ヒト胚性幹細胞は、神経前駆細胞に分化し、その後ニューロンアストロサイトとオリゴデンドロサイトに分化するように誘導することができる。しかしながら、ヒト胚性幹細胞の培養法及びそれらの神経前駆細胞への分化は非常に非効率的で、生体内での共同培養系で開かれた、ヒト胚性幹細胞を用いた高密度単細胞型培養を用いて容易に拡張可能な改良された堅牢な培養システムを提示する。ヒト胚性幹細胞の単一細胞型培養は、神経前駆細胞とその後の分化を含む様々で明確な分化系統に沿った多段階分化を調節する分子機構を研究するための迅速かつ効率的なシステムを提供する。ヒト胚性幹細胞修飾基膜マトリックス被覆プレートを調製する。少なくとも2〜3時間または一晩、摂氏4度で基膜マトリックス溶液をゆっくりと解凍する。解凍したら、冷たいDMEM/F-12でマトリックスを希釈し、2%よく混ぜ、希釈した溶液を1ミリリットルで6ウェルプレートの各ウェルをコーティングします。コーティングされたプレートを室温で少なくとも3時間または一晩摂氏4度でインキュベートします。コロニー型H9型ヒト胚性幹細胞のフィーダーフリー培養物を基部膜マトリックス上で増殖させるために、ウェルから培地を吸引し、1ミリリットルのDPBSで1回洗浄する。各ウェルに1ミリリットルのディスパーゼ溶液を加え、20分間摂氏37度でプレートをインキュベートします。ディスパーゼを取り出し、DMEM/F-12の2ミリリットルで細胞を穏やかに洗います。洗浄後、培地を取り出し、各ウェルに2ミリリットルのDMEM/F-12を加えます。コロニーを上下にピペットして穏やかに取り外し、15ミリリットルのチューブに移します。チューブを370回gで2分間遠心分離する。培地を吸引する。その後、細胞ペレットを1つの細胞に解離し、2ミリリットルの細胞剥離液を加え、37°Cで10分間インキュベートします。遠心分離を繰り返し、剥離液を取り除きます。mTeSR-1ヒトESC培地中の細胞を上下に軽くピペット化して再懸濁する。コロニー型ヒト胚性幹細胞を単一の細胞型培養物に適合させるために、約150万~200万個の細胞を、10ミクロモルロック阻害剤を含むmTeSR-1の2ミリリットルのプレートにコーティングされた基底膜マトリックスの各ウェルにプレートを入れる。24時間後、ROCK阻害剤なしで新鮮なmTeSR-1で培地を交換してください。毎日培地を変更する 3 日間、セルを単一のセル型として拡張できるようにします。4日目に、分離溶液中の細胞を解離し、先に述べたように再板を取り出します。原稿の方向に従って細胞を再中断してインキュベートした後、小さな胚体を15ミリリットルのチューブに移し、底に落ち着かせ、ピペットで穏やかに培地を取り除きます。EB培地に移し、7日間低い愛着料理で拡大できるようにします。神経前駆細胞またはNPC分化を誘導するために、単一細胞型ヒト胚性幹細胞を1ミリリットルの剥離液で解離し、摂氏37度で10分間培養する。370回gで2分間細胞を遠心分離する。剥離液上清を取り除きます。次に、DMEM/F-12のセルを再び中断します。10マイクロモルロック阻害剤を有するmTeSR-1の2ミリリットルのウェルあたり2倍の密度で6ウェルプレートをコーティングした基底膜マトリックス上の細胞をプレートします。24時間後、培養培地を1つの小臼歯ドルソモルフィンおよび5つのマイクロモルSB431542で補った神経誘導培地に置き換える。神経誘導の最初の4日間に1日おきに培地を変更し、7日目に合流するまで毎日変更します。7日間の神経誘導の後、1ミリリットルの剥離溶液を加え、摂氏37度で10分間インキュベートして細胞を解離する。細胞を370倍gで遠心分離し、剥離液上清を除去する。1ミリリットルのNPC膨張培地を加え、細胞を上下に静かにピペットして再中断します。次いで、6ウェルプレートを被覆した基質膜マトリックス上のNPC膨張媒体の2ミリリットルに10〜5番目の細胞を1回プレートする。培養が90%合流に達したときに必要に応じて細胞を通過し、最初の3〜4つの通路の間に10マイクロモルROCK阻害剤を加える。拡張中に、1 日おきにメディアを変更します。PLOラミニンコーティングされたプレートを調製するには、PBSまたは水でPLOストック溶液を1ミリリットル当たり10マイクログラムの最終濃度に希釈し、よく混ぜ、6ウェルプレートの各ウェルを1ミリリットルの溶液でコーティングします。プレートを摂氏37度で少なくとも2時間、摂氏4度で一晩インキュベートします。次に、PBSで各ウェルをよく洗い、その直後に原稿の指示に従って調製したラミニン溶液を1ミリリットルずつ塗布します。プレートを摂氏4度に1週間まで置きます。NPCをドーパミン作動性ニューロンに分化するには、24時間後に約50%の密度でNPC膨張培地にPLOラミニンコーティングされた皿の細胞をプレートし、その後1日おきに培地をDA1に変更し、その後1日おきに培地を交換します。合流に達したときに培養液を通過するには、1ミリリットルの剥離溶液で細胞を解離し、摂氏37度で10分間インキュベートする。次いで370回gで遠心分離し、剥離液上清を除去する。DA1培地の1ミリリットルで細胞を再懸濁し、PLOラミニンコーティングされた6ウェルプレート上の培地の2ミリリットルに10〜5番目の細胞を1回プレートします。NPCをアストロサイトに分化するには、PLOラミニンプレート上の細胞とNPC膨張媒体を50%の密度でプレートし、24時間後に培地をアストロサイト培地に変更し、前述のように細胞を通過させます。コロニー型ヒト胚性幹細胞を単一細胞型培養に適応させたところ、細胞を高密度に維持し、合流に達すると容易かつ効率的にサブ培養できることが分かった。これらの細胞は、短いG1相及び高割合のS相の細胞のようなコロニー型ヒト胚性幹細胞に典型的な細胞周期特性を保持した。定量PCR分析では、コロニー型細胞と同等のレベルでESCマーカーを発現することも確認された。さらに、ヒト胚性幹細胞の単一細胞が、3つの胚層すべて(エンドデラム、中胚、および外胚葉)の細胞を含む胚体を形成できることが示された。その後、典型的なヒト胚性幹細胞の形態の喪失とNPC形態の出現によって示されるように、単一細胞型ヒト胚性幹細胞がNPCに効率的に分化することが実証された。分化は、シグネチャーNPCマーカーの発現増加によって支持され、免疫染色およびFACS分析によって確認された。同じ分析では、90%以上の細胞がSOX1、PAX6、およびNCAMタンパク質に陽性に染色されたことも示されました。さらに、誘導NPCは、特徴的な形態の出現および系統特異的マーカーの発現によって示されるように、ドーパミン作動性ニューロンおよびアストロサイトに分化することができた。この手順を試みる際には、健康で未分化のヒト胚性幹細胞培養を維持するためには、ヒト胚性幹細胞を高密度にプレートすることが不可欠である。このプロトコルは、神経変性医療における基礎研究、薬物スクリーン、および応用のために拡張可能となる前駆細胞および分化細胞のシンプルで堅牢でスケーラブルな生産のためのプラットフォームを提供する。
