このプロトコルは、出生後小脳から幹細胞を精製するための簡単で費用対効果の高い方法を記述する。これらの幹細胞は、後期生まれの分子的に文字化けしたインターニューロンを占めるが、出生後由来の小脳アストロサイトも占めている。したがって、これらの細胞を研究することは、小脳の発達を理解するために明らかに重要である。このプロトコルを使用すると、幹細胞の収率は、通常10倍高価であるFACSベースの戦略の収量に匹敵する。このプロトコルは、腸および骨髄などの他の組織から幹細胞を発現するプロミニン-1を抽出するために一般化することができる。まず、解剖顕微鏡の下に脳で皿を置くことから始めます。細かい5つの鉗子を使用して小脳から髄液および大きな血管を取り除き、ヘラを使用して脳幹から小脳を分離する。小脳を5ミリリットルの氷冷HBSS溶液を含む15ミリリットルの遠心分離管に移します。その後、HBSSをデカントします。さらに2回洗い流します。最後のすすみ後、摂氏37度に予め温めたパパインベースの組織解離液を5ミリリットル加えます。37°Cの水浴で15分間組織をインキュベートし、3分ごとに3〜5回反転してチューブの内容物をゆっくりと混ぜます。広径と狭径ガラスパスツールピペットをバンゼンバーナーで磨火で準備します。インキュベーション後、5ミリリットルのHBSS溶液で組織を3回洗浄し、デカンティング中に組織を失わないようにします。最後のHBSS洗浄後、250マイクロリットルのDNAseを含むDPBSを組織に5ミリリットル加え、広い直径のパスツールピペットで10〜15回穏やかに三度ずつ解約します。次に、狭いパスツールピペットを使用して、組織スラリーをさらに10回トリチュレートします。より大きな組織が残っている場合は、ピペットチップでチューブの底に押し付け、細かい懸濁液が達成されるまでピペットを続けます。遠心分離管を氷の上に置きます。その後、解約細胞を40マイクロメートルの細胞ストレーナーを通して50ミリリットルのチューブにひずみます。10 ミリリットルの氷冷 HBSS 溶液でフィルターを上にして、細胞がメッシュを通過するようにします。濾過した細胞を新鮮な15ミリリットルの遠心分離管に移し、300倍gの懸濁液を摂氏4度で10分間遠心分離する。その後、真空吸入器を使用して上清を完全に除去します。160マイクロリットルの氷冷磁気カラムバッファーで細胞ペレットを再懸濁します。細胞懸濁液に40マイクロリットルの抗プロミニニン-1マイクロビーズを加えます。次に、チューブを冷蔵庫で15分間インキュベートし、抗体がプロミニン-1発現細胞に結合できるようにします。細胞を1~2ミリリットルのカラムバッファーXで洗い、遠心分離機を300倍gで10分間洗浄します。上清を吸引し、ペレットを1ミリリットルのカラムバッファX.磁気スタンドに置き、500マイクロリットルのバッファXで1回リンスします。500マイクロリットルのバッファーXでカラムを3回洗い、磁場からカラムを取り出し、1.5ミリリットルのチューブに入れます。1ミリリットルの培養培地を加え、プランジャーをカラムに押し込み、プロミニン-1ビーズでタグ付けされた細胞を洗い流します。神経球を通過するには、培養培地と一緒に滅菌15ミリリットル遠心管に移す。5分間300回gで遠心分離により神経球をペレット化し、上清を捨てる。ペレットをパパインまたは0.05%トリプシン溶液で5ミリリットルの解離培地で再懸濁し、37°Cで細胞を10分間インキュベートする。再び細胞懸濁液を遠心分離する。その後、神経球培地の5ミリリットルで細胞を再懸濁し、パスツールピペットで10回ゆっくりとピペット化することによってそれらを解約する。テキストプロトコルに記載されているように神経球培地に細胞をプレートし、前述のように培養で7〜10日後に二次神経球でそれらを豊かにする。ヘモサイトメーター上の細胞を数え、将来の実験のためにポリD-リジンコーティングされたプレート上の細胞の最適な数をプレートします。カラム精製プロミニン-1陽性出生後小脳幹細胞は、成長因子が豊富な神経圏培地で神経球を形成した。これらの神経球はプロミニン-1、単離のために使用されるマーカー、およびネスチンおよびGFAPのような他の幹細胞マーカーに陽性であった。フロースルー中の細胞は、幹細胞マーカープロミニン-1およびネスチンおよびニューロンマーカーβ-IIIチューブリンに対する陽性に対して陰性に染色された。成長因子の離脱時および白血病抑制因子またはPDGF-AAの存在下で、プロミニン-1陽性神経球はニューロン、アストロサイトおよびオリゴデンドロサイトに分化した。この技術を試みるとき、正常な細胞はまた、神経球のように見える塊を形成する傾向があるため、神経球がプロミニン-1抗体のために押し通され、通過できることを確認することが重要です。この分離法を用いて、健康や疾患の状態におけるニューロンやグリアのような誘導系統を特徴付けることができます。この方法は価値があり、小脳の発達における小脳幹細胞の役割と、二分脊髄性失調症および癌のような疾患状態に関する新たな洞察を提供する。
