1. 参与者招聘

</ol>2. 创建外围轨道系统 （图 3）

</ol>3. 使用异色闪烁光度计进行测量
注： 此步骤用于其他数据收集，对于使用反射计的外围测量不是必须的。

	<li>向学员解释该过程。</li>

</ol>6. 分析 （图 4）

我们的研究说明了使用反射计装置在各种波叶和对向区域进行体内MPOD测量的技术和方法。我们开发和校准了外围轨道系统，以便从中央固定处获得 1 度和 2 度的测量。研究结果表明，MPOD、L-OD和Z-OD可在光学无穷大下利用该协议在各种对叶和对向区域进行测量。当诊所没有长房间时，该协议可以适用于较短的距离。然而，在这种情况下，控制活动通融是必要的，但必须进行扩张（见表1）。
执行此实验时有两个关键步骤：1） 0 度校准和 2） 黑色校准。当使用外围轨道系统测量 MPOD 及其成分离中心时，0 度校准或浮力测量的外部固定至关重要。如果测量眼睛的参与者无法理解此过程或无法执行必要的步骤，则测量结果将受到影响并出错。黑色校准也至关重要，因为它允许 MPR 在无光时建立基线光谱仪测量，然后设备会将其与从受试者获得的所有值进行比较。因此，黑色校准是每个参与者的必备功能。
我们的研究结果表明，中央MPOD水平与以前发表的实验和组织学研究7、10、14的数据相匹配。此外，我们发现MPOD的水平随着视网膜偏心率的增加而下降，与对向区域相比，MPOD值在foveal上更大。叶黄素和玉米黄素水平在不同的视网膜位置也有所不同，叶黄素和玉米黄素比随着偏心的函数而变化。我们发现中央 L-OD 和 Z-OD 比率为 1：2.6，从中央固定 2 度时变为 2.08：1。这与先前研究10、29的报告一致。我们发现，叶黄素和玉米黄素水平表现出相当大的个体间变异。此前的体内实验室实验只评估了三个课题，而这方面的信息有限29。如果类胡萝卜素水平的显著个体间变化是正确的，那么这将支持需要获得类胡萝卜素的基线测量和定制个别补充剂。需要进一步的研究，以确认这一发现高个体间变异叶黄素和玉米黄素水平的健康个人。以前的出版物和与这种MPR器件的配合表明，在未扩张和扩张的阴孔条件下，MPOD可以获得可重复的测量，尽管当学生扩张25时，L-OD和Z-OD测量的可重复性得到了提高。在本研究中，我们用扩张的学生进行了所有MPR测量。鉴于类胡萝卜素水平在叶边缘和对叶区域较低，因此可能必须拉升孔，以便获得一致的信号强度和可靠的外围测量。
我们尝试了各种方法，并最终开发和测试了我们的轨道系统。事实证明，这是取得可靠结果的最有效方法。该系统通过多次检查三个参与者进行测试，看看每次尝试是否可以取得类似的结果。这包括在两个月内三次对参与者进行测量。其他尝试的方法包括修改反射仪目镜，方法是在中心外 0、1 和 2 度处创建具有预切切口的覆盖物。这种技术被证明是无效的，因为地道太接近了，无法充分区分受试者。
本研究有几个限制。研究要求受试者具有正常的双筒望远镜。这可确保受试者能够在测量另一只眼睛时固定目标。不符合此标准的主体将无法遵守说明，在使用刺激时无法正确固定，并且不能使用此技术成功测量。轨道系统是可靠的，但其局限性可以在今后的研究中解决。该协议可以通过内置红色 LED 固定灯和 Badal 光度计的一部分作为反射计的一部分来改进。这样，参与者可以固定到所需的偏心度，用适当的透镜来测量眼睛。
目前，没有替代技术来测量体内的L-OD和Z-OD。但是，存在测量 MPOD 的替代设备。其中一个装置是本研究中使用的异色闪烁光度计。异色闪烁光度计采用一种心理物理测试方法，无法确定单个叶黄素和玉米黄素值。使用异色闪烁光度计获得的中央 MPOD 测量值平均比 MPR 器件获得的平均值低 0.11，标准偏差为 0.16。使用这两种技术获得的MPOD测量具有与先前25所述的优异相关性。
虽然目前的研究样本量很小，但其目的是证明使用反射测量装置可以获得玉米黄素和叶黄素光学密度的外围测量。据我们所知，其他体内研究的样本数量比本研究中使用的样本要小得多。因此，我们相信，我们的结果表明，体内类胡萝卜素密度可以用反射计在foveola、foveal外围和对向转移区域测量。我们的研究进一步揭示了玉米黄素和叶黄素水平如何分布在人类视网膜的中央和周围黄斑区域。由于我们发现我们的研究参与者的价值有显著差异，因此需要在体内和体外进行更大的研究，以更好地了解黄素素和玉米黄素在一般人群中的分布、水平和比率。

