HHT患者では、ENG(CD105)の単一の突然変異が報告されている。この突然変異は、EC増殖の増加と流動媒介EC伸長の減少を招く11,2である。また、ENG欠乏症はゼブラフィッシュ3の内皮マーカー発現(すなわち、kdrl、cdh5、hey2)を低下hey2させることも以前に報告されている。 kdrl cdh5エンドグリンは、主に内皮細胞に発現し、膜貫通糖タンパク質であり、成長因子β(TGF-β)ファミリーメンバーを変換するための共レセプターとして機能する。これは、BMP9結合を細胞表面に指示して、Id1発現を含む下流遺伝子を調節し、幹細胞分化をECs4に誘導する。従って、eng遺伝子は血管形成およびヒト血管疾患55,66において重要な役割を果たす。我々は、ゼブラフィッシュ胚における血管形成に対するエンドグリンノックダウンの影響を検討し、続いて、ENG突然変異を有するHHT患者から取得したiPSC由来のICの分析を行った7。このプロトコルは、EN欠乏が内皮前駆細胞マーカー発現および管形成に及ぼす影響を示し、これはインビトロ血管新生を測定するための定量化可能な方法である。
空間的および時間的発現を研究するために、WISHは、生体内8において遺伝子発現を検出するために採用される。インサイチュハイブリダイゼーション(ISH)では、標的核酸(DNAまたはmRNA)の補数配列を有する標識プローブを用いて、固定検体中の標的核酸ハイブリッドを検出および可視化する方法である。このプロセスは、生体内の遺伝子発現シグナルを増幅し、顕微鏡による遺伝子の発現を検出するために使用されます。WISHは、特にゼブラフィッシュ9で、様々なモデル動物で広く使用されています。また、次のデータを取得するために使用されます: 1) 遺伝子の空間/時間発現パターン, 遺伝子機能と分類に関する情報を提供します;2)高スループット薬物または変異体スクリーニング10に使用される遺伝子マーカーを特異的に発現させる。
染色原性プローブは、従来の染色体を用いてそのシチュハイブリダイゼーション(CISH)で容易に分解され、高いバックグラウンドノイズと非特異的信号11,12,12を生じる。WISH法は、RNA配列を標的とするようにハイブリダイズするように設計された2つの独立した二重Zプローブを使用します。各プローブには、標的RNAと相補的な18~25個の配列と14個のベーステール配列(Zとして概念化)が含まれています。ターゲットプローブはペア(ダブルZ)で使用されます。2つのテールシーケンスは、アンプ用の20の結合部位を含むプリアンプ用の28ベースのハイブリダイゼーションサイトを形成します。このアンプは、標識プローブ用の20個の結合部位を含み、理論的には各ターゲットRNA配列に対して最大8,000個のラベルを生成できます。
この高度な技術は、組織形態13を維持しながら、単一分子可視化を達成するために同時に信号増幅とバックグラウンド抑制を促進する。WISH法のさらなる修飾は、以前の研究14に基づいており、追加の固定およびDAB染色を含む。ここで提供される改良されたWISHプロトコルは、標的RNAが部分的に減少または分解された場合でも動作することができる。利点はRNase自由条件なしで24時間で完了することができることである。複数のターゲットからの複数のチャネルを通じて同時に信号を検出することができ、結果は一貫しており、異なる高スループットの自動化プラットフォームの結果と互換性があります。
動物モデルの結果は、ヒトで起こるのと同じ現象を反映する必要はない。ENGには、孤児の領域で二硫化橋を形成するC30-C207とC53-C182の2組の保存されたシステインが含まれています。HHT患者におけるengの役割をさらに研究するために、HHT患者由来のiPSCを用いたチューブ形成アッセイは、eng変異を伴わない/またはeng変異を伴わない細胞で行われている(Cys30Arg,C30R)15。Kubotaらが最初にチューブ形成実験16を報告した後、アッセイはいくつかの方法で開発された。血管新生または抗血管新生因子を同定し、血管新生におけるシグナル伝達経路を定義し、血管新生17を調節する遺伝子を同定するために用いられている。
患者由来のiPSC-ICが利用できる前に、研究者は主に培養されたECを使用して血管新生16を研究した。しかし、内皮細胞の場合、ECが受けることができる通過数が限られているため、ウイルスを持つ外因性遺伝子をトランスデュースすることは技術的な課題です。これは、手術または一致した承認されたドナーのいずれかから人間の血管から収集されるのに十分な細胞材料がほとんどないためです。山中信也によるiPSC生成技術の発明により、iPSCに由来するヒトICは、前に報告された18のインビトロ実験で確実に使用することができる。
数と通過が限られたウイルス伝達されたICを使用すると、シグナル伝達研究には十分かもしれませんが、機能研究のためには、変異型多能性幹細胞株(iPSCまたはCRISPER/Cas9標的hESCのいずれか)を生成し、チューブ形成アッセイ19を使用する血管新生研究のためにそれらをECsに区別する方が良い。チューブ形成は、突然変異を有する内皮細胞の機能を評価するために使用することができる。このプロトコルは、簡単で費用対効果が高く、再現可能な方法であるμ-Slide血管新生プレート上のチューブ形成についても説明します。

