Aufgrund der spezialisierten Rolle bei der Bereitstellung von B-Zell-Hilfe bei der Erzeugung von Hochaffinitätsantikörpern wurden Tfh-Zellen ausgiebig in den Mechanismen der Differenzierung und Manipulation untersucht, um neue Strategien für das Impfstoffdesign zu liefern. Influenza-Virus-Infektion induziert kräftige Tfh und GC B Zellen Reaktionen, so dass ein geeignetes Modell für dieses Forschungsfeld. In den letzten zehn Jahren konzentrierten sich zahlreiche Studien auf die neu identifizierte CD4+ T-Zell-Untermenge, Tfh-Zell-Submenge, für ihre wesentliche Rolle in der Entwicklung des Keimzentrums (GC) B. B-Zell-Lymphom 6 (Bcl6), das hauptsächlich als Genrepressor betrachtet wird, ist der Liniendefinierende Faktor von Tfh-Zellen für den Beweis, dass die ektopische Expression von Bcl6 ausreicht, um die Tfh-Differenzierung anzutreiben, während ein Mangel an Bcl6 zu einer verschwundenen Tfh-Differenzierung1,2,3führt. Im Gegensatz zu anderen CD4+ T-Helfer-Untergruppen, die ihre Effektorfunktion durch Migration zu den Entzündungsstellen ausführen, bieten Tfh-Zellen die B-Zellhilfe hauptsächlich in der B-Zell-Follikelzone von Milz und Lymphknoten. Die kostimulierenden Moleküle ICOS und CD40L spielen eine wichtige Rolle in der Interaktion zwischen Tfh- und GC-B-Zellen. Während der Tfh-Differenzierung überträgt ICOS notwendige Signale von kognaaten B-Zellen und fungiert auch als Rezeptor, der Migrationssignale von Bystander-B-Zellen für die B-Zellzonenlokalisierung4,5empfängt. CD40L ist ein Mediator von Signalen von Tfh-Zellen für B-Zellen Proliferation und Überleben6. Ein weiterer Faktor, der die ähnliche Rolle wie CD40L spielt, ist das Zytokin IL21, das hauptsächlich von Tfh-Zellen abgesondert wird. IL21 reguliert direkt die Entwicklung und Produktion von Gc B-Zellen entwicklung und Produktion von hochaffinen Antikörpern, aber seine Rolle bei der Tfh-Differenzierung ist immer noch umstritten7,8. PD-1 und CXCR5, die heute am häufigsten bei der Identifizierung von Tfh-Zellen in der Durchflusszytometrie-Analyse verwendet werden, spielen ebenfalls eine wichtige Rolle bei der Differenzierung und Funktion dieser Teilmenge. CXCR5 ist der Rezeptor für B-Zell-Follikuläres Chemokin und vermittelt die Lokalisierung von Tfh-Zellen in B-Zellfollikel9. PD-1 ist nun nicht nur identifiziert, um die follikuläre Führungsfunktion zu haben, sondern auch kritische Signale im Prozess der GC B-Zellen Affinitätsreifung10zu übertragen. Basierend auf diesen Erkenntnissen könnte die Bewertung der Expression dieser Moleküle im Wesentlichen die Reifung und Funktion von Tfh-Zellen widerspiegeln.
GC ist eine induzierte transiente mikroanatomische Struktur in sekundären Lymphorganen und stark von Tfh-Zellen abhängig, was eine perfekte Auslesung zur Bewertung der Tfh-Antwort ist. In GC unterliegen B-Zellen nach dem Empfang von Signalen, die durch Zytokine und kostimulierende Moleküle vermittelt werden, Klassenumschaltungen und somatischer Hypermutation, um hochaffine Antikörper zu erzeugen11. Differential-Antikörper-Klassen-Switching tritt in Differential-Zytokin-Nische, in der IL4 und IL21 induzieren IgG1-Klasse Schalten, während IFN induziert IgG2-Klasse Switching12. Plasmazellen sind die Produzenten von abgesonderten Antikörpern und endlos differenzierte Zellen. Wie Tfh-Zellen ist die Entwicklung von B-Zellen in GC mit der dynamischen Expression vieler signifikanter Moleküle verbunden. Basierend auf der aktuellen Studie können GC B-Zellen als B220+PNA+Fas+ oder B220+GL7+Fas+ Zellen und Plasmazellen identifiziert werden, im Vergleich zu ihren Vorläufern, Downregulate Expression von B220 und upregulate CD138 Expression13. Darüber hinaus können beide Eigenschaften in der Durchflusszytometrie und Immunfluoreszenzanalyse nachgewiesen werden, was eine angemessene Bewertung der GC-Reaktion ist.
Robuste zelluläre und humorale Reaktionen werden bei Influenza-Virus-Infektionen induziert, wobei Tfh- und Th1-Zellen die CD4+ T-Zellreaktion14dominieren, was sie zu einem perfekten Modell für die Tfh-Zelldifferenzierungsstudie macht. Influenza A/Puerto Rico/8/34 H1N1(PR8), ein häufig verwendeter mausangepasster Stamm, wird häufig in dieser Studie verwendet14,15,16. Hier beschreiben wir einige grundlegende Protokolle des Tfh-Studien-relevanten Assays bei Influenza-Virusinfektionen: 1) intranasale Impfung des PR8-Virus; 2) antigenspezifische Tfh-Zellen, GC B- und Plasmazellen und IL21-Nachweis mit Durchflusszytometrie; 3) histologische Visualisierung von GC; 4) Nachweis von antigenspezifischem Antikörpertiter im Serum mit ELISA. Diese Protokolle liefern die notwendigen Techniken für neue Forscher in Tfh-assoziierten Studien.

T Follicular Helper (Tfh) Zellen sind eine unabhängige CD4+ T-Zell-Untergruppe, die auf die Unterstützung bei der Entwicklung von Keimzentren (GC) und der Erzeugung von Antikörpern mit hoher Affinität spezialisiert ist. Bei einer Influenza-Virusinfektion werden robuste Tfh- und GC-B-Zellreaktionen induziert, um eine effektive Virusausrottung zu erleichtern, die ein qualifiziertes Mausmodell für Tfh-assoziierte Studien ermöglicht. In diesem Beitrag haben wir Protokolle zum Nachweis der grundlegenden Tfh-assoziierten Immunantwort während einer Influenzavirusinfektion bei Mäusen beschrieben. Zu diesen Protokollen gehören: intranasale Impfung des Influenzavirus; Durchflusszytometrie Färbung und Analyse von polyklonalen und antigenspezifischen Tfh-Zellen, GC-B-Zellen und Plasmazellen; Immunfluoreszenznachweis von GCs; enzymgebundenen Immunsorbent-Assay (ELISA) von Influenza-virusspezifischen Antikörpern im Serum. Diese Assays quantifizieren grundsätzlich die Differenzierung und Funktion von Tfh-Zellen bei Influenza-Virusinfektionen und helfen so bei Studien zur Aufklärung von Differenzierungsmechanismus und Manipulationsstrategie.
