最近では、30以上の細胞マーカーを集中的にスクリーニングし、サンゴ細胞に標識が可能な24個を同定し、そのうち16個はユニークな集団18を区別するのに有用であり、分化クラスター(CD)を作った。ここでは、ポシロポラダミコーンスにおけるサンゴ細胞単離法が炭酸カルシウム骨格から細胞を除去することから、FACSを用いた特定の細胞集団の同定および単離に及ぶ過程について説明する(図1)。

このプロトコルは、これまで不可能な記述レベルで生きたサンゴ細胞集団の同定と単離を容易にする。この技術の主な利点は、これまでほとんど達成不可能であった細胞特異性のレベルへのアクセスを可能にすることです。この手順を実証するのは、シルベスター総合がんセンターのフローサイトメトリー共有リソースの研究員であるシャノン・サイグ博士です。ライブサンゴ細胞のソート用のフローサイトメーターを設定するには、フローサイトメーターソフトウェアで新しいプロジェクトテンプレートを開き、レーザーパネルで分析に適したレーザーを選択します。次に、適切なプロットに側面散布図と前方散布図を作成し、格子および解析戦略を設定します。フローサイトメトリック分析と生きたサンゴ細胞の選別のために、細胞計のサンプルチャンバーにコントロール未染色細胞サンプルをロードし、読み取りプロセスを開始します。サンゴ細胞は特に壊れやすいので、細胞のリシスを防ぐために細胞メーターの圧力を低く保ちます。散布図では、写真の乗数電圧を調整してポイントを中央に配置し、イベントの記録を開始します。前方散布図では、前方散布図の 10 から 4 番目のマークの周りに、セルの周囲にゲートを作成します。コントロールサンプルを染色した細胞の最初のサンプルに置き換え、一時停止する前に約10,000個のセルを記録します。次に、染色されたサンプルグループに固有のイベントをゲートします。生きたサンゴ細胞を選別するには、フローサイトメーターソフトウェアで関心のある細胞集団を選択した後、適切な回収液の250〜500マイクロリットルを含む1つのマイクロ遠心チューブを、各集団が選別するサイトメーターコレクションチャンバーに入れる。残骸を洗い流すために適切な時間が経過したら、所望の集団を選別して10,000から数百万個の細胞を収集します。新しい染色、種、またはソーターを使用するたびに、関心のある少なくとも20,000個の細胞を500マイクロリットルの染色媒体に分類して、対象の集団に対して純度チェックを行い、ソートされた細胞を再分析して、最初のソートに使用されるゲート内でイベントが読み取られていることを確認します。インビトロ研究では、並べ替えられた細胞をインキュベーターまたは滅菌環境に移すまで氷の上に保管します。サンゴ細胞と同じ形状や粒度を共有しない他の非細胞粒子を持ち込むためには、前方および側面散布図でゲート選択を作成することにより、解析から除去することができます。死細胞は、ヒストグラムを用いて染色されていない細胞懸濁液のDAPIシグナルを比較し、高DAPI発現細胞を外に出すことによって除外することができる。並行して、自己蛍光共生症をホストする細胞は、遠赤色チャネル内の高いシグナルを有する細胞に対して行うことによって除去することができる。この反復ゲーティングプロセスの後、残りの細胞は共生性を欠く無傷の生きたサンゴ細胞集団を表す。これらの細胞は、活性酸素種活性および/またはリソソーム含有量などの特徴を染色することによって、異なる細胞亜集団に分類することができる。一部のセルは他のセルよりも脆弱であり、このプロセスを可能な限り慎重に実行することが重要です。特定の細胞が単離されると、細胞培養の確立やトランスクリプトームプロファイルの作成などのx-vivo機能アッセイに使用できます。