このマウント全体は、その一方でのハイブリダイゼーションで、WISHは従来の方法と比較して高解像度と低いバックグラウンド結果を提供します。また、Uスライドプレートは、マウス胚、ショウジョウバエ胚および他の組織に適用できる願い物を少なくすることで同じ焦点距離で焦点を改善する。チューブ形成アッセイは、幹細胞分化アンドロゲン細胞に適用することができる。WISHは、変異補正の機能的意味を理解しやすくします。この手順をデモンストレーションする人は、私の研究室の研究助手であるヨン・ワンです。顕微鏡下で作業するフェムトジェットシステムを使用して、モルフォリノスのナノグラム2個とMRNA 500ピコグラムを1細胞胚に注入します。モルフォリノスのマイクロインジェクションを何度も練習し、細胞に正常に注入し、卵の完全性を保つことができます。彼らがデコリオネートされ、意図した実験に対応する開発の状態に達するまで、接合者をインキュベートします。原稿の表1に記載されているように。次に、プラスチックピペットを使用して、20〜40個の胚を1.5ミリリットルチューブに移します。室温で作りたての4%PFA溶液を1ミリリットル加えます。原稿に記載されているように胚を固定、洗浄、脱水した後、下部にナイロンメッシュを付けたふるいに胚を入れる。胚を75%50%および25%メタノールでそれぞれ5分間順次洗浄して水和する。PBSTで胚を洗浄した後、胚とチューブに50マイクロリットルのプロテイナーゼKを加える。消化後、プロテナーゼKを取り出し、毎回5分間PBSTで胚を3回洗浄します。胚を含む各チューブに50~100マイクロリットルの混合ターゲットプローブを加えます。胚を摂氏40度で一晩インキュベートする。原稿に記載されているように胚を洗浄した後、4%パラホルムアルデヒドを使用して、胚を室温で30分間固定する。その後、2倍のSSCT溶液で胚を室温で15分間3回洗浄します。SSCT溶液を取り外し、Amp 1の50マイクロリットルに交換してください。その後、40°Cで胚をインキュベートします。30分後、胚を室温で15分間、2倍のSSCT溶液で3回洗浄する。SSCT溶液を取り外し、Amp 2を50マイクロリットル加えます。15分間インキュベートし、先に行ったように胚を洗浄した後、50マイクロリットルのアンプ3を加え、チューブを軽くタップします。胚を摂氏40度でインキュベートする。30分後に先述のように胚を洗浄する。その後、Amp 4ドロップワイズの50マイクロリットルを追加し、慎重にチューブをタップします。その後、胚を摂氏40度で15分間インキュベートし、SSCTで3回洗浄します。原稿に記載されているようにイメージング用の胚を調製した後、DABペルオキシダーゼ基材キットを使用して標本を染色する。1ミリリットルの蒸留水に各色試薬A、B、Cを50マイクロリットル加えます。そして完全なDAB作動液を得るためによく混ぜる。試料に流体を加え、10分間覆います。検体を十分に洗浄した後、光学顕微鏡で画像を作成します。HHT iPSCの培養と制御を開始するには、ウェルの底部を覆う6つのウェル細胞培養プレートに基底膜マトリックスを追加します。プレートを摂氏37度で1時間インキュベートします。次に、使用した基膜マトリックス溶液をウェルから取り出し、各ウェルに2ミリリットルのmTeSR 1培地を加える。その後、最後の通路から収穫したiPSCを1回あたり約10倍から6個の細胞にプレートします。iPSCを約4日間培養した後、細胞培養培地を添加したBEL培地と交換する。中皮細胞を生成するために3日間細胞を成長させます。血管内皮細胞を拡大するには、培地を4日間補充されたBEL培地に置き換える。そして、同じ培地と培養物を含む細胞を、さらに3〜4日間治療します。キットの指示に従ってCD31ダイナビーズを使用して成熟した血管内皮細胞を精製します。次いで、溶出緩衝液によりCD31陽性細胞を収集する。VEGF165、bFGFおよびFBSを含むEC-SFM培地中の精製内皮細胞を維持および拡大する。内皮細胞をプレートするには、血管新生プレートにウェルあたり10マイクロリットルの基基膜マトリックスを加える。37°Cで30分間プレートをインキュベートします。内皮細胞を収穫し、繰り返しピペット処理することにより、内皮増殖培地2に再懸濁する。この細胞懸濁液の50マイクロリットルを各ウェルの固化マトリックスに加え、37°Cで細胞をインキュベートします。3~5時間のインキュベーションの後、高分解能顕微鏡を使用して内皮管形成を評価します。各グループに対して少なくとも10のエリアの画像をキャプチャします。チューブ全体の長さ、チューブ番号、分岐点を確認します。CISHおよびWISHは、受精後24時間の胚におけるエンドリンの発現を決定するために行われた。血液形成性内皮マーカーと内皮前駆体マーカーを用いて、エンドグリンサイレンの効果を調べた。赤い矢印は、これらのマーカー、aplnrnaおよびnpr1aの発現が有意に減少した領域を示す。内皮管形成アッセイを、エンドグリン変異体およびコントロールiPSC由来内皮細胞に対して行った。チューブ形成を評価し、3時間後に撮影した。チューブの長さ、チューブ番号および分岐は、ImageJソフトウェアを用いて定量化した。変異型内皮細胞は、コントロール内皮細胞よりも少ない枝を形成した。そして、血管内皮増殖因子による刺激後に変異内皮細胞の枝が有意に増加した。また、この方法は末梢血からのiPS生成にも使用できます。そして、この方法により、インビトロおよびインビボにおける血管形成における内分泌の役割を明らかにする。突然変異を矯正すると血管新生に違いが生じる可能性があることを知って、潜在的な幹細胞療法のために患者に細胞を移植することが可能である。
