Schaalbare en efficiënte methoden voor de differentiatie van hESCs in verschillende kilometerstanden en het genereren van voldoende aantallen gedifferentieerde cellen zijn belangrijke criteria voor geneesmiddelen screening en stamceltherapie. Verschillende methoden voor het passeren van één cel zijn gepubliceerd, waarin cellen worden gekweekt in de aanwezigheid van rotsremmer of andere kleine moleculen om de overleving te verbeteren, maar de eindproducten van deze cultuur methoden zijn kolonie type hescs<sup class="xr...

Menselijke embryonale stamcellen (hESCs) hebben het potentieel om te differentiëren in de drie primaire kiem lagen, die vervolgens differentiëren in verschillende multipotente voorlopercellen. Deze kilometerstanden geven vervolgens aanleiding tot alle celtypen in het menselijk lichaam. In vitro cultuur systemen van hESC hebben het vermogen om menselijke embryonale ontwikkeling te bestuderen getransformeerd en hebben gediend als waardevol hulpmiddel voor het verkrijgen van nieuwe inzichten in hoe deze processen op het biologische en moleculaire niveau worden gereguleerd. Evenzo, studies van geïnduceerde pluripotente stamcellen (iPSCs) gegenereerd uit herprogrammering somatische cellen geïsoleerd van menselijke patiënten bieden nieuwe inzichten in verschillende ziekten. Bovendien kunnen voorlopercellen en gedifferentieerde cellen die zijn afgeleid van hescs nuttig zijn voor onderzoek waarbij stamceltherapie en geneesmiddelen screening1,2,3,4worden behandeld.
hESCs kunnen worden geïnduceerd om te differentiëren in neurale voorlopercellen (Npc's), die multipotential cellen met een uitgebreide zelfvernieuwings capaciteit zijn. Vervolgens kunnen deze cellen worden gedifferentieerd in neuronen, astrocyten, en oligodendrocyten5,6. Npc's bieden ook een cellulaire systeem voor in vitro studies van neuro ontwikkelingsbiologie en verschillende neurologische aandoeningen. Echter, huidige kolonie type cultuur methoden met hescs en hun differentiatie in npc's zijn inefficiënt en vaak betrekken cocultuur, alsmede embryoid lichaam (EB) en rozet vorming5,7,8,9. Deze protocollen vertonen lagere overlevingspercentages en spontane differentiatie en zijn meer tijdrovend.
Gepresenteerd hier is een verbeterde en robuuste cultuur systeem dat is gemakkelijk schaalbaar en maakt gebruik van hoge dichtheid eencellige type cultuur van hESCs10. De opname van ROH-kinase (Rock) remmer droeg bij aan een significant verbeterde overlevings efficiëntie tijdens het type eencellige cultuur van hesc10,11,12,13,14. In dit cultuur systeem kunnen hESCs gemakkelijk worden onderhouden en uitgebreid. Daarnaast presenteert het protocol een efficiënte methode om npc's te genereren van de eencellige type cultuur van hescs, waardoor de productie van zeer zuivere NPCs. remming van BMP/tgfβ/activin signalering trajecten met alk remmers efficiënt induceren differentiatie van eencellige type hescs in npc's15,16, die vervolgens kan worden geïnduceerd om te differentiëren in functionele neurale lijnen, zoals Dopaminerge neuronen en astrocyten.
Samengevat, het eencellige type cultuur protocol met behulp van hESCs biedt een aantrekkelijk model om de differentiatie van deze cellen te bestuderen in verschillende kilometerstanden, waaronder Npc's. Dit protocol is gemakkelijk schaalbaar en daarom geschikt voor het genereren van cellen voor onderzoek waarbij regeneratieve therapie en geneesmiddelen screening worden betrokken.

Efficient Neural Differentiation using Single-Cell Culture of Human Embryonic Stem Cells

