תאי T Follicular Helper (Tfh) הם תת-קבוצה עצמאית של תאי CD4+ T המתמחה במתן עזרה לפיתוח מרכז נביטה (GC) ויצירת נוגדנים בעלי זיקה גבוהה. בזיהום וירוס שפעת, חזק Tfh ו- GC B תגובות תאים המושרים כדי להקל על מיגור וירוס יעיל, אשר מעניק מודל עכבר מוסמך למחקר הקשורים Tfh. במאמר זה תיארנו פרוטוקולים באיתור תגובה חיסונית בסיסית הקשורה ל-Tfh במהלך זיהום בנגיף השפעת בעכברים. פרוטוקולים אלה כוללים: חיסון תוך-אנלתי של וירוס שפעת; כתם cytometry זרימה וניתוח של תאי Tfh פוליקלוניים ואנטיגן ספציפי, תאי GC B ותאי פלזמה; זיהוי חיסוני של מחשבים אישיים; בדיקה אימונוסורבנטית (ELISA) הקשורה לאנזימים של נוגדן ספציפי לנגיף שפעת בסרום. מבחנים אלה בעצם לכמת את הבידול והתפקוד של תאי Tfh בזיהום וירוס שפעת, ובכך לספק עזרה למחקרים במנגנון בידול מבהיר ואסטרטגיית מניפולציה.
