このプロトコルは、好中球の群れのインビトロ分析を可能にし、現在の実験不能の多くの制限を克服するため、重要です。この技術の主な利点は、好中球が群れの間に放出し、定量的な画像分析を可能にする分泌されたサイトカインおよび脂質メディエーターへの容易なアクセスを提供することです。この技術を好中球に応用した一方で、単球やT細胞のような他の白血球の移動を分析するために拡張することができます。適切な量のCP粉末を水に加え、一晩かすべての固形物が溶解するまで攪拌板に混合することによって、1ミリリットルカチオン性ポリエレクトロライト(CP溶液)あたり1.6ミリグラムを調製することから始めます。必要に応じて、FITCで標識したポリL-リジンを添加することにより、溶液を蛍光化することができる。標準的なフォトリソグラフィ法を使用して、マスターシリコンウエハを生成します。ここで使用される設計は、直径30マイクロメートルの4ミリリットルの矩形配列で、中心間に150マイクロメートルの中心がありますが、異なる用途に合わせて変更することができます。シリコンウエハーに負のフォトレジストの厚さ40マイクロメートルの厚い層をスピンコートします。その後、ウエハースを摂氏65度で5分間焼きます。そして10分間摂氏95度。150~160ミリジュール/センチメートルのクロムフォトマスクを通してウエハーをUV光に当てよばします。ウエハースを摂氏65度で再び5分間、摂氏95度で10分間焼きます。その後、フォトレジスト開発者に10分間水没させます。そして、イソプロピルアルコールでそれをすすいでください。パターンがウエハーに表示されるようになりました。次に、ポリジメチルシロキサン、またはPDMS、プレポリマー及びその硬化剤の10対1の比率を十分に混合する。そして、ペトリ皿のマスターウエハースの上に未硬化混合物を注ぎます。真空は、気泡が存在しなくなるまで、未硬化PDMS混合物を処理します。その後、摂氏65度で一晩それを治します。翌日、メスを使ってウエハーのパターン化された部分の外側を切り回します。そして、ゆっくりと硬化したPDMSスラブを取り外し、パターン化された側面を上に向けたきれいなまな板の上に置きます。PDMSスラブから8ミリメートルの生検パンチで個々のスタンプをパンチアウトし、各スタンプを粘着テープに伏せして破片を取り除きます。これらのスタンプを上に向けて、各スタンプに100マイクロリットルの事前準備されたCP溶液を盛り付け、溶液とスタンプの間に気泡が形成されないようにします。次に、CP溶液の層にスタンプを反転し、1時間後にそれらを削除します。各濡れたスタンプを清潔なガラススライドに6〜8回下にして、余分な液体を取り除きます。その後、1〜2分間スタンプを真空処理します。スタンプ配置のガイドとして、きれいなガラススライドの上に8ウェル、直径8ミリメートルのイメージングスペーサーを付着させます。画像スペーサの各ウェルの中央にあるガラススライドの上にスタンプを下に置きます。その後、各スタンプの上に5.6グラムのバランスのとれた重量を置き、スタンプのために10分を許可します。スライドから重量とスタンプを取り除き、CP層が室温で24時間乾燥してからバイオ粒子を添加します。CPにFITCがタグ付けされている場合は、蛍光顕微鏡でスタンプの有効性を確認してください。空白のPDMSスラブをイメージングスペーサーのサイズにカットし、8ミリメートルの生検パンチを使用して、スペーサーの井戸に合わせたPDMSの井戸を作成します。次に、PDMSスラブをガラススライドに付着させます。バイオ粒子の溶液を解凍し、注射のために水で1ミリリットルあたり500マイクログラムに希釈します。ガラススライド上の各PDMSウェルに100マイクロリットルの生物粒子溶液を加え、スライドを30分間揺らします。水で井戸を十分に洗い流し、蛍光顕微鏡でスライド上のパターンを確認します。この生物粒子マイクロアレイは、最大3ヶ月間、摂氏4度のほこりのない環境に貯蔵することができます。0.4%のヒト血清アルブミンでIMDMで1ミリリットル当たり5番目の細胞に7.5倍10回で再懸濁して細胞を調製します。100マイクロリットルの懸濁液を、生物粒子マイクロアレイを含むPDMSウェルに加え、細胞懸濁液がPDMSの上部に凸状であり、気泡が含まれていないことを確認します。直径12ミリメートルのカバースリップで井戸を密封し、ピンセットで軽く押し下げて余分な細胞サスペンションが井戸の端まで脱出し、組織を使用して過剰を取り除きます。細胞を画像化するには、37°C、5%の二酸化炭素、および90%相対湿度に設定されたケージインキュベーターを装備した顕微鏡の生きた細胞イメージングステーションに細胞をマイクロ粒子アレイにロードします。タイムラプス、蛍光、明視野顕微鏡を使用して、405ナノメートル、594ナノメートル、および明るいフィールドで10秒ごとに10倍の画像を記録します。37°Cのバイオ粒子と5%の二酸化炭素をウェル内の好中球を3時間インキュベートし、所望の時点でサンプルを採取する。明視野顕微鏡を使用して、マイクロアレイ上に群れが形成されていることを確認します。単一の井戸の上清の全容を収集し、0.45マイクロメートルの遠心フィルターチューブにロードし、トリプリケート内の各時間ポイントを分析することを確認します。チューブをG190倍、摂氏20度で5分間遠心し、濾過したボリュームを集めます。次に、サンプルを処理時間までマイナス80°Cで保存します。このプロトコルを試みるとき、フォトレジストとヒュームフードの開発者と一緒に働く。人間のドナーから血液を採取する前に、IRB承認プロトコルを用意してください。ヒトの血液由来の物質はBSL2であることを覚えておいてください。好中球を微生物配列に添加すると、生物粒子クラスターに接触する好中球が活性化し、群れの応答を開始します。この生物粒子マイクロアレイは、経度経過蛍光顕微鏡を用いて、生物粒子クラスターへの好中球の移動を追跡するために検証された。安定好中球の群は、30〜60分の曝露後に各クラスターの周りに形成される。対照的に、好中球は、生物粒子クラスターがない場合に集団移動を示さない。染色された好中球核の蛍光強度を用いて、生物粒子クラスター周辺の平均群れサイズが約1490マイクロメートル平方メートルであると判定した。クラスターがない場合、対象となる特定の領域の蛍光は時間の経過とともに一定のままでした。好中球移動のトラックは、好中球が存在したときに生物粒子クラスターに収束したことを示しているが、制御系には収束は認められない。群れの速度と非活性化好中球の速度を測定し、速度分布に統計的に有意な差を見つけた。群れの好中球の平均速度は毎分20.6マイクロメートルで、対照好中球の平均速度は毎分2マイクロメートルであった。群れの間に好中球が放出する16個のタンパク質の濃度を時間をかけて分析した。10タンパク質が濃度で増加した。2人が減少した。そして、残りの4つは群れの最初の1時間の間に増加したが、その後減少した。この手順を実行している間、これらは細菌粒子のパターニングを成功させるために不可欠であるため、すべてがきれいで、スタンプが適切に乾燥していることを確認してください。この技術の開発により、研究者は、遊離メディエーターや細胞外小胞が好中球細胞間コミュニケーションにどのように関与しているかなど、好中球の群れの分野で新たな疑問を探求することができます。