Menselijke embryonale stamcellen kunnen worden geïnduceerd om te differentiëren in neurale voorlopercellen en vervolgens in neuron astrocyten en oligodendrocyte. Om neurale voorloper cel of NPC differentiatie induceren, dissociaal het eencellige type menselijke embryonale stamcellen met een milliliter van detachement oplossing en incubeer ze op 37 graden Celsius gedurende 10 minuten.Centrifugeren de cellen gedurende twee minuten op 370 keer g. Verwijder de onthechtingsoplossing supernatant. Dan resuspend de cellen in DMEM/F-12.Plaat de cellen op een kelder membraan matrix gecoate zes-put plaat op een dichtheid van twee keer 10 tot de vijfde cellen per put in twee milliliter van mTeSR-1 met 10 micromolar ROCK remmer. Vervang na 24 uur het kweekmedium door neurale inductiemedium aangevuld met één micromolar Dorsomorphin en vijf micromolar SB431542. Verander het medium om de andere dag tijdens de eerste vier dagen van neurale inductie, dan elke dag tot de samenvloeiing wordt bereikt op dag zeven.Na zeven dagen van neurale inductie, dissociate de cellen door het toevoegen van een milliliter onthechting oplossing en incuberen ze gedurende 10 minuten op 37 graden Celsius. Voeg een milliliter NPC expansiemedium toe en pipette de cellen voorzichtig op en neer om opnieuw te worden opgetrokken.Dan plaat een keer 10 tot de vijfde cellen per put in twee milliliter van NPC expansiemedium op de kelder membraan matrix gecoat zes-put platen. Passage de cellen als dat nodig is wanneer culturen bereiken 90%-samenloop toe te voegen 10 micromolar ROCK remmer tijdens de eerste drie tot vier passages. Verander het medium om de andere dag tijdens de uitbreiding.Om PLO laminine gecoate platen te bereiden, verdun de PLO-voorraadoplossing in PBS of water tot een uiteindelijke concentratie van 10 microgram per milliliter, meng goed, en bestrijkt elke put van een zes-put plaat met één milliliter van de oplossing. Broed de platen minstens twee uur bij 37 graden Celsius of 's nachts bij vier graden Celsius. Om culturen door te geven wanneer ze samenvloeiing bereiken, distantieer de cellen met een milliliter onthechtingsoplossing en broed ze gedurende 10 minuten bij 37 graden Celsius.Vercentrifugeren ze vervolgens op 370 keer g en verwijder de ontwijdingsoplossing supernatant. Resuspend de cellen in een milliliter VAN DA1 medium en plaat een keer 10 tot de vijfde cellen in twee milliliter van medium op de PLO laminine gecoate zes-put platen. Om de NPC's te differentiëren in astrocyten, plaat de cellen op de PLO laminine platen en NPC expansie medium bij een dichtheid van 50%veranderen het medium tot astrocyten medium na 24 uur, en passage de cellen zoals eerder beschreven.Toen menselijke embryonale stamcellen van kolonietype werden aangepast aan de eencellige kweek, bleek dat de cellen bij hoge dichtheid konden worden gehandhaafd en vervolgens gemakkelijk en efficiënt werden gesubcultuurd wanneer de samenvloeiing werd bereikt. Deze cellen behielden de celcycluskenmerken die kenmerkend zijn voor menselijke embryonale stamcellen van kolonietype, zoals een korte G1-fase en een hoog percentage cellen in de S-fase. Kwantitatieve PCR-analyse bevestigde ook dat zij ESC-markers uitdrukken op niveaus die vergelijkbaar zijn met die van kolonietypecellen.Bovendien werd aangetoond dat eencellige menselijke embryonale stamcellen embryonale lichamen konden vormen die cellen uit alle drie de kiemlagen bevatten:endoderm, mesoderm en ectoderm. Vervolgens werd aangetoond dat eencellige embryonale stamcellen efficiënt gedifferentieerd in NPC's, zoals aangegeven door het verlies van typische menselijke embryonale stamcelmorfologie en het uiterlijk van NPC morfologie. Differentiatie werd ondersteund door de toegenomen expressie van kenmerkende NPC-markers en bevestigd door immunostaining en FACS-analyse.Dezelfde analyse toonde ook aan dat meer dan 90% van de cellen positief gekleurd voor SOX1, PAX6, en NCAM eiwitten. Bovendien waren de afgeleide NPC's in staat om zich te onderscheiden in dopaminerge neuronen en astrocyten, zoals aangegeven door het verschijnen van karakteristieke morfologieën en expressie van afstammingsspecifieke markers. Bij het proberen van deze procedure is het essentieel om de menselijke embryonale stamcellen met een hoge dichtheid te plaat om een gezonde, ongedifferentieerde menselijke embryonale stamcelkweek te behouden.Dit protocol biedt een platform voor eenvoudige, robuuste en schaalbare productie van voorloper en gedifferentieerde cel die schaalbaar zal zijn voor basisstudie, medicijnscherm en toepassing in neurodegeneratieve geneeskunde.
