームまたはモジュラーベースの処理チェーンを実装することによって実現できます。CliniMACS Prodigy(ミルテニ・バイオテック)、コクーン(オクタンバイオテック)、量子(テルモBCT)など、自家用細胞製造に市販されている完全に統合されたプラットフォームがいくつかあります。「GMP-in-a-box」と呼ばれることが多いこれらの統合プラットフォームは、インフラストラクチャに対する要求が少なく、運用が容易です。ただし、完全に統合されたセットアップの製造能力は、システムに取り付けられたインキュベーターによって制限される場合があります。例えば、Prodigyの培養能力は、その400 mLチャンバー8に制限され、量子カートリッジは、2.1m2(120 T175フラスコに相当)7に設定された制限表面積を有し、より高い細胞用量9、10を必要とする患者には十分ではないかもしれない。さらに、ProdigyおよびQuantumは、その使用を制限する共通の属性を有する:操作単位は、細胞膨張期間を通じて細胞の単一のバッチによって占有され、したがって、各ユニット11によって製造することができるバッチの数を制限する。自動化へのモジュラーアプローチは、商業製造プロセス12、13をシミュレートする複数のモジュラーユニットを有する製造チェーンを作成することです。このアプローチは、培養装置と細胞洗浄装置を分離し、これにより製造効率を最大化することができる。理想的な処理装置は、製造ニーズ12に適応性と拡張性を備えたものであろう。
1970年代にさかのぼる逆流遠心分離(CFC)技術は、細胞処理14に長い歴史を持っています。遠心力と逆流力のバランスをとることで細胞濃度と分離を実現します。典型的には、細胞懸濁液は、遠心力を受けながら一定の流量下で細胞チャンバの狭い端から入る(図1A)。流体の流れは遠心力とは逆の方向に発揮される。これは、細胞チャンバ内に勾配を形成する逆流力と呼ばれる。逆流力は、細胞チャンバーが円錐形の細胞チャンバの先端から広がるにつれて減少します。より高密度で大きな直径を持つ細胞は、より高い沈下速度を有し、したがって、それらは円錐形の細胞チャンバーの先端に向かって力平衡に達する。より小さい粒子は、チャンバーの基部に向かって平衡に達するか、またはチャンバーに保持するには小さすぎて洗い流される。CFC技術は、樹状細胞療法のための単球を単球を分離するなどの血液アペレーシス製品の処理における応用で主に知られている15、16。緩衝液交換の面では、CFC技術は大規模な製造17年にのみ適用され、自家細胞療法の小規模な製造にはまだ使用されていません。
小規模なセル製造に適した装置の必要性に対処するために、自動化されたCFC装置(材料表参照)が最近開発された。自動セル処理装置は、逆流遠心分離技術を使用して細胞破片を除去し、バッファ交換を容易にします。この装置は細胞移動袋に無菌接続することができる単一使用キットとの緩衝交換を行い、細胞は無菌の封入されたシステムの中で処理することを可能にする。ここでは、自動プロトコルで哺乳動物細胞培養物の緩衝交換を行うカウンターフロー遠心装置の使用を検討する。本研究では、ユルカト細胞と間葉系間質細胞(MSC)を用いて緩衝交換プロトコルを試験し、それぞれ非付着細胞型と付着細胞型をモデル化した。ユルカート細胞は、急性T細胞白血病19、20の研究にしばしば使用される不死化T細胞である。MSCは、広範囲の疾患9に対するヒト臨床試験で研究されている成体幹細胞である。