年龄相关黄斑变性（AMD）是导致失明的主要原因，占全世界失明的8.7%1。与AMD相关的风险因素包括年龄增加、女性性别、吸烟、光虹膜颜色、脂质失衡、终生暴露在阳光和紫外线辐射下、系统性低水平的抗氧化剂、较低的黄斑色素光密度（MPOD）、遗传学和种族2。其中，可改变的危险因素是戒烟、口服抗氧化剂补充剂和类胡萝卜素。类胡萝卜素是植物和微生物中的天然色素，是有效的抗氧化剂3。它们由光合生物产生;人类从他们的饮食中获得类胡萝卜素3，4。黄斑色素由三种类胡萝卜素组成：叶黄素、玉米黄素和中角黄素4。黄素叶黄素和玉米黄素5在视网膜中被发现，特别是黄斑，并给fovea其黄色6。在视网膜5、7光受体的斧子和内丛状层中观察到较高浓度的xhophylls。类胡萝卜素的摄入量，如叶黄素和玉米黄素，增加黄斑色素的水平。叶黄素和玉米黄素是从膳食摄入或补充营养品中获得的，而中叶黄素只是叶黄素3、7、8代谢的副产品。叶黄素和玉米黄素浓度在视网膜的不同区域不同。中央，在叶黄素中，玉米黄素浓度大于叶黄素浓度，比例为2.3：19，10。类胡萝卜素的浓度在叶面外围每毫米降低100倍，其中叶黄素比玉米黄素更普遍，比例为2.4：19，10。
视网膜中黄斑虫的存在保护视网膜电路，特别是在叶面和黄斑中，并且对中央视觉至关重要。Xanthophylls通过两种可能的机制保护视网膜：1）过滤蓝光和2）减少氧化应激5，11，12，13。蓝光在视网膜中散射最多，黄斑色素水平较高，集中吸收散射光，从而改善视力。此外，可见光谱的蓝色部分由高能、短波长组成，可能导致视网膜中产生过多的活性氧。因此，人们认为类胡萝卜素通过作为内在视网膜和光受体视网膜色素上皮复合物的抗氧化剂，通过淬火这些自由基5，12，13，14来减轻黄斑的氧化负担。
视网膜类胡萝卜素的测量对全身健康有较大影响。最近的一项试验表明，类胡萝卜素治疗改善糖尿病患者的视网膜功能，而血糖水平没有任何改变15。视网膜中的类胡萝卜素密度水平也与大脑16的水平密切相关。胡萝卜素水平可能是在发育年17，18和大脑水平下降与19岁的关键。MPOD水平与儿童和老年人的神经保护和神经效率有关20，21。因此，有必要在临床上测量MPOD及其特征。这将在诊断、管理和治疗各种眼和系统疾病方面发挥作用7、15、16、17、18、19、20、21 。
目前市售的MPOD测量技术是异色闪烁光度计（HFP），它基于心理物理测试。这些测量在fovea上1度补丁，这相当于一个±0.30毫米直径的圆22。虽然这些类型的设备已被证明是可靠的，但它们受其主观性质的限制，使用时间，并且无法区分形成MPOD13，22，23，24的个别数量的xtohophylls。黄斑颜料反射计（见材料表），也称为反射计（见图1），通过客观地测量MPOD及其叶黄素和玉米黄素（黄黄素）的个别成分，解决了这些限制。反射仪利用紫外线/红外过滤和准直石英卤素源向视网膜发送受控光束（见图2），内部滤波器吸收大部分产生的辐射。因此，参与者很少或没有辐射暴露的风险。在文献26、27、28中，人们注视着人眼的各种色光和结构以及相应的吸收和反射模式。内部光谱仪处理的反射光分析允许定量隔离和测量叶黄素和玉米黄素光密度（L-OD、Z-OD）以及整体MPOD。第三个视网膜类胡萝卜素中青素在光谱上与玉米黄素没有区别，因此Z-OD代表两种类胡萝卜素29的组合。先前的工作表明，在测量中央 L-OD、Z-OD 和 MPOD25、29时，反射测量是可靠的。
目前研究的目的是创造一种技术，可用于在体内产生人类叶黄素和对叶酸水平的估计。其他目的是将研究结果与先前发表的实验室及动物学结果比较14、29。本手稿中开发和描述的方法及其与反射测量学一起测量的周游MPOD是新颖的。该技术可与任何现有的反射测量单元一起使用，无需进行重大修改，即可在各种视质和对硫位置测量单个类胡萝卜素（如 L-OD 和 Z-OD）的视网膜水平。
本手稿中提出的研究包括 8 名参与者，年龄从 22 到 29 岁不等。我们的方法包括首先进行常规眼科检查，以确保研究参与者符合纳入标准。在获得知情同意后，每个研究参与者接受以下四项测试：1） 使用市售的异色闪烁光度计设备进行中央 MPOD 测量;2） 利用反射计装置进行两次中央测量;3） 将同一反射仪装置与外围轨道系统结合使用，测量类胡萝卜素水平在1度偏心处，即直径0.30毫米的圆，中心距中央圆点0.30毫米;4） 使用相同的设置，类胡萝卜素水平在2度偏心，一个0.30毫米直径圆放置在fovea的边缘（对向区域），也测量。
MPR测量是在用1%的热带酰胺眼液扩张每个参与者的学生后进行的。众所周知，使用反射测量获得MPOD值不需要小孔扩张，但可以提高L-OD和Z-OD测量25、29的可重复性。这可能是因为使用反射计从视网膜获得的测量在学生扩张时具有更好的信噪比。为了进行精确、稳定的外围反射测量，参与者使用被放置在光学无穷大30，31的固定目标。
我们获得了 30 s 的反射计测量值，并丢弃了前 10 s 的数据。此过程有两个优点：1）信号源明亮，使眼睛能够适应和适应任务;2） 最重要的是，光感受器颜料漂白在前10。因此，消除前10s的测量允许一个更稳定和准确的信号29。在本研究中，我们进行了两次反射测量测试，之后我们对测量值求平均值，以获得平均 MPOD、L-OD 和 Z-OD 值以及每个参与者的 Z-OD/L-OD 比率。
