骨形態形成タンパク質(BMP)は、成人構造の早期胚発生およびパターニングに不可欠です。BMPは、応答細胞の増殖および細胞分化に必要な標的遺伝子の転写に影響を与えるために産生され、分泌される。デカペンタペティック(Dpp)は、翼1、2、3、4のような胚および成人構造の発達に重要であるBMP4のショウジョウバレギである。いくつかのグループは、1)翼が簡単に評価することができる一貫したベナミズパターンを持つ2つの透明な上皮シートで構成されているため、大人のフライウィングのパターン化におけるDppの役割に焦点を当てています。2)翼ディスクも合理的に平らであり、幼虫の外で培養することができ、パターンの違いを画像化し、定量化することが簡単です。3)翼パターンの発達は、経路内の小さな摂動が翼の静脈パターンに影響を与えるようなDppに敏感である。
Dppは、翼ディスク5、6、7、8の前/後部境界に位置する細胞内で産生される。Dppは、タイプ1およびタイプ2セリン/スレオニンキナーゼ受容体9、10の複合体に結合する。Dpp結合時に、タイプ2受容体は、Dpp(マッド)に対して母親をリン酸化するタイプ1受容体をリン酸化し、スマッド1/5/8ホモログである。リン酸化SMADは、標的遺伝子を調節する核に入ることを可能にする追加の共スマド(Medea)を募集し、増殖または分化4、11などの下流効果をもたらす。
近年、ベイツラボは、翼ディスク内のDppの不適切な放出が、マッドリン酸化の減少、標的遺伝子発現の減少、および翼パターニング欠陥12、13につながる可能性があることを示している。いくつかのイオンチャネルは、ショウジョウバアラ翼および関連構造物14、15の発達に影響を与える。これらのイオンチャネルは、Dpp リリースにも関与する可能性があります。モルホゲン放出のメカニズムを決定する際には、リリースイベントを可視化する方法が重要です。

ここでは、Dpp-GFP を使用して Dpp リリース イベントを視覚化するために使用する方法について説明します。Dpp-GFPは、UAS-GAL4システム19を使用してショウジョウバエウィングディスクにおけるそのネイティブパターンで表現することができる。これは、Irk チャネルが Dpp リリース18に影響を与えることを判断するために使用された方法です。z スタックをライブイメージングしてメソッドを検証しました。Dpp-GFP punctaが焦点の1つの平面で取得した場合、時系列の焦点の平面内で移動するのを見ていません。また、Zスタックで画像を作成した場合、Dpp-GFPパンクタの動きは見当たりません。我々は、この方法を用いて見られるDpp-GFPパンクタは、細胞内で小胞の動きではなく、放出事象であると結論付ける。Dpp-GFP のライブイメージングのこの方法は、Dpp ダイナミクスへの影響について Dpp リリースの他のポティティブ修飾子をテストするために使用される可能性があり、または他のリガンドのダイナミクスを調べるように変更できます。
