열은 모든 M. 결핵 균을 비활성화 제어(live cells)의 광학 밀도(OD)는 시간이 지남에OD따라 증가하였고(0.04 OD-0.85OD)열 불활성화 샘플에서 OD 변화가 관찰되지 않았으며, 각각 성장 및 성장이 없음을 의미하였다(도1)27. 유사하게, 제어 임상 가래는 MGIT에서 3일째까지 양성으로 성장했으며 열 불활성화 임상 샘플은 인큐베이션이 끝날 때까지 양성 플래그를 표시하지 않았습니다. MGIT에서 Mtb의 성장은 지엘-넬센 얼룩 현미경 검사법 및 항원 MPT6429에의해 확인되었다.
불활성화 된 샘플의 RNA는 4 일 동안 37 °C에서 안정적입니다. 열 불활성화 샘플은 세포의 열 불활성화 에 따라 RNA가 저하되는지 여부를 결정하기 위해 37 °C에서 배양하였다. BCG 배양물 모두에서 D1, 2, 3 및 4에서 단리된 열 불활성화 직후의 데이(D) 0에서 수확된 RNA와 4의 RNA 사이에는 차이가 발견되지않았다(도 2A-C)및 결핵 양성 가래(도2D).
외인성 RNase는 열 불활성화 분획에서 RNA 분해속도를 증가시킵니다. RNA가 분해되지 않는 이유를 확인하기 위해 RNase A 효소는 열 불활성화 전 및/또는 열 불활성화 전후에 1,000 U/mL에서 외인성으로 첨가되었습니다. 이것은 세균 부하 에 상응하는 RNA 손실을 일으켰습니다 1.5± 0.3 -, 1.8±0.2 -, 및 1.3±0.1 –80°C, 85°C 및 95°C에서 각각 4일간의 배양에 걸쳐 10eCFU/mL. 16S rRNA를 기준 마커로 사용하여 TB-MBLA에 충분한 RNA가 보존됩니다. rRNA에 대한 열 불활성화의 효과는 결핵 양성 환자로부터 BCG 배양 및 가래에서 측정되었다. 대조군 BCG 배양의 측정된 세균 부하, 5.3±0.2 로그10 eCFU/mL, 결합된 5.1±0.3 로그10 eCFU/mL보다 높은 열 불활성화 배양(ANOVA p < 0.0001)에서 80°C, 85°C, 25°C, 45°C에서 45°C, 25°C, 25°C, 2725°C에서 유사하게, 대조군 환자 가래의 세균 부하, 7.1 로그10 eCFU/mL, 결합된 6.3±0.41 로그10eCFU/mL 80°C, 85°C 및 95°C(도4B)27보다0.8±0.1 로그10 eCFU/mL 높았다. 세균 부하 감소는 시험된 두 가지 유형의 샘플에서 <1log였다.
Sidak의 다중 비교 시험은 80°C 및 85°C(p = 0.001)의 95°C에서의 세균 부하 간의 유의한 차이를 밝혀냈습니다. 모든 온도에서 TB 샘플에서 차이가 발견되지 않았습니다, p = 0.8.
